Immunocytochemical localization of bone morphogenetic proteins (BMPs) in salivary gland pleomorphic adenoma.

The localization of bone morphogenetic protein (BMP)-1, -2, -3 and transforming growth factor (TGF)-beta in normal salivary gland and pleomorphic adenoma of the salivary gland has been examined immunocytochemically. Tumor cells with BMP immunostaining in pleomorphic adenoma were associated with some solid cellular and tubuloglandular patterns, and with stellate cells in the myxoid area. In addition, in the chondroid area of three pleomorphic adenomas, chondrocyte-like cells were positive for BMPs. It is speculated that BMPs secreted by the tumor cells play a role in the formation of the chondroid component in pleomorphic adenoma by inducing some tumor cells, probably neoplastic myoepithelial cells, to differentiate to chondrocytes by metaplastic change. No tumor cells specifically immunostained with TGF-beta were found. TGF-beta was positive in fibrous and hyalinized stroma. In the submandibular gland, only anti-BMP-1 antibody specifically reacted to apical portions of degenerated serous acinar cells.

[A study on Fgr expression and its associated molecules in murine lymphoid and haematopoietic tissues].

The c-fgr gene is a member of the src family of proto-oncogene tyrosine kinases. A monoclonal antibody (2H2 mAb) which recognizes the unique N-terminal domain of the murine c-fgr gene product (Fgr) has been established. Immune complex kinase assay with a monocytic leukemia cell line demonstrated that 2H2 precipitated a 59 kilodalton (kDa) protein, which corresponds to the molecular weight of murine Fgr. Fgr was expressed highly in lymph nodes, slightly in spleen and peripheral blood leukocytes and barely in thymus. The Fgr was hardly detectable in bone marrow. Immunohistochemical analysis showed that the expression of Fgr was restricted to cells of monocyte/macrophage lineage located in the marginal zone of the spleen and in the paracortical zone and medulla of lymph nodes. However, various haematopoietic or lymphoid tumor cell lines different from a lineage of monocyte/macrophage were shown to express Fgr molecule by immune complex kinase assay. Although normal resting haematopoietic or lymphoid cells did not express Fgr protein, activated T and B cells expressed Fgr. In the presence of a mild detergent, the Fgr was co-immunoprecipitated with a 75 kDa protein (p75) and several other molecules expressed on the cell surface membrane. Furthermore, the molecule co-immunoprecipitated with Ly6C molecule from a macrophage cell line showed protein tyrosine kinase (PTK) activity. Peptide mapping showed that this kinase activity was derived from Fgr. The similarity of the relationship between this intramembrane p75 and/or Ly6C and the cytoplasmic Fgr to the relationships previously reported between T cell antigen receptor complex including CD4 and CD8 coreceptors, and Lck or Fyn in T cells and between surface IgM and Lyn or Blk in B cells suggested that the Fgr and p75 or Ly6C are indeed associated each other and responsible for recognition of extracellular substances (either cellular or non-cellular) and for signal transduction. It seems likely that these molecules are involved in activation of cells of monocyte/macrophage lineage.

[Randomized controlled trial of MMC + UFT and MMC + 5-FU therapy in advanced gastric cancer].

To compare the effects of MMC + UFT (A) with MMC + 5-FU (B) therapy on the response rate, survival time and changes with time in quality of life (QOL) in unresectable or recurrent gastric cancer, we carried out a study by a multicenter, randomized controlled trial from June 1990 to August 1992. 39 patients were randomly divided into A group and B group, including 6 incompletely evaluated cases and 3 in eligible cases. Responses were recognized in 3 out of 13 evaluable cases (23.1%) treated with regimen A and in one out of 15 evaluable cases (6.7%) treated with regimen B. Mean survival time was 150 days (A) and 116 days (B), respectively. These results suggested that MMC + UFT therapy is one of the effective regimens for advanced gastric cancer and as useful as MMC + 5-FU therapy. Moreover, it showed that regimen A was superior to B on QOL.

[An autopsied case of pleuro-pulmonary cryptococcosis].

We report a case of pleuro-pulmonary cryptococcosis. An 86-year-old man was admitted to the hospital with dyspnea, a high fever and an oppressive sensation in the chest. Chest X-ray on admission revealed massive pleural effusions, interstitial shadows and consolidation with air bronchograms in both lung fields. The cause of pleural effusion was unknown. The patient died one month later. Autopsy revealed severe pleural cryptococcosis and organizing pneumonia with a small number of cryptococci but without granuloma. To our knowledge, there have been very few documented cases of pleuro-pulmonary cryptococcosis with such massive bilateral pleural effusions.

Production of an interleukin-8-like chemokine by cytokine-stimulated rat NRK-49F fibroblasts and its suppression by anti-inflammatory steroids.

