Interaction between dual NADPH-dependent reactions of paraquat in mouse brain microsomes.

Paraquat stimulated NADPH-dependent lipid peroxidation in mouse brain microsomes, while it suppressed lipid peroxidation induced by NADPH plus ascorbate. The stimulation of lipid peroxidation by paraquat was inhibited by N-ethylmaleimide (NEM). However, NEM attenuated the inhibitory effect of paraquat on lipid peroxidation elicited by NADPH in combination with ascorbate. These results suggest that an NADPH-dependent reaction which is susceptible to NEM might share an electron from NADPH with the alternative reaction, lipid peroxidation, in the presence of paraquat in mouse brain microsomes.

[A case of infiltrative transitional cell carcinoma of the kidney with hypercalcemia].

A 73-year-old man was admitted to our hospital with chief complaint of macrohematuria. Computerized tomography revealed enlargement of right kidney, tumor thrombi and lymph nodal involvement. Distant metastases were found in liver, lung and pelvis. We performed conservative therapy including right renal embolization. However, serum calcium levels continued to increase. Patient died from renal failure and pneumonia after 2 months, and autopsy was performed. Histological examination revealed infiltrative transitional cell carcinoma of the right kidney. PTH like peptide was measured 1 ng/gram wet tissue from the primary tumor and 16.6 ng/gram wet tissue from the metastatic liver tumor.

[Evaluation of modern therapy for neuroblastoma].

The clinical results were evaluated periodically in 50 cases of neuroblastoma, 4 survived cases out of 15 from 1965 to 1971, 6 survived of 16 (38%) from 1972 to 1982 and 11 survived of 19 (58%) after 1983. In the early period, before 1971, our systemic wide resection was not performed. From 1972, we operated on by our wide resection's technique which based on to preserve main vessels from tumor and lymph nodes. To separate the vessels from the lymph nodes was begun from both iliaca vessels to caeliac vessels. The systemic wide resection might contribute to improve survival rate from 1972 to 1982, however, all of 7 cases in stage IV did not survive. The best result after 1983, 58% survival rate, depends on new intensive chemotherapy accompanying with autologous bone marrow transplantation. All of 3 cases in stage III and 5 of 11 cases in stage IVA survived and better survival rates. Five alive cases in stage IVA received delayed wide resection after 3 or 4 cycles of new regimen A1, and then underwent bone marrow transplantation preconditioned by Melphalan .

[A clinical evaluation of AMS Hydroflex in the treatment of impotence].

The AMS Hydroflex penile prosthesis was implanted in 34 organic impotence patients between June 20, 1986 and Aug. 6, 1990. Patients ages ranged between 26 and 71 with an average of 50.5. The causes of impotences were; 17 cases of post-radical pelvic surgery, 7 cases of injury, 6 cases of diabetes and 4 of others. First evaluation was made after 12 weeks of implantation in terms of patient satisfaction with sexual intercourse and post operative complication. Sixteen patients (47.1%) were highly satisfied with intercourse, 13 (38.2%) satisfied with intercourse, 2 (5.9%) slightly dissatisfied even with successful intercourse, 1 (2.9%) with no improvement and 2 (5.9%) could not follow. Utility of the prosthesis, e.g. effectiveness and safety was; high utility in 20 (58.8%), good utility in 11 (32.4%), slight utility in 1 (2.9%), no favorable in 1 (2.9%) and no judgement in 1 (2.9%). The overall effectiveness and safety of the Hydroflex was demonstrated based on the above findings. Long term evaluation was made from the initial implantation of the device up to the termination of the trial with average of 19.2 months and was mentioned altogether.

[Results of the surgical treatment of patients with advanced small lung cancer].

From 1970 to 1988, 603 patients with non-small cell lung cancer were operated in our department. Among them, patients with small lung cancer (diameter less than 3 cm) with advanced disease were analyzed. There were 19 patients with mediastinal node metastasis, 2 with contralateral or supra-clavicular node metastasis, 3 with distant organ metastasis and one with pleural dissemination. In T1N2 group, adenocarcinoma was predominant in 17 patients and only 2 with squamous cell carcinoma. Sixteen patients underwent curative lobectomy with mediastinal node dissection. In T1N3 group, lobectomy and mediastinal and cervical node dissection were carried out. In T 1M1 group, 3 patients had lobectomy and removal of metastatic lesions, separately or simultaneously. One patient with pleural dissemination had segmentectomy and extirpation of disseminated foci. There were no operative deaths or hospital deaths in this series. Survival of the patients with T1N2 with curative resection was 93.8% in one year, 38.2% in 3 years and 22.9% in 5 years after operation. Among them, patients with intraoperative diagnosis of negative mediastinal node had relatively well survival. However, patients with N3, D+, or M1 had poor prognosis.

