Presence of lysosomal hyaluronidase in human corneoscleral tissue.

Hyaluronidase activity in human corneoscleral tissues was biochemically studied. The corneoscleral junction, particularly the inner layer, showed higher activity than other corneoscleral tissues. The possible role of lysosomal hyaluronidase in the trabecular meshwork is discussed.

IgA nephropathy with subendothelial deposits.

IgA nephropathy with subendothelial deposits in the capillary walls of the glomeruli (IgA type 2) was compared histometrically and clinically with IgA nephropathy without subendothelial deposits (IgA type 1) and membranoproliferative glomerulonephritis with subendothelial deposits (MPGN). Study cases consisted of 32 biopsies from 26 patients of IgA type 1, 25 biopsies from 20 patients of IgA type 2 and 31 biopsies from 27 patients of MPGN. Histological changes of the glomeruli consisted of an increase in the mesangial matrix and hypercellularity in the mesangium in both types of IgA nephropathy, and the degree of the changes was a little higher in IgA type 2 than in IgA type 1 (0.02 < P < 0.05). Mesangial changes of MPGN were marked as compared with IgA type 1 and IgA type 2 (P < 0.001). Histometry of the mesangium on the cases followed up showed that the degree of mesangial thickening increased with lapse of time in IgA type 2 and MPGN, whereas it remained unchanged up to 13 years in IgA type 1. Proteinuria tended to be mild in IgA type 1, moderate in IgA type 2, and marked in MPGN. The impairment of renal function was observed in 21.9% of IgA type 1, in 36.0% of IgA type 2 and in 58.1% of MPGN. IgA type 2 has been shown to be pathologically and clinically intermediate between IgA type 1 and MPGN. These results suggest that there is a clinicopathological overlap between IgA nephropathy and MPGN with IgA deposition.

Acid phosphatase in eyes with pseudoexfoliation.

We studied the activity of acid phosphatase in the anterior segment of the eye with pseudoexfoliation histochemically and biochemically. Light and electron microscopic histochemistry revealed lead precipitates indicating acid phosphatase activity in pseudoexfoliative materials. Biochemical assay for acid phosphatase was significantly higher in the aqueous humor of eyes with pseudoexfoliation than in that of cataractous eyes without pseudoexfoliation.

Adherence of Yersinia enterocolitica to mammalian epithelial cell lines.

Yersinia enterocolitica RIMD 2501003 grown at 25 C avidly adhered to various kinds of cultured epithelial cell lines (HeLa, FL, Y-1 adrenal, human intestine, human conjunctiva) but the bacteria grown at 37 C did not adhere. This phenomenon paralleled the temperature-dependent motility of the bacteria. To clarify the adherence mechanism, we obtained two kinds of mutants, an immobile mutant and a nonadherent mutant, by treatment with N-methyl-N'-nitro-N-nitrosoguanidine. The immobile mutant did not move on soft agar but retained the capacity to adhere to cultured epithelial cells when grown at 25 C. The nonadherent mutant did not adhere to cultured epithelial cells but retained the ability to move on soft agar when grown at 25 C. When the bacteria were killed by heat, ultraviolet light irradiation or formaldehyde they lost their capacity to adhere to the cultured epithelial cells. Antiserum against Y. enterocolitica RIMD 2501003 grown at 25 C was absorbed with the bacteria grown at 37 C, with the bacteria grown at 25 C, with the nonadherent mutant grown at 25 C and with the bacteria killed by various means. Only the antiserum absorbed with bacteria grown at 37 C inhibited the adherence of bacteria. These data indicate that motility does not correlate with adherence of Y. enterocolitica. It appears that the adherence factor involves both a temperature-dependent surface factor and a factor synthesized de novo during the interaction of susceptible cells with the bacteria.

Complement proteins and macrophages. II. The secretion of factor B by lipopolysaccharide-stimulated macrophages.

The secretion of factor B by mouse peritoneal macrophages was found to be enhanced following in vivo or in vitro stimulation with lipopolysaccharide (LPS). The intravenous administration of LPS to mice of various strains caused an increased release of factor B but not the release of acid phosphatase by the peritoneal macrophages obtained from the stimulated mice. In vitro stimulation of cultured macrophages with LPS resulted in an enhanced secretion of both factor B and acid phosphatase. The dose-dependent augmentation of factor B secretion by LPS was found in the macrophages from LPS-responsive C3H/HeN mice, whereas the macrophages from LPS-unresponsive C3H/HeJ mice did not respond to either phenol-extracted LPS or butanol-extracted LPS. The ability of LPS to cause the enhancement of factor B secretion by macrophages was abolished by alkali or acid treatment of LPS, indicating that its lipid A part was responsible for the observed effect.

Hyperglycemic effect of hydralazine in rats.

The effect of hydralazine in the alteration of blood sugar, tissue glycogen and blood cAMP levels in rats were investigated. Hydralazine was found to increase blood sugar level in intact rats when administered i.p. On the other hand, this hyperglycemia was partially blocked either by the treatment of intact rats with propranolol, beta-adrenergic blocker, or by adrenalectomy. Hydralazine treatment increased blood cAMP level, leading to the hyperglycemia, because pretreatment with phentolamine partially blocked the hydralazine-induced hyperglycemia.

Japanese minimum requirements for albumin preparations: recent amendments and current problems.

The following amendments have been introduced to the Minimum Requirements for Human Serum Albumin (HSA) and Plasma Protein Fraction (PPF): 1) The upper limit of sodium content was fixed at 3.7 mg/ml; 2) test for chloride content was introduced; 3) sodium and chloride contents were required to be stated on the label; 4) use of zone electrophoresis was introduced in addition to the free-boundary electrophoresis; and 5) albumin content in HSA was lowered from 97% to 96%. Moreover, current problems in quality control which may require further amendments were discussed, namely: 1) limulus amoebocyte lysate test to replace or supplement the pyrogen test in rabbits; 2) attempt of quantitative expression of visual turbidity; 3) establishment of reference A and B blood group substances; 4) gel permeation analysis and scanning at 370-800 nm; and 5) test for prekallikrein activator (PKA).

Reproductive performance in women with sex chromosome mosaicism.

Among chromosomal analyses performed in 23 couples with a history of 2 or more spontaneous abortions, 5 women had mosaicism of the sex chromosomes, 46,XX/47,XXX, but all the men had normal 46,XY chromosomal complements. A review of the literature revealed that 75% of the fetuses of mothers with sex chromosome mosaicism were abnormal. Fifty percent of theses pregnancies ended in abortion and one third of the infants exhibited a chromosomal or physical abnormality. The possible effect of sex chromosome mosaicism on outcome of pregnancy is emphasized.
