[A case of non-neoplastic pineal cyst presenting Parinaud's syndrome].

A case of non-neoplastic pineal cyst with Parinaud's syndrome is presented. A 54-year-old woman was admitted to the department of neurosurgery. Fukui Medical School on October, 1985, complaining of the paralysis of upward gaze. A computed tomography (CT) demonstrated a low density mass in the pineal region and the mass was not enhanced after the injection of contrast agent. Metrizamide CT cisternogram delineated the mass clearly. Bilateral carotid and vertebral angiograms showed no significant findings. On November 5, an operation was performed by means of an occipital transtentorial approach. A cyst filled with clear yellowish fluid was found in the pineal region. The cyst was evacuated and its wall was subtotally resected. The histological examination revealed the cystic cavity lined by fibrous astrocytes which were surrounded by normal pineal tissue. Therefore the cyst was diagnosed as a non-neoplastic pineal cyst. The postoperative course was uneventful. The paralysis of upward gaze was gradually improved. On December 27, the patient was discharged in excellent condition. Non-neoplastic small cysts which do not cause an enlargement of the pineal body are common incidental findings at autopsy, whereas large cysts which cause symptoms due to compression of the corpora quadrigemina and the production of internal hydrocephalus are rare. In this paper, non-neoplastic pineal cyst is discussed.

Differences in clinical and immunological findings of systemic lupus erythematosus related to age.

Five hundred and seventy patients with systemic lupus erythematosus were classified into 3 groups (younger onset, adult onset and older onset) according to their ages at the time of diagnosis. There were more patients with HLA-Bw35 and/or DR2 in the younger onset group than the adult onset group. Typically related systemic features and serological abnormalities were more frequently found in the younger onset group than the other groups. The disease patterns of the adult onset group were more heterogeneous than those of the other 2 groups. The causes of death in the older onset group were mainly complications such as infections and perforated peptic ulcers.

Use of 4-bromomethyl-7-methoxycoumarin for derivatization of pyrimidine compounds in serum analysed by high-performance liquid chromatography with fluorimetric detection.

Derivatization of the pyrimidine nucleobases and nucleosides with 4-bromomethyl-7-methoxycoumarin was studied with the aim of developing a sensitive and selective column liquid chromatographic method for these substances in serum. The labeling reactions and the nature of derivatives are discussed, together with the chromatographic properties of these derivatives. The derivatives are stable for at least several weeks. Typical detection limits are 50 pg for inosine, 150 pg for uridine, 50 pg for uracil, 50 pg for thymine and 100 pg for fluorodeoxyuridine, respectively. Within-day coefficients of variation averaged 5.0% for the stored-frozen serum pools; the mean day-to-day value was 5.2%. Thirty samples could be processed per working day.

Physical and functional association of the atrial natriuretic peptide receptor with particulate guanylate cyclase as demonstrated using detergent extracts of bovine lung membranes.

Coupling of the atrial natriuretic peptide (ANP) receptor to particulate guanylate cyclase has been demonstrated kinetically and chromatographically using bovine lung plasma membranes and their detergent extracts. Addition of ANP to the membrane suspension stimulated guanylate cyclase activity 2-5-fold indicating the presence of ANP-sensitive particulate guanylate cyclase. The enzyme retained the ability to respond to ANP even after solubilization with digitonin. Characterization of the solubilized enzyme by gel filtration and affinity chromatography revealed that the ANP receptor and particulate guanylate cyclase exist as a functionally but not covalently linked stable complex.

Methylation of the c-myc gene changes during aging process of mice.

The degree of methylation at c-myc proto-oncogene was found to change during aging process of mice by the use of methylation-sensitive restriction enzymes. The spleen DNA showed hypomethylation as mice aged, while hypermethylation was observed in the liver DNA. The brain DNA on the contrary revealed no appreciable difference between young and old mice. When the DNAs were examined at actin and dihydrofolate reductase (DHFR), no significant change was observed. It suggests that an age-related change of oncogene structure may be one of the factors which are related to an age-associated increase of cancer incidence rate.

Fucosterol decreases angiotensin converting enzyme levels with reduction of glucocorticoid receptors in endothelial cells.