[A case of acute postinfarction mitral insufficiency and cardiogenic shock caused by total rupture of a papillary muscle].

A 61-year-old male in cardiogenic shock was transferred to our hospital with an intra-aortic balloon pumping (IABP). He had had acute posterior myocardial infarction previous day. He had developed progressively increasing dyspnea and physical signs of severe congestive failure. On admission, his blood pressure was 60 mmHg even with IABP. He was anuric with a high blood urea nitrogen and serum creatinine level. An echocardiogram demonstrated a rupture of a posterior papillary muscle and massive mitral insufficiency. His deteriorating condition did not allow us to perform cardiac catheterization. He underwent an emergency operation. Total rupture of a posterior papillary muscle was confirmed. Mitral valve replacement with a SJM prosthetic valve was performed. A postoperative course was uneventful.

Peptide behaviour and analysis on a chemically stable C18-bonded vinyl alcohol copolymer column with alkaline and acidic eluents.

C18-bonded vinyl alcohol copolymer (ODP) gel showed no weight loss or decrease in column efficiency for alkyl alcohols after being immersed in aqueous solutions of pH 2 and 10 at 50 degrees C for 48 h, and only a 1% weight loss and a slight decrease in alkyl alcohol retention volumes after similar immersion at pH 13. The chemical stability of the ODP gel was further demonstrated in analyses of acidic, neutral and basic peptides on an ODP column with eluents of pH 3-10, which showed that the peptides differ considerably in the sensitivity of their retention behaviour to eluent pH, even though hydrophobic interaction invariably appeared to be the main retention mechanism. The ODP column was therefore applied to the analysis and alignment of lysilendopeptidase (LEP) peptides derived from reduced and S-carboxymethylated carboxyl proteinase (Rcm-P-CP) of Pseudomonas sp. 101. One acid-soluble and seven alkaline-soluble LEP peaks were found in analyses on the ODP column using acidic and alkaline eluents, respectively. The chymotryptic peptides of Rcm-P-CP were first separated on an ODS column with an acidic eluent, and the eight eluates which contained lysine residue, as determined by amino acid analysis, were then analysed on the ODP column with an alkaline eluent, resulting in a further separation of each into several peaks and thus in the recovery of fractions of pure peptides. The LEP peptide alignment was then determined by overlapping the sequences of the chymotryptic peptides with the C- and N-terminal regions of the LEP peptides.

Monoclonal antibodies against human synovial phospholipase A2.

Four monoclonal antibodies (HP-1, HP-2, HP-3 and HP-4) with differing reactivities were raised against human synovial fluid phospholipase A2. None of them bound to exocrine phospholipases A2, such as those from pancreas or snake venom. However, antibodies HP-1 and HP-3 showed cross-reactivity with rabbit and rat platelet secretory phospholipases A2, which share common enzymatic and structural features with the human synovial enzyme. Antibodies HP-1, HP-2 and HP-3 inhibited the activity of human synovial phospholipase A2. The antibodies were used to develop a rapid immunoaffinity column chromatographic procedure for enzyme purification. In some preparations, the recovery of total activity after immunoaffinity column chromatography was more than 100% suggesting the existence of endogenous inhibitory factors of phospholipase A2 in human synovial fluid.

Cloning and sequence analysis of a cDNA for cellulase (FI-CMCase) from Aspergillus aculeatus.

