Malignant lymphoma in the Miyazaki district: analysis of 237 biopsy cases.

Pretreatment biopsy specimens of 237 consecutive patients with malignant lymphoma, who presented to us from 1979 to 1982, were reviewed and reclassified. According to the new classification proposed by the Lymphoma Study Group of Japan (LSG), there were 226 patients with non-Hodgkin's lymphoma (NHL) which was further classified as diffuse lymphoma (216 cases), follicular lymphoma (4 cases), mycosis fungoides (4 cases), and others (2 cases). The 216 cases of diffuse NHL were subdivided into small cell (2 cases), medium-sized cell (71 cases), mixed (7 cases), large cell (92 cases), pleomorphic (40 cases), lymphoblastic (3 cases), and Burkitt's type (1 case) lymphoma. Cell surface marker studies using conventional methods were performed on 65 NHL patients, of whom 45 showed T-cell marker and 11 B-cell marker, and 8 had neither marker. The average survival periods were 13.1 mo for 107 patients with NHL, 27.9 mo for those with mycosis fungoides and 70.0 mo for 10 patients with Hodgkin's disease. Patients with adult T-cell leukemia survived for an average of only 5.5 mo. Histologically diffuse pleomorphic type had the worst prognosis. T-cell lymphoma appeared to have a poorer prognosis than B-cell lymphoma.

Fibrinolytic activity of the retinae in streptozotocin-diabetic rats.

Fibrinolytic activity of the retinae in control and diabetic rats was assayed quantitatively in twenty male rats made diabetic by giving a single injection of streptozotocin. All these rats were killed at either 3 months or 12 months. Ten saline-injected rats and five rats treated with 3-0-methylglucose and streptozotocin served as controls. As the plasminogen activator activity in diabetic rats maintained for 12 months was significantly lower than that in controls, we postulate that there may be a poor defense mechanism against microthrombus formation in the retinal vasculature of diabetics, which may contribute to the development of diabetic retinopathy.

[Intra-aortic PGE1 infusion combined with anti-malignant chemotherapy in the treatment of advanced neuroblastoma].

Neuroblastoma is one of the malignant childhood tumors which have a dismal prognosis, but is known to occasionally mature into benign histology spontaneously. Two cases with either stage III or IV neuroblastoma, whose tumors had not been extirpated at the initial operation, were treated by intra-aortic PGE1 infusion combined with oral papaverine and conventional anti-malignant chemotherapy. This therapeutic modality was effective in one case, where the tumor decreased in size and was removed in a second exploratory operation. The histology of the extirpated tumor was benign ganglioneuroma. In this case, however, distant bone metastases were found after the second operation. The role of PGE1 in the mechanism of bone metastasis remains to be determined.

[Regenerative capability in the hindlimb of Xenopus laevis during ontogenetic development].

The regenerative capacity of hindlimb of Xenopus laevis was investigated by amputating the limbs at four levels in various developmental stages including younger postmetamorphosed froglets. Amputations of limbs were performed at the base of limb in stages 50, 51, 52, 53, 54, 55, 58, and 60 (Nieuwkoop and Faber's table), at the middle of limb bud in stages 50, 51, 52 and 54, and at mid-thigh and mid-shank in stages 58 and 60, and the froglets in 2 and 3 cm in snout-vent length. In the present experiments the regenerative capacity of limbs was expressed by the rate of regeneration and morphogenesis. Tadpoles in the stages after 55 failed to regenerate when the limbs were amputated at base level, but individuals in all the other experimental series exhibited regeneration in various rates irrespective of the level of amputation and the stage. The regenerative capacity increased distally along the proximo-distal axis of the limb when amputated at the same stage, while regeneration was better in younger stages than that in older stages when amputations were made at the same levels. The regenerates obtained by amputation of limbs in stages between 50 and 54, were mainly digitated in that they had 5 toes with 3 claws which is the same pattern with the normal limb, 4 toes with 2 claws, 3 toes with 2 claws or one, and 2 toes with one claw etc. Tadpoles at stage 50 could regenerate toes and claws without defect, but in the later the regenerative capacity gradually declined by reducing the number of toes and claws and accompanied by malformation of skeleton as the stage proceeded. The tadpoles in stages after 58, and the froglets of 2 and 3 cm, produced various types of heteromorphic regenerates of shapes such as cone, spike or rod of which the centra were occupied with cartilage rods. However these regenerates showed no morphological differences according to the developmental stages. These heteromorphic regenerates continued their growth even after one year without any sign of development of digitated feet.

