Neovascularization in the pre- and postnatal rabbit corpora cavernosa penis: light and electron microscopy and autoradiography.

In the late prenatal stage, the rabbit corpora cavernosa penis (cavernous bodies) are occupied by a large number of mesenchymal cells actively proliferating by mitosis. Most profiles of the cavernous sinuses show growing capillaries consisting of immature endothelial cells and enclosing a narrow lumen. Such growing capillaries are often associated with the mesenchymal cells that contact with each other and eventually with the endothelial cells. Between 1 and 7 postnatal days, the capillary network develops extensively and the maturation of the capillaries results in a flattened endothelium with a continuous basal lamina. The mesenchymal cells are also associated with the helicine arterial sprouts at this time period. No endothelial mitotic figures were observed within the growing capillaries in pre- and postnatal specimens examined. Single pulse labeling with 3H-thymidine in the postnatal rabbits demonstrates that the labeled endothelial cells of the growing capillaries increase in number at 48 hours after injection compared with those at 24 hours, whereas the labeled mesenchymal cells inversely decrease in number. These findings indicate that the incorporation of the mesenchymal cells as vessel-forming cells into the growing capillaries accelerates the neovascularization in the rabbit cavernous bodies.

Cytotoxic T lymphocyte clone recognizing self idiotype: its regulatory role in antibody production.

MOPC 104E idiotype-specific cytotoxic T lymphocyte (CTL) clones were established. The clone KA1.02 specifically killed MOPC 104E cells and inhibited anti-dextran antibody production in an idiotype-specific manner. Both cytotoxicity and antibody inhibition were restricted to the cross-reactive idiotype and class I histocompatibility antigen. The data presented here provide definitive evidence that the self idiotype is the immunogenic determinant recognized by T lymphocytes in association with the major histocompatibility complex product. A regulatory role of CTL in antibody response is proposed.

Electron-microscopic and immunocytochemical analyses of Weibel-Palade bodies in the human umbilical vein during pregnancy.

The present study was done to elucidate the biological significance of the Weibel-Palade body of human umbilical vein endothelial cells. Quantitative determinations of these endothelial-specific granules throughout pregnancy revealed that their numbers and size per cell profile were maintained at low levels from 12 to 19 weeks of gestation; then both rapidly increased from 33 weeks to full term. This increase coincided with the development of the rough endoplasmic reticulum and an increase in the number of endothelial cell pinocytotic vesicles. Light-microscopic peroxidase anti-peroxidase and electron-microscopic protein A-gold techniques provided evidence that factor VIII-related antigen was localized in the Weibel-Palade bodies. Furthermore, in vitro treatment of incubated umbilical vein tissue with compound 48/80, a histamine releaser, induced degranulation of Weibel-Palade bodies from the endothelium. The present study indicates that Weibel-Palade bodies are storage sites of both histamine and factor VIII-related antigen and have an important role in the obliteration of this vessel.

The presence of an aberrant type of human chorionic gonadotropin in patients with gastric or colorectal cancer.

Beta subunits of human chorionic gonadotropin (beta-hCG) and human chorionic gonadotropin-like substance (hCGLS) were measured radioimmunologically in the serum and malignant tissue from patients with gastrointestinal cancer. Since serum beta-hCG and hCGLS correlate closely to those in cancer tissues, it is assumed that these two gonadotropins originate from cancer tissues. The serum hCGLS levels in 54 patients with gastrointestinal cancer were significantly higher, when compared with the findings in 19 healthy volunteers and 10 peptic ulcer patients. The frequency of high levels of serum hCGLS accounted for 71 per cent of those with operable gastric cancer, 44 per cent of those with inoperable gastric cancer, 100 per cent of those with operable colorectal cancer, and 67 per cent of those with inoperable colorectal cancer. On the contrary, serum beta-hCG levels did not differ between the volunteers and the cancer patients. In the 17 sera and 15 cancer tissues assayed, beta-hCG did not correlate to hCGLS. Moreover, the high levels of beta-hCG in cancer patients occurred in only 1/14 (7.1 per cent) of the assayed serum, and in 5/14 (35.7 per cent) of the cancer tissue. The increased production of these two hCGs may result from neoplastic transformation of an unrestrained fetal genome responsible for hCG production during gestation. It is assumed that the increased producibility of a defective hCG, i.e., an aberrant hCG such as hCGLS, is characteristic of malignant tumors.