The modulation of angiotensin converting enzyme (ACE) levels was studied using fucosterol, one of phytosterols, in cultured bovine carotid endothelial cells. Addition of fucosterol to the culture medium resulted in the decrease of ACE activity of endothelial cells; however, fucosterol did not directly inhibit ACE activity. Dexamethasone elevated the levels of ACE in normal cells, but this effect was not seen in the fucosterol-treated cells. Receptor assays showed that the amount of glucocorticoid receptors in fucosterol-treated cells decreased to an undetectable level. These results indicate that fucosterol lowers the ACE levels on the endothelial cells by inhibiting the synthesis of glucocorticoid receptors involved in the regulation of ACE levels.

Effect of polyamines on in vitro reconstitution of ribosomal subunits.

The effect of polyamines on in vitro reconstitution of Escherichia coli 30S and 50S ribosomal subunits has been studied. Spermidine stimulated the reconstitution of 30S particles from 16S rRNA lacking the methyl groups on two neighboring adenines and total proteins of 30S subunits at least 1.6-fold. The reconstitution of 30S particles from normal 16S rRNA and total proteins of 30S subunits exhibited only slight spermidine stimulation. However, the optimal Mg2+ concentration of the reconstitution was decreased from 20 mM to 16 mM in the presence of 3 mM spermidine. In the absence of spermidine the assembly of 30S particles from normal 16S rRNA was more rapid than the assembly from 16S rRNA lacking the methyl groups on two neighboring adenines. The reconstitution of 50S particles from 23S and 5S rRNA and total proteins of 50S subunits was not influenced greatly by spermidine. Gel electrophoresis results, from reconstitution experiments of 30S particles from 16S rRNA lacking the methyl groups on two neighboring adenines and total proteins of 30S subunits, showed that the assembly of S1 and S9 proteins to 23S core particles was stimulated by spermidine during reconstitution. The relationship of polyamine effects on in vitro ribosome assembly from its constituents to in vivo ribosome assembly is discussed. The reconstitution of Bacillus subtilis 30S particles from 16S rRNA and total proteins of 30S subunits was also stimulated approximately 1.3-fold by 3 mM spermidine.

Cellular basis of in vitro anti-DNA antibody production: evidence for T cell dependence of IgG-class anti-DNA antibody synthesis in the (NZB X NZW)F1 hybrid.

An in vitro system was designed to measure anti-DNA antibody synthesis, and the cellular basis of this autoantibody production in NZB X NZW (B/W)F1 (B/W F1) mice was analyzed. The spleen cells from old B/W F1 mice contained a number of B cells that spontaneously produced anti-DNA antibodies of both IgM and IgG classes in the absence of stimulants, thereby demonstrating that these B cells had been activated in vivo. These activated B cells could be removed by Sephadex G-10 column (G-10) filtration. Such G-10-passed, homogeneously small B cells were activated by the stimulant lipopolysaccharide (LPS) and produced both IgM and IgG class anti-DNA antibodies. The G-10-passed cells contained both B and T cells, and the cytotoxic treatment of the cells with monoclonal antibodies to T cells, anti-Thy-1 and anti-L3T4, abolished the LPS-induced IgG class, but not IgM class, anti-DNA antibody syntheses. Thus, the LPS-induced production of IgG class anti-DNA antibodies in B/W F1 mice is regulated by T cells. Reconstitution experiments revealed the requirement of T-B cell contact but not of the proliferative response of T cells. Moreover, there was no apparent adherent cell requirement. Such IgG class anti-DNA antibodies were produced only by spleen cells from old B/W F1 mice, but not from young B/W F1, NZB, NZW, and C57BL/6 mice. Like IgM class anti-DNA antibodies, LPS-induced synthesis of polyclonal IgM was T cell-independent. Only a slight reduction in the polyclonal IgG synthesis was observed after the G-10-passed cells had been treated with anti-Thy-1 antibody plus complement. This study should facilitate investigation of cell to cell interactions in the formation of autoantibodies and their correlations to immunologic abnormalities in autoimmune disease.

Uveitis induced in primates by interphotoreceptor retinoid-binding protein.