Complete amino-acid sequences of two isocytochromes c of the housefly, Musca domestica L., and their developmental variation in different tissues.

The complete amino-acid sequences of two isocytochromes c (larval- and adult-type cytochromes c) purified from the housefly Musca domestica L. were determined. Their sequences differed at six positions from each other. More than 90% of the total cytochrome c was larval-type during larval stages. The amount of adult-type cytochrome c increased rapidly from 1 day before adult emergence, making the total cytochrome c content increase to approximately 2.5-times as much cytochrome c content as in larvae. Although the major cytochrome c species in flight muscles was adult-type, imaginal disks contained mainly larval-type cytochrome c.

Meal feeding schedule and ventromedial hypothalamic lesions do not affect rhythm of pineal serotonin N-acetyltransferase activity in rats.

Effects of meal feeding schedule and bilateral lesions of the ventromedial hypothalamus (VMH) on the circadian rhythm of pineal serotonin N-acetyltransferase (SNAT) activity were examined in rats, under LD (12:12) condition. Neither meal feeding nor VMH lesions affected the phase of the circadian rhythm of pineal SNAT activity, but the VMH lesions reduced the level. Meal feeding caused a shift of the phases of the daily rhythms of phosphoenolpyruvate carboxykinase and tyrosine aminotransferase activities in the liver. These findings suggest that the circadian rhythm of pineal SNAT activity is not entrained by the food intake, and that the VMH does not function as a master oscillator of the rhythm.

Reductive cleavage of Xaa-proline peptide bonds by mild alkaline borohydride treatment employed to release O-glycosidically linked carbohydrate units of glycoproteins.

During the deglycosylation reaction of fish egg polysialoglycoproteins under the conditions of 1 M NaBH4 in 0.1 M NaOH at 37 degrees C for 48 h, a marked loss of the glycine content has been encountered, besides the serine and threonine residues to which the carbohydrate units are linked. The chemical basis behind this phenomenon has been elucidated by amino acid analysis first of the major glycopeptides (carbohydrate-(O)Thr-Gly-Pro-Ser) derived from desialylated polysialoglycoproteins and subsequently six proline-containing peptides before and after treatment under similar conditions. It has thus been established that -Xaa-Pro- sequences are remarkably susceptible to reductive cleavage under such mild aqueous conditions. In view of the finding that the reductive cleavage of insulin B-chain, which contains a single proline residue adjacent and C-terminal to a threonine residue, led to about 80% loss of the threonine residue, deglycosylation with alkaline borohydride reagents warrants a special comment. The decreased amounts of serine or threonine residues cannot be related simply to the degree of glycosylation of these residues. The above results are therefore discussed in the relation to other work.

Enzymatic dehydrogenation of tryptophan residues of human globins by tryptophan side chain oxidase II.

Enzymatic dehydrogenation of tryptophan residues in human alpha- and beta-globins has been demonstrated with tryptophan side chain oxidase II, a hemoprotein from Pseudomonas. In 1 M ammonium acetate, pH 4, the enzyme catalyzed the conversion of Trp-14 of alpha-globin to alpha, beta-dehydro-Trp. Under identical conditions, Trp-15 of beta-globin was converted to alpha, beta-dehydro-Trp, whereas Trp-37 was unmodified. No modification was obtained in 0.4% sodium dodecyl sulfate or 8 M urea. The dehydrogenase component of tryptophan side chain oxidase II was also active toward globins in the presence of ferricyanide, whereas tryptophan side chain oxidase I was inert. Upon analyses of the tryptic fragments of the modified beta-globin, two forms of alpha, beta-dehydro-Trp-15 distinguishable by ultraviolet absorption and fluorescence were identified. Each of them rapidly equilibrated with another form on irradiation at 300-380 nm. We tentatively assigned them to two isomers of alpha, beta-dehydro-Trp distinguishable by the configuration around its beta-carbon atom. alpha, beta-Dehydro-Trp in globins and certain polypeptides used herein showed a blue fluorescence at 420 nm. In [alpha, beta-dehydro-Trp 15]beta-globin, the fluorescence was exceptionally intense, the fluorescence yield being comparable to that of NADH. From several lines of evidence, the result was ascribed to the excitation energy transfer from Trp-37 to alpha, beta-dehydro-Trp-15 in the modified beta-globin.

