Extracellular phospholipase A2 activity in peritoneal cavity of casein-treated rats.

An extracellular phospholipase A2 was detected in the peritoneal cavity of rats injected intraperitoneally with casein. This activity required Ca2+ ion and had a pH optimum of 9. The increase of phospholipase A2 activity in the peritoneal fluid as a function of time roughly paralleled the increase in the number of polymorphonuclear (PMN) leukocytes found in the peritoneal cavity. Phospholipase A2 activity was also observed in rat PMN homogenates, and its characteristics were similar to those of the enzyme found in cell-free peritoneal fluid fraction. PMN leukocytes may release non-lysozomal phospholipase A2 into the extracellular space as 'inflammation reactions' proceed.

Purification and characterization of proteinase inhibitors from winged bean (Psophocarpus tetragonolobus (L.) DC.) seeds.

Seven proteinase inhibitors were isolated from winged bean seeds by ion-exchange chromatographies. These inhibitors had molecular weights of around 20,000, included four half-cystine residues, and were Kunitz-type inhibitors. Two (WTI-2 and 3) inhibited bovine trypsin strongly and four (WCI-1, 2, 3, and 4) inhibited bovine alpha-chymotrypsin, but in different ways. One mole of WCI-2 or -3 could inhibit 2 mol of alpha-chymotrypsin. The remaining inhibitor (WTCI-1) could bind both bovine trypsin and alpha-chymotrypsin at the molar ratio of 1:1, but not simultaneously. All four chymotrypsin inhibitors cross-reacted with rabbit anti-WCI-3 serum, while the other inhibitors did not.

Leiomyosarcoma of the spermatic cord. A case report and a brief review of literature.

A case of leiomyosarcoma of the spermatic cord, occurring in a 79-year-old male patient, is reported. The tumor was resected with high inguinal ligation and orchiectomy. It was located at the scrotal part of the spermatic cord on the left side and was shown to have originated from the spermatic duct using the dissecting method. Histologically, it was identical to leiomyosarcoma showing a typical morphological appearance of smooth muscle cell tumor in the conventional histochemical preparations as well as a focal positive reaction to antidesmin antibody, a marked nuclear pleomorphism, and abundant mitotic figures. The patient had been untroubled for six months after the resection of the tumor. Leiomyosarcoma arising in spermatic cord is quite rare.

A case of partial 5q trisomy associated with partial 7q monosomy.

A 1-year-old girl with partial 5q trisomy and partial 7q monosomy had ocular abnormalities that included bilateral blepharoptosis and Leber's congenital amaurosis. A single bright-flash electroretinogram (dark-adapted, white stimulation) disclosed subnormal bilateral responses. Her maculas showed a reddish spot surrounded by a broad, greyish retinal zone. Cytogenetic studies disclosed deletion of q22 to the terminal of chromosome 7 and partial trisomy of q31 to the terminal of chromosome 5. Because all reported patients with 5q trisomy have not had Leber's congenital amaurosis, the ocular abnormalities noted in our patient may be explained by the 7q monosomy.

Vaso-occlusive crisis episodes in sickle cell disease.

Seventeen adult males and females with Hb-SS, Hb-SC (1) and Hb-S Thal (1) hemoglobinopathies were continuously studied for 3 years. Various hematological and biochemical parameters were measured in the venous blood of the subjects for blood gases, CBC profile, blood chemistry (SMA-18), fibrinogen, alpha-HBD and myoglobin levels, percent sickling, blood viscosity, oxygen affinity of whole blood, osmofragility of red blood cells and calcium and zinc contents in plasma and in RBC. The results were compared between those subjects who encountered more frequent vaso-occlusive crisis episodes (frequent sicklers) and those with fewer crisis episodes (infrequent sicklers), along with parameters between crisis and non-crisis states of frequent sicklers. Our studies showed that percent sickling, P50 for O2, CBC profile, PO2, serum calcium, ALP, LDH, alpha-HBD level, zinc and calcium levels in plasma and in RBC varied between crisis and non-crisis states of frequent sicklers and also between frequent sicklers and infrequent sicklers. A logical explanation of such variations may help in understanding the etiology of vaso-occlusive crisis episodes in sickle cell disease.