Normal rat kidney fibroblasts (NRK-49F cells) stimulated with interleukin-1 beta (IL-1 beta) and tumor necrosis factor-alpha (TNF-alpha) produced mainly cytokine-induced neutrophil chemoattractant (CINC) which is the rat counterpart of human gro/melanoma growth stimulatory activity. In addition, the cytokine-stimulated cells produced two minor neutrophil chemoattractants which are highly related to murine macrophage inflammatory protein-2 in their NH2-terminal amino acid sequences. IL-1 beta was a stronger stimulator than TNF-alpha, and addition of both the cytokines into the NRK-49F cell culture caused an additive stimulation for rat gro/CINC production. The anti-inflammatory steroids (dexamethasone, prednisolone and hydrocortisone) at 10(-9)-10(-6) M significantly suppressed the production of rat gro/CINC by the IL-1 beta-stimulated NRK-49F cells in a dose-dependent manner. The relative potencies of the inhibitory activity of the steroids on the rat gro/CINC production were dexamethasone > prednisolone > hydrocortisone. On the other hand, the non-steroidal anti-inflammatory drugs (indomethacin and piroxicam) at 10(-7)-10(-5) M showed no apparent inhibitory effect on rat gro/CINC production by NRK-49F cells stimulated with IL-1 beta.

Expression of bone morphogenetic protein in human adenocarcinoma cell line.

A subclone termed as HSG-S8 was previously isolated from human salivary adenocarcinoma cell line, HSG, and proliferated in a serum-free culture. When HSG-S8 and parental HSG cells were transplanted into nude mice i.m. or s.c., both cells reproducibly induced adenocarcinoma. Furthermore, injection of HSG-S8 consistently induced formation of cartilage i.m. or bone s.c. in tumors, but did not parental HSG cells. The cytoplasm of HSG-S8 cells was specifically immunostained by anti-bone morphogenetic protein (BMP)-2 antibody. The conditioned medium of HSG-S8 cells contained a protein of M(r) 18,000 which specifically reacted with anti-BMP-2 protein antibody. Northern blot analysis also revealed that HSG-S8 cells expressed transcripts for BMP-2. On the other hand, parental HSG cells gave a very slightly positive signal only in Northern blot analysis. These results indicate that HSG-S8 cells synthesize and secrete BMP-2 protein, which is probably involved in the formation of cartilage and bone in the tumor tissues in nude mice.

Production of cytokines belonging to the interleukin-8 family by human gingival fibroblasts stimulated with interleukin-1 beta in culture.

Three major neutrophil chemotactic peptides belonging to the interleukin-8 (IL-8) family were purified to homogeneity from the conditioned medium of the IL-1 beta-stimulated human gingival fibroblasts culture. On the basis of their molecular masses and NH2-terminal amino acid sequences, these three neutrophil chemotactic factors were identical to melanoma growth stimulatory activity-alpha (MGSA/gro-alpha), MGSA/gro-gamma and Ala-Val-Leu-Pro-Arg-IL-8 (AVLPR-IL-8), each belonging to the IL-8 family. It has been shown by the chemotaxis studies in vitro that the chemotactic activity of MGSA/gro-gamma is similar to that of MGSA/gro-alpha for both human and rat neutrophils, while AVLPR-IL-8 is chemotactic for human neutrophils but not for rat neutrophils. The in vitro findings were supported by the histological studies in vivo on the intradermal injection of either MGSA/gro-alpha or AVLPR-IL-8 into the back of the rats. The results obtained in the present experiment suggest that human gingival fibroblasts play an important role in gingival inflammation through the production of neutrophil chemotactic factors newly characterized as the IL-8 family in response to inflammatory chemical mediators including IL-1 beta.

The coexistence of cancer cells of different origin within the same lymph nodes.

The coexistence of two differently originating cancer cells within the same lymph node is reported. An 83-year-old male patient died from severe carcinomatous lymphangitis of the lungs five months after gastrectomy for gastric cancer. At autopsy, prostatic cancer metastasis to lymph node was found. Histologically, the prostatic carcinoma cells had a cribriform pattern and the gastric cells showed papillary and tubular features. Immunohistochemically, the former was immunoreactive to prostate specific antigen (PSA) and the latter was stained positively for carcinoembryonic antigen (CEA). Fifty-one of the retroperitoneal and intrapelvic lymph nodes were histologically examined; the coexistence of both types of cancer cells was found in two lymph nodes. Both types were adjoined at the hilum, as was also confirmed by an immunohistochemical double staining technique. These two lymph nodes were located in the pre-aortic and left lateral aortic areas, and they belonged to the terminal lymph node group of both the intra-pelvic and intra-abdominal intestinal organs.

Effects of simetryne on growth of various freshwater algal taxa.

The sensitivity of 56 algal strains, representing 7 taxonomic groups to the triazine herbicide, simetryne, was examined using EC50 values for growth. There was a wide range of values from 6.5 to 1500 microg litre(-1). The Volvocales (Chlorophyceae, Chlorophyta) and Cyanophyceae (Cyanophyta) as a whole were the most sensitive, whereas the Desmidiales (Charophyceae, Chlorophyta) and Bacillariophyceae (Chromophyta) were the most tolerant, although sensitivity differed among strains of a single species. Sensitive and tolerant species were both isolated from samples collected at the same site. The results suggest that changes in species composition and relative abundance will occur when herbicides are applied in natural habitats.

Small bowel obstruction in children: review of 10 years experience.

