Submandibular glands of both sexes from one week to six months-old were serially sectioned, and mucous cells showing a strong positive stain to alcian blue pH 2.5, first detected at two weeks, showed a high degree of inter-sectional and inter-individual variability. In males, the percentage of animals having mucous cells increased with sexual maturation and attained 100 per cent at age six months. In females, the maximum (60 per cent) was reached by four weeks and then decreased with sexual maturation. Only 28.5 per cent of the glands of adult female rats contained mucous cells. In the six-month-old females, the percentage of female rats having mucous cells increased three-fold after injection of testosterone (total dose 17-21 mg; p less than 0.05, Fisher test). Mucous cells in testosterone-injected females displayed morphological features which suggest a transition from original seromucous cells to mucous cells.
Chronic effects of Cd on the growth and reproduction of the guppy (Poecilia reticulata) were studied using a food chain model, midge larvae as prey and guppy as predator. The transfer rate of Cd from the midge to the guppy was between 0.5 and 1% during the 30-day experiment. Growth rate of the guppy fed Cd-accumulated midge larvae (270 micrograms/g dry wt) for 30 days was not impaired. Cumulative numbers of fry produced by the guppy fed Cd-accumulated midge larvae (210 micrograms/g) for 2 months decreased to ca. 80% of the control. Guppies had been fed the Cd-accumulated midges from 30 days old for 7 months. Cumulative numbers of fry produced by the guppy fed midge larvae-accumulated 500, 800, and 1,300 micrograms Cd/g for 6 months decreased to 79, 65, and 55% of the control, respectively. Similar effects of Cd on the reproduction of guppy were shown between the guppies fed the Cd-accumulated midge larvae (500 micrograms Cd/g) and exposed to 10 or 20 micrograms Cd/liter for the 6 months. The Cd concentrations of the digestive tract, liver, and kidney increased strongly, indicating that such Cd accumulation was brought on mainly through the Cd-accumulated midges. Mortality of the female guppies fed larvae-accumulated 1,300 micrograms Cd/g increased abruptly from the 6 months of the experiment, whereas no male guppy died during the experiment.
Chronic effects of Cd on reproduction of Polypedilum nubifer (Chironomidae) were investigated by Cd-exposure from the egg stage using a flow-through aquarium. No significant effect of Cd on reproduction was observed in the midge larvae which had been exposed to 10 or 20 microg Cd litre(-1). Emergence of the Cd-exposed larvae peaked several days before that of the control, although growth was impaired in 1st and/or 2nd instars. The percentage emergence success decreased to about 46% of the control at 40 microg Cd litre(-1). However, other reproductive processes (adult sex ratio, oviposition success and egg hatchability) were not impaired. The emergence success decreased to less than 3% of the control at 80 microg Cd litre(-1). The emergence success of midge larvae which had been fed Cd-contaminated food with 220 or 1800 microg Cd g(-1) decreased to nearly 60% of the control. However, other reproductive processes were not impaired. Emergence success of larvae exposed to food containing 22 microg Cd g(-1) was unaffected.
Gibels were exposed to cadmium in their aquarium at a concentration of 10 micrograms Cd/l for up to 39 weeks. Distributions of cadmium, copper and zinc in the liver soluble fraction were determined along with sulfur by high performance liquid chromatography-inductively coupled argon plasma-atomic emission spectrometry. Cadmium was sequestered by the two major isoforms of gibel metallothionein as in the case of cadmium injected intraperitoneally into gibel. Several peaks with cadmium, copper, zinc and sulfur were observed other than the two major isoforms and their relative ratios were different between the control and cadmium-exposed fishes.
