Mitotic asynchrony of multinucleate cells in tissue culture.

A high degree of mitotic asynchrony is reported for spontaneously arising multinucleate cells of HeLa and Pt-K1, tissue culture cell lines. Neither nuclear number, nor nuclear size nor the presence or absence of nucleoli, could be directly related to the asynchronous behaviour of the cells. Suboptimal levels of nutrients led to a higher frequency of asynchrony. A partial compartmentation of the cytoplasm could be detected in some of the multinucleate cells. The compartmentation of the cytoplasm may lead to an uneven distribution of inducer and non-inducer metabolites. All these factors together may be responsible for the high degree of mitotic asynchrony.

Laparoscopy for the confirmation and prognostic evaluation of pelvic inflammatory disease.

A presumptive diagnosis of pelvic inflammatory disease (PID) is usually made in our gynecologic clinics when obscure, acute lower abdominal pain is accompained by fever and abnormal vaginal discharge, urinary and rectal discomfort, marked tenderness of the pelvic organs to palpation, or pelvic masses. In the present study, during a 2-year period, 223 women underwent laparoscopy to confirm this diagnosis. PID was confirmed in 103 (46.2%) of the cases; other serious conditions were diagnosed in 69 (30.9%) of the cases; and no evidence of disease was found in 51 (22.9%) of the cases. The authors conclude that laparoscopy is a valuable tool in the diagnosis of PID.

Studies on alkaline phosphatase in normal and nitrofurantoin resistant Vibrio el tor.

Activity of alkaline phosphatase as well as its substrate specificity in Vibrio el tor and in nitrofurantoin resistant Vibrio el tor have been studied. A lower level of activity is observed in Vibrio el tor after its acquisition of resistance towards nitrofurantoin. The enzyme activity in both the strains is significantly inhibited by EDTA, and also by metal ions like Mg++, Zn++ and Mn++. The normal strain is found to possess two isoenzymes for alkaline phosphatase whereas the resistant strain has only one isoenzyme.

Orientation of interphase chromosomes as detected by Giemsa C-bands.

The orientation of Giemsa C-bands has been studied in mitotic and interphase cells of Allium cepa. A sativum and of Aloe vera. The C-bands in these three species are located at the telomeres, secondary constriction region of the nucleolar chromosomes and the centromeric regions, respectively. Observations in A. cepa and Aloe indicate clearly that the interphase chromosomes are non-random in their orientation and possibly maintain their telophase configuration through the attachment of telomeres and perhaps of kinetochores with the nuclear membrane. Electron micrographs of onion cells also reveal that certain heterochromatic segments are associated with the nuclear membrane.--The nucleolar interstitial C-bands in A. sativum remain free in the nucleoplasm and may come close to each other due to heterochromatic attraction. Such a heterochromatic attraction is also evident between telomeric regions and between centromeres. However, a two by two attachment could not be noticed. A diagrammatic representation of the orientation of interphase chromosomes has been presented.