Interphotoreceptor retinoid-binding protein was found to be highly uveitogenic in primates. All six monkeys immunized with bovine interphotoreceptor retinoid-binding protein developed ocular inflammation. The main clinical changes were sheathing of retinal vessels and deep well-circumscribed yellow-white lesions. Histologic changes in the retina included shortening of the outer segments and foci of inflammation, which extended through all retinal layers. Particularly profound were the choroidal changes, such as severe inflammatory infiltration, which often formed focal granulomas with epithelioid and giant cells. Some granulomas had the structure of Dalen-Fuchs nodules. These structures and other histologic changes in the monkeys closely resemble those in uveitic conditions in humans, such as sympathetic ophthalmia or Vogt-Koyanagi-Harada syndrome. These findings show that antigen that localizes specifically in the retina may initiate an immunopathogenic process affecting mainly the choroid.

Naturally occurring antibody response to DNA is associated with the response to retroviral gp70 in autoimmune New Zealand mice.

The spontaneous occurrence of retroviral gp70 immune complexes (ICs) in the blood of autoimmune New Zealand black and (New Zealand black X New Zealand white)F1 ([NZB X NZW]F1) mice is determined by a single dominant locus of the NZB strain (provisionally designated Agp-1). A combined effect of 2 unlinked dominant NZB loci (Ads-1 and Ads-2) is required for the production of anti-double-stranded DNA (anti-dsDNA) antibodies in these mice. The present genetic studies using (NZB X NZW)F1 X NZW backcross mice and their second through fourth generation progeny revealed that Agp-1 and Ads-1 exist in common or are closely linked on chromosome 17 of NZB mice and are related to the occurrence of renal disease. The renal disease and the serum level of both anti-dsDNA antibodies and gp70 ICs were more intense in (NZB X NZW)F1 hybrids than in NZB mice, indicating the contribution of NZW genes. In (NZB X NZW)F1 mice, a single dominant locus from the NZW strain, Agp-3, intensified the magnitude of gp70 IC formation, and a combined effect of 2 unlinked dominant loci from the NZW strain, Ads-3 and Ads-4, acted to increase the serum titers of anti-dsDNA antibodies. This study clearly indicates that the NZW loci Agp-3 and Ads-3 exist in common or are tightly linked on chromosome 17, are closely linked to the H-2 complex, and are associated with the severity of renal disease in (NZB X NZW)F1 hybrids.(ABSTRACT TRUNCATED AT 250 WORDS)

Does the deletion within T cell receptor beta-chain gene of NZW mice contribute to autoimmunity in (NZB X NZW)F1 mice?

To determine the transacting genetic factors of NZW contributing to the development of autoimmune disease in (NZB X NZW)F1 (B/W F1) mice, we examined the relationship between the T cell receptor beta chain gene deletion and the severity of autoimmune manifestations in 76 B/W F1 X NZB backcross mice. Very high association between the T cell receptor beta chain gene deletion and the development of autoimmune manifestations including the production of IgG anti-DNA antibodies and circulating retroviral gp70 immune complexes was observed, indicating that a defect in the NZW T cell receptor beta chain gene or a locus closely linked to it contributes to the autoantibody formation in B/W F1.

Requirement of H-2 heterozygosity for autoimmunity in (NZB X NZW)F1 hybrid mice.

In the F1 hybrid of autoimmune New Zealand Black (NZB) and phenotypically normal New Zealand White (NZW) mice, there occurs a severe systemic lupus erythematosus (SLE)-like autoimmune disease more fulminant than that found in the parental NZB mice. To determine the role of the H-2 complex in the pathogenesis of autoimmune disease of the (NZB X NZW)F1 hybrid, we developed H-2-congenic NZB (NZB.H-2z) and NZW (NZW.H-2d) strains, and compared the degree of autoimmune features between congenic H-2d/H-2d and H-2z/H-2z homozygous F1 hybrids and the original H-2d/H-2z heterozygous (NZB X NZW)F1 hybrid. We found that autoimmune features such as productions of IgG class anti-DNA antibodies and retroviral gp70 immune complexes and the development of renal disease were to a great extent reduced in both H-2 homozygous F1 hybrids, as compared with the H-2 heterozygous (NZB X NZW)F1 hybrid. It would thus appear that the heterozygosity of H-2d haplotype derived from NZB and H-2z from NZW is essential for the autoimmune disease characteristic of the (NZB X NZW)F1 hybrid.