Acidic lens-protein degrading activity in bovine ciliary body. III. Lens crystallin degradation by fractions A and B.

We studied the in vitro digestion of lens crystallin by two fractions (Fractions A and B) of acidic lens-protein degrading activity from the bovine ciliary body. With whole lens crystallin as substrate, Fraction A degraded beta L-crystallin most significantly, and beta H-, gamma-, and alpha-crystallins moderately. Fraction B degraded alpha-crystallin most significantly, beta H- and beta L-crystallins moderately, and gamma-crystallin slightly. Insoluble protein was most resistant to digestion by Fractions A and B. Crystallins from the lens cortex and nucleus were degraded in the same manner as those from the whole lens.

[Renal cell carcinoma in a long-term hemodialysis patient: a case report of its interesting clinical course].

A 61-year-old man with a history of hemodialysis for 10 years, and complaining of fever and right flank pain was introduced to us with the suspicion of right renal tumor and admitted on May 19, 1984. Right radical nephrectomy was done and the histological diagnosis was renal cell carcinoma (clear cell type) with acquired cystic disease of the kidney (ACDK). On the 27th day after operation, spontaneous rupture of bladder, despite its lack in functioning was suspected and an emergency operation was done. The bladder wall was very thin and weak and it was lacerated about 5 cm. After this operation the blood pressure was unstable and the patient died on the 64th day after nephrectomy due to sepsis. In Japan, 9 cases (containing this case) of renal cell carcinoma associated with ACDK were reported. The mean age and the mean duration of dialysis were 37.4 years old and 6.7 years. The cause of bladder rupture (1973-1983) in 87 cases is discussed.

[A case of non-resectable thymoma which responded to the administration of adriamycin, vindesine and 5-FU].

Surgery and radiotherapy have been considered to be the primary therapies for malignant thymoma. However, the results of these therapies for invasive thymoma are poor. Recently, combination chemotherapy for invasive thymoma has been tried and good results have been reported. A 50-year-old female was admitted to our hospital. Her chest film showed a large anterior mediastinal tumor which was diagnosed as invasive thymoma. In order to remove the tumor, median sternotomy and 4th intercostal thoracotomy was done but the mass was found to have invaded to the adjacent tissues, and so only partial resection of the tumor was performed. Postoperatively, the patient was treated with Adriamycin, Vindesine and 5-FU, as a result of which the mass completely disappeared and CR was achieved. Our regimen, AVF, for non-resectable thymoma is therefore thought to be a potentially effective treatment.

[Adult neuroblastoma: a case report].

A rare case of adult neuroblastoma is presented. A 20-year-old woman complaining of fever and left abdominal tumor was admitted on July 14, 1984 and diagnosed as left nonhormonal adrenal tumor after DIP, CT scanning, and hormonal assay in serum and urine. Left radical nephrectomy was done and the histological diagnosis was neuroblastoma. The chemotherapy of EDX, 5FU was done in 5 courses, but the patient had multiple metastasis and died on November 30, 1984. Neuroblastoma in the adult is rare and 40 cases (greater than 15-year-old) have been reported in the past 5 years, 20 of which were olfactory neuroblastoma. Particularly in urological sites, only 13 cases (greater than 15-year-old) have been reported for the past 20 years. Thus it is a characteristic of adult neuroblastoma that the occurrence in the retroperitoneal cavity is very low, compared with 60-70% of neuroblastoma in children.

Determination of alpha-keto acids in serum and urine by high-performance liquid chromatography with fluorescence detection.

A simple, rapid and highly sensitive high-performance liquid chromatographic method for the determination of alpha-keto acids in serum and urine is described. In dilute hydrochloric acid, alpha-keto acids are converted by 1,2-diamino-4,5-dimethoxybenzene into highly fluorescent quinoxalinone derivatives. The derivatives are isocratically separated simultaneously within 14 min by reversed-phase chromatography on a Radial-Pak cartridge C18 and detected fluorimetrically. The limits of detection are 10-300 fmol in an injection volume of 10 microliters (40-1200 pmol/ml of serum or urine). This sensitivity permits precise determination of several alpha-keto acids in 5 microliter of serum or urine from healthy persons, and also the determination of phenylpyruvic acid in normal urine which cannot be simultaneously determined by other methods.