Small bowel obstruction, excluding postoperative adhesive ileus, in patients > 1 month old treated between June 1982 and May 1992 at Gunma Children's Hospital Medical Center is reviewed. There were 32 patients, 22 boys and 10 girls, whose ages ranged from 1 month to 6 years (median 9 months). Intussusception was the most frequent cause of obstruction and was seen in 17 patients (53.1%). Causative lesions were identified in five patients, and were ileal duplication cysts in four and Meckel's diverticulum in one. Incarcerated inguinal hernia and mesenteric cysts resulted in bowel obstruction in six and three patients, respectively. Other causes included mesodiverticular band, ileal volvulus without malrotation, abnormal adhesion of omentum, abnormal band, vitelline duct remnant and trapping in a mesenteric defect. As for the age distribution, there was no significant correlation between the causes of obstruction and the age of patients. Ultrasonography was useful in differential diagnosis, and this modality should therefore be used in every patient with signs of small bowel obstruction.

Saussureamines A, B, C, D, and E, new anti-ulcer principles from Chinese Saussureae Radix.

Five new amino acid-sesquiterpene adducts, saussureamines A, B, C, D, and E, were isolated from Chinese Saussureae Radix, the dried root of Saussurea lappa CLARKE, together with a new lignan glycoside, (-)-massoniresinol 4"-O-beta-D-glucopyranoside. Their structures were determined on the basis of chemical and physicochemical evidence. Among of these compounds, saussureamines A, B, C showed anti-ulcer effect on HCl/ethanol-induced lesions in rats, and saussureamine A also exhibited a inhibitory activity on stress-induced ulcer formation in mice. During the course of these studies, facile conversion from sesquiterpene having an alpha-methylene gamma-lactone function to amino acid-sesquiterpene adduct has been accomplished by means of Michael type addition reaction.

[Qualitative and quantitative analysis of bioactive principles in Zingiberis Rhizoma by means of high performance liquid chromatography and gas liquid chromatography. On the evaluation of Zingiberis Rhizoma and chemical change of constituents during Zingiberis Rhizoma processing].

As a continuing study on the evaluation of various Zingiberis Rhizoma and the chemical characterization of the processing, a quantitative method by high performance liquid chromatography (HPLC) for 6, 8, 10-gingerol (1, 2, 3), 6,8-shogaol (4, 5), 6-dehydrogingerdione (6), and galanolactone (7) has been developed. By the use of this HPLC method, the contents of these compound in twenty kinds of Zingiberis Rhizoma [originating in China, Taiwan, Vietnam, and Japan (Shizuoka Prefecture)] and fresh ginger root cultivated in Shizuoka Prefecture were examined. It was found that Japanese Zingiberis Rhizoma and fresh ginger root contained 6-gingerol (1), 6-dehydrogingerdione (6), and galanolactone (7) as major constituents, whereas 7 was not detected in imported Zingiberis Rhizoma and 6 was detected in Vietnamese Zingiberis Rhizoma. Furthermore, the contents of 1 and 7 in fresh ginger root decreased remarkably during the processing procedure for Zingiberis Rhizoma. In addition, anti-ulcer sesquiterpene constituents in seven kinds of Zingiberis Rhizoma were analyzed by means of gas liquid chromatography (GLC).

[Chemical fluctuation of the constituents during the drying of Ginseng radix and Ginseng radix Rubra. Crude drug processing by far-infrared treatment].

In order to evaluate the utility of far-infrared drying method for crude drugs, the efficiency in the drying process of Ginseng Radix and Ginseng Radix Rubra was examined. Furthermore, chemical fluctuation of the constituents of Ginseng Radix and Ginseng Radix Rubra, which may occur during their drying process, has been investigated by means of HPLC quantitative analysis for ginsenosides and malonyl-ginsenosides, and TLC qualitative analysis for lipophylic constituents. It has been found that the far-infrared drying method (oven temperature 45 degrees C) dried Ginseng Radix faster without reducing both ginsenosides and malonyl-ginsenosides in comparison with the conventional drying methods such as an air drying and a hot-air drying.

[Pulmonary aspergillosis with air-fluid level in a cavity].

We experienced 17 cases of pulmonary aspergillosis with air-fluid level in a cavity. The incidence of this phenomenon was 39% in 44 patients who were diagnosed as having pulmonary aspergillosis during the period from 1983 to 1991 at Yaizu Municipal Hospital. There were two clinical patterns: primary air-fluid type and secondary air-fluid type. Primary air-fluid type disease began with cavitation with an air-fluid level. Our 14 cases of this type had severe symptoms and inflammatory reaction. In secondary air-fluid type, the fungus ball disappeared and an air-fluid level appeared in a cavity during treatment. Our 3 cases of this type had only mild cough and sputum. The fluid disappeared later in two of these three cases. Some cases of primary air-fluid type changed to fungus ball type or productive aspergilloma on the inner wall of a cavity. Primary air-fluid type thus seems to represent incipient pulmonary aspergilloma, and to be important for early diagnosis and treatment. Of the immunological tests, precipitins to Aspergillus fumigatus proved most useful for early diagnosis.