Enhanced antitumor efficacy with a combination of hyperthermochemotherapy and thermosensitization with polyamine antimetabolites in nude mice.

In an attempt to enhance the antitumor effects of hyperthermochemotherapy, methylglyoxal-bis-guanylhydrazone (MGBG) and alpha-difluoromethylornithine (DFMO) were used in combination with hyperthermochemotherapy of 1-(4-amino-2-methyl-5-pyrimidinyl)-methyl-3-(2-chloroethyl)-3-nitrosoure a (ACNU) against human gastric cancer (ST-2) xenotransplanted into nude mice. After priming with DFMO and MGBG, ACNU was given ip and subsequently, a 23 minute-hyperthermia was carried out by placing the leg with the tumor into a water bath of a temperature of 43.5 +/- 0.1 degrees C. The second hyperthermic treatment was given in the same manner after 48 hours. MGBG and DFMO were administered for 4 successive days from the previous day of the first hyperthermia. In mice treated with DFMO plus MGBG, either tumor growth or tumor tripling time was much the same as in the control, while in mice given MGBG, DFMO plus heat, there was a diminution in tumor growth. Hyperthermia together with MGBG, DFMO plus ACNU brought about remarkable antiproliferative effects on ST-2 tumor growth, compared to three regimens with MGBG, DFMO plus heat, MGBG, DFMO plus ACNU, as well as ACNU plus heat. These data suggest that a combination of MGBG with DFMO leads to a favorable thermosensitization to the antitumor efficacy of ACNU.

Contradictory antitumor efficacies produced by the combination of DNA attacking drugs and polyamine antimetabolites.

The antitumor effects of two polyamine antimetabolites, alpha-difluoromethylornithine (DFMO) and methylglyoxal-bis-guanylhydrazone (MGBG), when combined with cis-diamminedichlroplatinum (CDDP) or mitomycin C (MMC), were studied using human gastric cancer cells xenotransplanted into nude mice. DFMO 1000 mg/kg and MGBG 50 mg/kg were given intraperitoneally for 6 successive days, while CDDP 3 mg/kg or MMC 2 mg/kg was given every second day. Although DFMO and MGBG plus MMC did suppress the tumor growth, the combination with CDDP led to no suppression, and rapid growth occurred after the cessation of therapy. The inhibition of tumoral DNA biosynthesis and a decline in polyamine levels, were also not observed. The polyamine antimetabolites when used with CDDP did not produce the desired antitumor efficacy, even though the platinum concentration in the tumor tissue was high. On the contrary, however, DFMO and MGBG when combined with MMC did suppress tumor growth, inhibited DNA biosynthesis, and tissue polyamine levels were low. These results suggest that though CDDP and MMC belong to a similar category of DNA attacking, bifunctional alkylating agents, the findings of these two drugs are contradictory. Here, the mechanism of action no doubt plays a contributory role.

Gastric cancer with synchronous unresectable hepatic metastasis and a positive response to chemotherapy--report of two cases.

We report herein the clinical events seen in two longterm survivors of gastric cancer with unresectable synchronous hepatic metastases. Partial gastrectomy was performed against the primary lesion and continuous intra-arterial infusion of anti-tumor drugs, followed by prolonged systemic antitumor drug treatment, were prescribed. A 43-year-old man with moderately-differentiated tubular adenocarcinoma had an uneventful course during his first 21 postoperative months. However, multiple liver metastases became manifest at 24 months and enlargement of the intrahepatic foci led to death 27 months after gastrectomy. A 51-year-old woman with moderately-differentiated tubular adenocarcinoma had an uneventful postoperative course and is now well with no signs of recurrence 6 years and 6 months after gastrectomy.