The purpose of this study was to examine the effect of glucocorticoid on human neoplastic salivary duct epithelial cell line (HSG). Dexamethasone was found to inhibit cell growth and to increase cell size and the ratio of protein content to DNA content in a cell. The inhibition of cell growth was dose-dependent; in comparison to the control (33.8 +/- 3.1 h), the population doubling time was 1.57-fold longer in 10(-5) M dexamethasone (P less than 0.01, N-K test). [3H] thymidine incorporation was inhibited in 45.5% of the control at 10(-5) M. Plating efficiency was 20.5 +/- 3.0% in 10(-5) M and 47.0 +/- 4.4% in the absence of dexamethasone. Cell diameters increased 1.29 fold in 10(-5) M dexamethasone in comparison to the control size (16.0 +/- 2.1 micron). The ratio of total protein content of DNA content increased 1.46 fold in 10(-5) M dexamethasone-treated cells on the seventh day of cultivation. Scatchard plot analysis using [6, 7-3H]-triamcinolone revealed that the HSG cells had apparent cytosolic glucocorticoid receptors with an equilibrium dissociation constant (Kd value) of 6.48 nM, whose number of binding sites (NBS) was 57.8 fmol/mg protein.
In the bone marrow of leukemic patients (31 cases), the number of immunoglobulin containing cells per unit square significantly decreased to the level of approximately 20-50% of the age-matched control, although the serum level of immunoglobulin in these patients was almost comparable to the control level. The number of immunoglobulin containing cells in the bone marrow was inversely related with the degree of infiltration of leukemic cells and the correlation of Ig-G containing cell count (Y) and percentage of blast cells (X) was Y = 91.8-1.9X (r = -0.436, P less than 0.05). Follow-up examination of the bone marrow after chemotherapy revealed that the recovery of the number of immunoglobulin containing cells in the remission state appeared to occur in younger patients, but not in older ones.
Neoplastic epithelial duct cell line from human salivary gland (HSG cell line) contains the specific glucocorticoid receptor. The time course study on the uptake of [3H]triamcinolone acetonide (TA), a synthetic glucocorticoid, by intact HSG cells in a growing monolayer culture showed that translocation of glucocorticoid receptors into nuclei occurred at 37 degrees C, but not at 0 degrees C. To elucidate the subcellular distribution of glucocorticoid receptor from HSG cells, a scaled-up-culture was employed. When the cells were incubated with [3H]TA at 0 degrees C, 94% of the receptors were found in the cytosol fraction, while only 6% of the receptors existed in the nuclei. When the cells were incubated at 37 degrees C, 49% of the receptor complexes were distributed in the nuclei and 74% of these nuclear receptor complexes were extractable with 5 mM pyridoxal phosphate.

A 51-year-old woman with adenocarcinoma of the esophagogastric junction (E-G J), which spread from the E-G J to the cervical esophagus, is reported. Only two cases of carcinoma in the E-G J with the extension of the tumor to the upper or middle portion of the intrathoracic esophagus have been reported in the Japanese literature in the past two decades and there have been no previous reports of E-G J carcinoma with extension to the cervical esophagus. At first, the invasion to the esophagus in our case was thought to be continuous. Histological examination, however, revealed that the tumor in the esophagus was discontinuous in some portions and that the lymph vessels were remarkably invaded. These findings suggested that the extension of the tumor to the esophagus was lymphogenous metastasis.
An autopsy case of a 52-year-old man suffering from chronic manganese poisoning (CMP) is reported with determination of the manganese distribution in the brain. The patient had been working in a manganese ore crushing plant since 1965. In 1967 he began to complain of difficulties in walking and diminished libido. Later, he developed various neuropsychiatric symptoms including euphoria, emotional incontinence, masked face, monotonous speech, "cock-walk", increased muscle tone, weakness of upper and lower extremities, tremor of the eye lids, and exaggeration of knee jerks. The major neuropathological change was degeneration of the basal ganglia, in which the pallidum was severely affected. The pallidum disclosed a loss and degeneration of nerve cells, which was especially marked in the medial segment, a prominent decrease of myelinated fibers, and moderate astrocytic proliferation. The substantia nigra was intact. Distribution of manganese in the brain of the present case of CMP was determined using flameless atomic absorption spectrometry and compared with control cases and also a case of Parkinson's disease (PD). There was no significant difference between the control cases and the case of PD in average concentration of manganese and its distribution in the brain. The present case of CMP showed no elevation in average concentration of manganese in the brain. However, there were some changes in its distribution. Thus, the continuance of neurological disorders in CMP is not linked to an elevated manganese concentration itself in the brain. CMP appears to be different from PD in neuropathology and manganese behavior in brain.
