Regulation of primary antibody response in mice by two types of suppressor cells resistant to anti-thy-1 serum treatment.

The anti-SRBC antibody response of normal young adult mice (about 3 months old) was specifically suppressed, when the spleen cells derived from the syngeneic donor mice that had been previously primed with a high dose of SRBC were adoptively transferred at the time of antigenic challenge. The suppression was antigen-specific and was mediated by a fraction of the immune spleen cells which appeared to belong to either B cells or their progeny antibody-forming cells. The differences were observed in the properties of the suppressor cells in terms of the period after the priming and the radiosensitivity; i.e., immune spleen cells taken from the mice which had been immunized with SRBC 5 days earlier were able to suppress only 19S PFC, whereas those taken from the mice which had been immunized 14 days earlier were able to suppress both the 19S and 7S PFC responses, and the former cells were susceptible to 400 R X-rays, but not the latter. These two types of suppressor cells appeared to play an important role in the regulation of the sequential change of 19S and 7S antibodies during the primary immune response.

Argyrophil cells in adenomas of familial polyposis coli.

The presence of argyrophil cell was demonstrated in the adenomas of familial polyposis coli (FPC) with the Sevier-Munger method. Usually, argyrophil cells were distributed sparsely near the base of the crypts, rather close to the normal mucosa. But, adenomas with severe or moderate dysplasia did not contain argyrophil cells. The frequency of argyrophil cells in the adenoma was remarkably higher in the sigmoid than in the ascending colon. Because this phenomenon was found both in the non-adenomatous mucosal crypts of FPC and of normal control cases, it is conceivable that the appearance of the argyrophil cells in the adenomas of FPC is affected by the structural arrangement of the original non-tumorous mucosa, as a background. Furthermore, a tendency, that many argyrophil cells were located within the adenomas, was observed in one of the seven families examined. It showed that the argyrophil cell which is thought to be one of the elements constituting the adenoma might actively participate in the growth of the adenoma.

In vitro steroidogenesis in testes of three infants, two with ambiguous external genitalia and one with true precocious puberty: Evidence for the presence of active 17 beta-hydroxysteroid oxidoreductase in immature human testes.

The present report investigated steroidogenesis in vitro in testis tissues obtained from two boys aged 8 months and 4 years with ambiguous external genitalia and male vagina, and a 4-year-old body with true precocious puberty. Histologically, testes of the former two boys are still immature and the testis of the last one contains differentiated Sertoli cells and primary spermatocytes, but no mature Leydig cells are recognized in any of them. In each testis, 17 beta-hydroxysteroid oxidoreductase is active for androstenedione in the presence of an excess amount of NADPH, while delta 5-3 beta-hydroxysteroid dehydrogenase and delta 4-steroid 5 alpha-reductase activities are limited. 17 alpha-Hydroxylase and C17--20 lyase are significantly active in each testis and are enhanced in the testis of the boy with precocious puberty. Although the testis tissue used in the present study may not be biologically normal and the number of cases investigated is still limited, the above results indicate that active 17 beta-hydroxysteroid oxidoreductase is present in immature human testes and that delta 5-3 beta-hydroxysteroid dehydrogenase may become active in the human testis at the advanced stage of the development of testicular function during the puberty.

Demonstration of immunoreactive somatostatin-like substance in villi and decidua in early pregnancy.

With the use of the radioimmunoassay for growth hormone--releasing inhibiting factor (GIF), it was found that measurable amounts of GIF-like substance existed in the chorionic villi and decidua of pregnant women. The indirect immunofluorescent method revealed that the higher intensity of GIF-like immunofluorescence was presented in cytotrophoblasts rather than in syncytiotrophoblasts of the villi and in stromal cells of the decidua.

Ultrastructural study of benign osteoblastoma of the maxilla.

The ultrastructure of benign osteoblastoma of the maxilla in a 14-year-old boy was studied. Morphologically, the tumor tissue was composed of abundant osteoid of a trabecular form, and cellular and vascular connective tissue. The cellular components were osteoblasts, osteoclasts, osteocytes entrapped within the osteoid, and small perivascular cells of two types. The small cells of one type were ovoid and had a relatively large nucleus and a dark cytoplasm. The cells of the other type were elliptical and had a clear cytoplasm. The former seemed to be a preosteoblast and the latter could not be identified. The osteoblast was the predominant cell in this lesion and was characterized by the presence of abundant rough-surfaced endoplasmic reticulum and several Golgi complexes. These characteristics indicated the pronounced activity of collagenous fiber synthesis and the matrix of the osteoid tissue.