Immunocytochemistry of human chorionic gonadotropin in human chorionic villi.

The immunocytochemical localization of human chorionic gonadotropin was investigated in chorionic villi from the seventh to twelfth week of gestation. By the light microscopic peroxidase-antiperoxidase technique, positive reactions of human chorionic gonadotropin were found exclusively in the syncytiotrophoblast. Immunoelectron microscopy by means of the protein A-gold technique reveals localization of the immunoreactive gold particles in two kinds of membrane-bound granular inclusions in this cell; one type is granules of 200 to 300 nm in diameter with moderate electron density and the other is large electron-dense bodies of 500 to 1000 nm. The former seems to be Golgi-derived secretory granules that play a role in the release of human chorionic gonadotropin from the syncytiotrophoblast. Although the origin of the latter is still uncertain, a certain amount of this hormone might be stored or treated by lysosomal digestion in the large bodies during these stages.

Long-term-cultured mouse B-lymphocyte line. I. Establishment and characterization of mouse B-lymphocyte line.

Mouse B-cell line was established by culturing anti-Thy-1 antibody and complement-treated splenic B cells with the conditioned medium of concanavalin A-stimulated spleen cell-culture supernatant. At the eighth week of culture, it was revealed that 100% of the long-term-cultured cells had both cytoplasmic and surface immunoglobulin. These cells were then maintained in the conditioned medium together with T-cell-depleted splenic and then splenic adherent feeder cells. Flow cytometric studies of the B-cell line showed that they had surface mu, delta, and kappa but no gamma, lambda, Lyt-1, or Lyt-2. The growth of the B-cell line was dependent on the factor(s) derived from concanavalin A-stimulated conditioned medium. It was found that IL-2 was the major factor supporting the B-cell growth. The B-cell line did not secrete immunoglobulin spontaneously, but it could differentiate into antibody-forming cells through the stimulation of bacterial lipopolysaccharides. The technique for obtaining mouse B-cell lines are reproducible in our laboratory and one of those lines has been propagated and maintained for 16 months to the present.

Long-term-cultured mouse B-lymphocyte line. II. Modulation of Ia-antigen expression on B-lymphocyte line.

Mouse B-cell line, established by culturing anti-Thy-1 and complement-treated splenic B cells with concanavalin A-stimulated conditioned medium, expressed immunoglobulins and Ia antigens on its surface. The long-term-cultured B-cell line was split in two and maintained with or without 3300 R X-irradiated T-cell-depleted syngeneic splenic adherent cells (SAC). Interestingly, the B-cell line cultured without SAC lost its Ia antigen but not its Ig expression, whereas the cell line with SAC maintained both Ia and Ig expression. The ability to express Ia antigens was restored by culturing them only in the presence of Ia-positive feeder cells. Neither recombinant interferon-gamma or lectin-stimulated conditioned medium nor cell-free culture supernatant SAC had the ability to restore Ia antigen expression on the B-cell line. Incubation of Ia-negative B-cell line with phorbol esters restored the Ia expression. It is suggested that the expression of Ia antigen on B lymphocytes was controlled differently from that on macrophage lineage. The B-cell line expressing Ia antigens acts as stimulator cells for alloantigen-activated T lymphocytes and as antigen-presenting cells on the KLH-specific Ia-restricted proliferative T-cell clone in the presence of a specific antigen.

Factors associated with clinical reactions to rubella vaccination in women.

Two types of rubella vaccine were given to 272 seronegative healthy Japanese adult women. Clinical and laboratory studies on clinical reaction after vaccination were carried out on HI antibody response, HLA types, basal body temperature (BBT), menstrual cycles and serum progesterone levels of vaccinees. Clinical reactions were not associated with HLA types of vaccinees, but arthropathy occurred significantly more often in women vaccinated at the progestational stage than in women vaccinated at the estrogenic stage.