Sinus hyperplasia occurring in the congestive splenomegaly associated with portal hypertension is a interesting model of steady-state hyperplasia in an adult tissue. In vitro labeling of human splenic tissue with 3H-thymidine and the demonstration of S-phase cells by autoradiography was performed to investigate the proliferative activity of sinus-lining and cordal reticular cells in congestive splenomegaly compared with that in the normal spleen. The labeling index (%) of sinus-lining cells after 2 h incubation was 0.07 +/- 0.02 in the normal spleen and 0.26 +/- 0.03 in congestive splenomegaly (t = 4.553, P less than 0.005, n = 11). This result indicated that sinus-lining cells have a long life span and that their proliferative activity is increased in congestive splenomegaly. On the other hand, the labeling of cordal reticular cells and arterial and venous endothelial cells was sparse or absent in both congestive splenomegaly controls, and these cells do not appear to be involved in sinus hyperplasia.
Two kinds of hybridoma clones, one producing monoclonal antibodies against Sertoli cell (TM-1) and the other the basal lamina of the seminiferous tubule (TM-2), were raised by fusion between P3X63Ag8-653 mouse myeloma cells and spleen cells of BALB/c mice (H-2d) immunized with testicular homogenate of the same inbred mice. Immunohistochemically, TM-1 reacted specifically with cytoplasmic component of Sertoli cell and TM-2 with basal lamina of the seminiferous tubule. Using these monoclonal antibodies, spermatogenic disturbance was induced experimentally in BDF1 (H-2b/d) by intratesticular injection of a set of these two antibodies. Single injection of either TM-1 or TM-2 failed to induce the lesion. This fact indicated that TM-1 antibody could reach the Sertoli cell to impair its function, which was otherwise inaccessible without coincidental action of TM-2 antibody. TM-2 antibody appeared to alter the permeability of the basal lamina of the tubule and lower its barrier effect.
Two rare cases of autopsy and surgery presenting extrahepatic biliary obstruction due to intrabile-duct growth of hepatocellular carcinoma were reported. Clinically obstructive jaundice was predominant in comparison with the other symptoms in both cases. In one autopsy case, hepatocellular carcinoma developed in the right lobe of the cirrhotic liver (posthepatitic). It involved the secondary branch of the right hepatic duct and grew into the common hepatic duct. In the other case of surgical operation, hepatocellular carcinoma, which developed in the posterior portion of the right lobe of the cirrhotic liver (posthepatitic), destroyed the posterior wall of the bifurcation of the bilateral hepatic duct and obstructed the common hepatic duct due to the intraductal cancer growth. From the site of the bile duct invasion or permeation by the tumor, two cases were classified into the peripheral (the former case) and proximal (the latter case) types, respectively. Furthermore, as far as obstructive jaundice is clinically concerned, the possibility should be kept in mind that hepatocellular carcinoma may proliferate into the large bile ducts, apart from that of cholangiocarcinoma or cholelithiasis.
Neoplastic epithelial duct cell line from human salivary gland (HSG cell) contained cytosol glucocorticoid receptor. Scatchard analysis of cytosol indicated that the dissociation constant (Kd) was 5.6-6.5 nmol/l and the number of binding sites was 83-92 fmol/mg protein. A competitive assay showed that the binding sites for [3H]triamcinolone acetonide were specific to glucocorticoid. Glycerol density gradient centrifugation displayed that the [3H]triamcinolone acetonide receptor complexes sedimented in the 8.5 S region under low salt conditions and in the 4.2 S region under high salt condition (0.4 M KCl). The same high salt conditions induced an increased binding of [3H]triamcinolone acetonide complexes for DNA-cellulose.

Cortical cerebellar degeneration was found in a 28-year-old man with testicular neoplasm. The patient, who had undergone a left orchidectomy for the testicular tumor, developed progressive cerebellar symptoms with mental changes 7 months later. The autopsy revealed the spread of a malignant germ cell tumor of the testis, and cortical cerebellar degeneration in the central nervous system (CNS) which was characterized by almost complete loss of Purkinje cells and degeneration of the both dentate nuclei and superior cerebellar peduncles. The present case is the first of cortical cerebellar degeneration combined with testicular neoplasms.