We have cloned and characterized the cDNA coding for a major component of cellulase, endoglucanase (FI-CMCase), produced by Aspergillus aculeatus. The cDNA was isolated from a A. aculeatus cDNA library using synthetic oligonucleotide mixtures that correspond to the internal amino acid sequence of the mature FI-CMCase protein. Nucleotide sequence analysis of the cloned cDNA insert revealed a 711 bp open reading frame that encoded a protein of 237 amino acid residues. The primary structure of FI-CMCase deduced from the nucleotide sequence of cDNA agreed with that found by amino acid sequencing of peptide fragments obtained by digestion with several proteinases and cyanogen bromide cleavage. There may be a signal peptide sequence of 16 amino acid residues at the N-terminus. The molecular mass of the mature protein calculated from the cDNA is 24002 daltons, which compares favorably with molecular mass estimates of purified FI-CMCase obtained from SDS-PAGE (25000 Da). No distinct homology was found between the amino acid sequence of FI-CMCase and known cellulase sequences of other microorganisms. This study is the first example of cDNA cloning of an endoglucanase from the genus Aspergillus.

The effects of a new immunosuppressive agent, FK506, on the glomerular injury in rats with accelerated nephrotoxic serum glomerulonephritis.

The aim of the present work was to study the effects of a new immunosuppressive drug, FK506, in accelerated nephrotoxic serum glomerulonephritis. Glomerulonephritis was induced in female Wistar rats by the preimmunization with normal rabbit IgG (Day-4) and the subsequent intravenous injection of rabbit anti-GBM serum (Day 0). Without treatment with FK506, rats developed proteinuria at Day 6 and onward. Rat anti-rabbit IgG was strongly detected at Day 6 and the titer was maintained through Day 20. Moderate hypercellularity and focal crescent formation were observed at Day 20. Rats injected intramuscularly with 0.3 or 1 mg/kg of FK506 did not develop proteinuria and the anti-rabbit IgG titer was much less or was undetectable throughout the experiments. These data suggest that FK506 is effective in the present model of glomerulonephritis.

NADPH-dependent reaction of paraquat in mouse brain microsomes.

When paraquat was incubated with mouse brain microsomes in the presence of NADPH, a Nash-reagent-reactive substance (NRRS) (but not formalin) was produced. It was found that NRRS production was decreased in a dose-dependent manner by N-ethylmaleimide, a sulfhydryl reagent, which also inhibited NADPH-cytochrome P-450 reductase in parallel with the decrease in NRRS production. NRRS production was reduced by radical scavengers (catechin, glutathione, mannitol, superoxide dismutase and catalase), or under anaerobic conditions. In addition, inhibitors of adrenal cortex mitochondrial cytochrome P-450 (metyrapone, aminoglutethimide and amphenone B) inhibited NRRS production without causing a significant decrease in NADPH-cytochrome P-450 reductase activity. These findings suggest that active oxygen species and the mixed-function oxidase system may play important roles in NRRS production from paraquat in brain microsomes.

Postmortem changes in the levels of monoamine metabolites in human cerebrospinal fluid.

Four monoamine metabolites, 3,4-dihydroxyphenylacetic acid (DOPAC), homovanillic acid (HVA), 3-methoxy-4-hydroxyphenylglycol (MHPG) and 5-hydroxyindoleacetic acid (5-HIAA), were determined in the cerebrospinal fluid (CSF) of cadavers, whose causes of death had been suicidal hanging (SH) or ischemic heart failure (IHF). The concentration of DOPAC increased in parallel with the increment of the postmortem interval (PMI) (r = 0.626), whereas the concentrations of HVA, MHPG and 5-HIAA did not. The correlation coefficient was further increased by considering each cause of death separately: i.e., SH, r = 0.761; IHF, r = 0.705. These findings suggest the possible usefulness of the DOPAC level in CSF for estimating PMI.

A rapid purification procedure and computer-assisted sulfide ion selective electrode assay for O-acetylserine sulfhydrylase from Salmonella typhimurium.

An improved method for purifying O-acetylserine sulfhydrylase from Salmonella typhimurium is described as well as a new computer-controlled assay making use of the sulfide ion selective electrode. The purification method uses gradient elution from Q-Sepharose Fast Flow and phenyl-Sepharose columns to give 75 mg (50% yield) of the enzyme starting from 300 g of starting material in 3 days. The sulfide electrode assay makes use of sulfide and calomel electrodes attached to a signal buffer which serves as an impedance match. The output of the signal buffer is linked in parallel to a strip chart recorder and a Keithley Model 575 data acquisition and control system. The system 575 is interfaced to a Packard-Bell AT computer. In addition, two BASIC computer programs have been written to convert potential measured by the electrode to sulfide concentration and to convert the time course data to rates.