[Demonstration of immunoreactive GIF-like substance in villi and decidua by radioimmunoassay and immunofluorescence (author's transl)].

Since Arimura et al (1975) reported the radioimmunoassay for somatostatin (GIF), the concentration of GIF in various organ and brain regions were determined by radio-immunoassay. Dubois et al (1975) reported that immunohistochemically somatostatin was located in the discrete cells of the pancreas as well as the hypothalamus, and from this result, they presented the concept of local hormone instead of systemic hormone which was up to that time accepted in endocrinology. In this study, we developed the high specific anti-GIF serum using rabbits, and with the micro immunodiffusion method, we demonstrated that the precipitin band formed a circular fusion between the GIF and anti-GIF serum. This pattern of reaction was also seen in decidual immunodiffusion. In addition, we developed the radioimmunoassay for GIF using this anti-serum and measured immunoreactive GIF-like substances in villi and decidua of early pregnancy. The concentration of GIF-like substances with 2 N acetic acid extracted of villi and decidua were 0 to 30 pg/0.1 g dry weight. At the same time, we demonstrated the presence of GIF-like substance-containing cells in the villi and decidua by indirect immunofluorescent method. The intensity of immunofluorescence was in cytotrophoblast rather than syncytiotrophoblast, and decidual stromal cell also reacted to the immunofluorescence.

Median rhomboid glossitis associated with amyloid deposition.

There are some controversies over the pathogenesis and clinical features of median rhomboid glossitis. A case of median rhomboid glossitis associated with amyloid deposition was presented. Clinically there was no organ involved in amyloid other than the tongue. It was suspected that median rhomboid glossitis occurred first and that amyloid was induced later.

Age-associated changes in microvasculature of human adult testis.

Age-associated architectural changes of the human testicular microvasculature from 70 autopsy cases were stereoscopically examined with a silicone-rubber injection technique. In the testis of a young subject, the interlobular main arteries run straight. The coiling phenomena of the interlobular centripetal or centrifugal arteries, which are commonly seen in adult testis, have been so far considered as physiological transformation of the vasculature. It was confirmed that the coiling changes in the interlobular main arteries of the human testis appear as an age-dependent alteration of the vasculature closely related to the volume of the gland. The practical importance of the spirallin or coiling of arteries is that it results in a considerable reduction of blood flow. The age-related coiling of the interlobular arteries is virtually accompanied by varying degrees of collapse of the peritubular capillary networks. The reduction of blood supply to the seminiferous tubules plays an active role in promoting aging of the testis. These stereoscopical observations of age-related transfiguration of testicular microvasculature were ascertained also by histometrical examinations.

Ultrastructural study of adenomatoid odontogenic tumor.

Two cases of adenomatoid odontogenic tumor were examined by light and electron microscopy. Morphologically the tumors could be divided into four layers. The ultrastructure of the tumor cells of each layer was revealed to be similar respectively to that of four layers seen in the enamel organ of a normal tooth germ. Duct-like structures and eosinophilic small areas were frequently present in compactly proliferating cell layers. Short columnar cells forming duct-like structures and eosinophilic small areas appeared similar ultrastructurally to ameloblasts during predentin formation. Therefore it seems reasonable to assume that this tumor originates from the enamel organ. The contents in the lumen of each eosinophilic small area varied, and seemed to be secreted into the stroma by circumscribing epithelial tumor cells. A fine filamentous layer was present in both of the duct-like structures and some of the eosinophilic small areas, and the former was different from the latter with respect to the alcian blue (pH 2.5) and toluidine blue (pH 4.4) staining reactions.

Studies on the adjuvant effect of water-in-oil-in-water (w/o/w) emulsion of sesame oil. 1. Enhanced and persistent antibody formation by antigen incorporated into the water-in-oil-in-water emulsion.

Water-in-oil-in-water (w/o/w) emulsion developed in our laboratory is as effective as water-in-oil (w/o) emulsion of Freund's incomplete adjuvant (FIA) in the stimulation of antibody formation. The emulsion is prepared by redispersion of water-in-sesame oil emulsion of an antigen solution in phosphate buffered saline with emulsifier, Tween 80. The emulsion can be stored at 4 degrees C for at least 3 months without any evidence of change in the adjuvanticity and in the w/o/w state. Even a single injection of bovine serum albumin (BSA) in the w/o/w emulsion elicited a high antibody response in mice over the period of almost whole lifespan. 10 microgram BSA in w/o/w could stimulate antibody formation up to 2(12) in hemagglutination titer, while the same dose in free solution did not elicit any detectable antibody. The tissue reactions caused by the w/o/w emulsion at the injected site and in the regional lymph nodes were much less prominent than those by FIA.