Novel oligosaccharide chains on polysialoglycoproteins isolated from rainbow trout eggs. A unique carbohydrate sequence with a sialidase-resistant sialyl group, DGa1NAc beta 1 leads to 4(NeuGc2 leads to 3)DGa1NAc.

A series of novel carbohydrate chains all possessing a hitherto unknown trisaccharide unit, Ga1NAc beta 1 leads to 4-(NeuGc2 leads to 3)Ga1NAc beta 1 leads to, have been isolated from trout egg polysialoglycoproteins, a new class of glycoproteins, on alkali-borohydride treatment. On the basis of chemical (methylation, Smith degradation, and hydrazinolysis-nitrous deamination) and direct-probe mass spectrometric methods. deamination) and direct-probe mass spectrometric methods, the structures of a series of the first major type of oligosaccharide alditols having a sialidase-resistant N-glycolyneuraminic acid residue in each molecule were determined. The structures thus determined are novel and all possess a unique carbohydrate sequence (sialidase-resistant unsubstituted sialyl group italicized): Ga1NAc beta 1 leads to 4(NeuGc2 leads to 3)Ga1NAc beta 1 leads to 3Gal beta 1 leads to-4Gal beta 1 leads to 3[(leads to 8NeuGc alpha 2)n leads to 6]Ga1NAcol (n = 0 through 3).

Fish egg polysialoglycoproteins: circular dichroism and proton nuclear magnetic resonance studies of novel oligosaccharide units containing one sialidase-resistant N-glycolylneuraminic acid residue in each molecule.

Long-core units having the common sequence Ga1NAc beta 1 leads to 4(NeuGc2 leads to 3)Ga1NAc beta 1 leads to 3Gal beta 1 leads to 4Ga1 beta 1 leads to 3Ga1NAc are one of the major constituents of rainbow trout egg polysialoglycoproteins. The existing ambiguity regarding the anomeric configuration of the sialidase-resistant unsubstituted sialyl group present in this novel type of oligosaccharide chains has been resolved by a circular dichroism difference spectral method. The fact that the negative band originating from the carbohydrate n leads to pi transition for this sialyl group was observed offers conclusive proof of the alpha-anomeric configuration. Next particularly interesting is the fact that the chemical shifts of the sialidase-resistant sialyl H-3eq and H-3ax protons were respectively found at relatively higher and lower magnetic field than for the corresponding protons of other sialyl groups. A consideration of molecular models shows that the observed anomalies are all in the directions compatible with expectations on the basis of the magnetic anisotropy effect due to the carboxylate group and steric compression effects by van der Walls interactions between groups that are sterically compressed. In addition to the observed resistance to bacterial sialidases of this sialyl group, it did not behave even as a competitive inhibitor of the sialidase, Arthrobacter ureafaciens, indicating that inaccessibility of this unique sialyl group toward the enzyme. Finally, the analysis of the proton nuclear magnetic resonances of sialidase-sensitive mono- and oligosialyl groups present in the long-core units was based on comparisons of diagnostically important regions in the spectra of homologous oligosaccharides of N-glycolylneuraminic acid.

Fish egg polysialoglycoproteins: structures of new sialooligosaccharide chains isolated from the eggs of Oncorhynchus keta (Walbaum). Fucose-containing units with oligosialyl groups.

A series of acidic oligosaccharide alditols having different neutral core oligosaccharides were isolated from salmon egg polysialoglycoproteins by alkali-borohydride treatment followed by anion-exchange chromatography and Iatrobead chromatography. Their structures were determined by methylation analysis, molecular secondary ion mass spectrometry of underivatized oligosaccharides, and enzymatic desialylation. The molecular secondary ion mass spectra of intact sialooligosaccharides exhibit pronounced quasi-molecular-ion peaks, (M + H)+, (M + Na)+, (M + 2Na - H)+, and/or (M + K)+, as well as some diagnostic sequence ion peaks. Of a number of oligosaccharide alditols, the following are novel: Fuc alpha 1 leads to 3GalNAc beta l1 leads to 3Gal beta 1 leads to 4Gal beta 1 leads to 3[(leads to 8NeuGc alpha 2)n leads to 6]GalNAcol (n = 1-6). The proton nuclear magnetic resonance spectra of these oligosaccharides are also reported and discussed.