Technetium-99m serum albumin measurement of gastrointestinal protein loss in a subtotal gastrectomy patient with giant hypertrophic gastritis.

Gastrointestinal protein loss was measured using Tc-99m labeled human serum albumin in a patient with giant hypertrophic gastritis. Gastric secretion was aspirated via a nasogastric tube and measured for radioactivity after intravenous injection of Tc-99m albumin. Assessment of radioactivity of the collected gastric secretion yielded a total radiocount of 98.7 kilocounts per minute within 6 hours, which is equivalent to 1.1% of the total dose. Therefore, at least 1.1% of the circulating albumin was excreted into the gastric cavity within 6 hours, and, since simultaneous abdominal imaging did not demonstrate obvious accumulation of tracer in the gastrointestinal tract, protein loss was thought to be due to giant gastric rugae of the resected stomach. It was concluded that Tc-99m albumin is a valuable means for detection of the site of protein loss in patients with protein-losing gastroenteropathy. This method has several advantages in the clinical setting; it is less time consuming, easy to perform, and provides quantitative and qualitative assessment of protein loss.

Protection against Campylobacter jejuni infection in suckling mice by anti-flagellar antibody.

We obtained two monoclonal antibodies of IgM class and IgA class of immunoglobulin prepared from mouse spleen cells immunized with crude flagellar preparation, and a polyclonal antibody raised against purified flagellin monomer of Campylobacter jejuni in a rabbit. The specificity of the reaction of these antibodies for flagellar filament was confirmed by Western blotting and by immunoelectron microscopy. These antibodies caused agglutination of the bacteria and inhibited the motility of the bacteria. When a strain of C. jejuni was treated with IgM class monoclonal antibody before being inoculated into suckling mice, it reduced colonization of the intestinal tract by this bacteria. Inhibition of the colonization by IgA class monoclonal antibody was less effective than that of IgM class, and the polyclonal antibody consisting mostly of IgG class immunoglobulin was without effect.

Progressive dementia associated with Sheehan's syndrome--an autopsied case.

This is a report of the study of progressive dementia associated with Sheehan's syndrome in a 61-year-old woman who had a 13-year history of insidious progressive decline in global cognitive abilities. At the onset, there were only slight difficulties with abstract thoughts, and a shortened attention span, but the symptoms gradually increased in severity until death at the age of 61. The clinical deficits fluctuated in severity. During the last years of life she was at all times confused, apathetic, and incapable of self-care. A necropsy revealed a moderately severe neuronal loss in the central nervous system with marked destruction of the anterior pituitary lobe, and severe atrophy of the thyroid and adrenal glands. No senile plaques or neurofibrillary tangles were observed in the cerebral cortex. Possible pathophysiological mechanisms, which may have been present in this case, could be the hypometabolic effects on the central nervous system, mediated through a multi-endocrine system malfunction, caused by postpartum uterine hemorrhage and a marked disappearance of the pituitary anterior lobe cells.

Ultrastructural and immunological studies of virus particles observed in a T-cell line derived from a mouse spontaneous lymphoma.

Intracytoplasmic type A virus particles in clusters, associated with budding, immature type B and mature type B-like particles were observed in a T-cell line derived from a spontaneous lymphoma of an A/J mouse. These particles shared the antigenic determinants with murine mammary tumor virus from RIII, C3H, and GR strains as revealed by both indirect immunofluorescence and immunoperoxidase methods at ultrastructural level.

Immunocytochemistry on the localization of 5-hydroxytryptamine in monkey and rabbit taste buds.

Immuno-electron microscopy with the protein A-gold method demonstrated immunoreactive gold particles against 5-hydroxytryptamine localized in cored vesicles aggregating around presynaptic terminals of the gustatory cells in monkey and rabbit taste buds. The positive reactions were also found in the intragemmal and subepithelial nerve fibers. The role of these cored vesicles in taste transduction is still uncertain but the data strongly suggest that they may participate in a serotonergic modulation of a cholinergic synaptic transmission from the gustatory cells to the nerve endings.
