A new cell line having the ability to metastasize to the lungs in nude mice was established from GCH-1 (human choriocarcinoma cell line). Twenty CD-1 nude mice were injected with GCH-1 cells subcutaneously. A solitary metastatic pulmonary nodule appeared in only one of these mice. The pulmonary nodule was removed and minced, and the dissociated cells obtained were transplanted subcutaneously into new mice. By repeating this process, metastatic pulmonary nodules were seen clearly to have grown in size and number by the 4th transplantation. An in vitro cell line originating in the pulmonary lesion of the 5th transplantation was designated GCH-1 (m). Pulmonary metastasis occurred in all of the mice inoculated with GCH-1 (m) grown in vitro. The doubling time of the parent line GCH-1 and its subline GCH-1 (m) was 22.6 and 36.2 hours, respectively. GCH-1 (m) cells seemed to be heterogeneous in their size and shape compared with GCH-1 cells. GCH-1 (m) secreted more hCG and beta-hCG into the culture medium than GCH-1. By the immunoperoxidase technique using poly- or monoclonal antibodies, GCH-1 (m) showed a clear positive reaction but GCH-1 was only slightly positive for hCG and beta-hCG. Both GCH-1 and GCH-1 (m) were weakly positive for HLA-A, B,C and negative for HLA-DR, SP1 and hPL. Addition of the extracts from mouse lungs to the culture medium obviously promoted the growth of GCH-1 (m) but not of GCH-1.(ABSTRACT TRUNCATED AT 250 WORDS)

The effects of human chorionic gonadotropin (HCG), forskolin, cyclic nucleotides, the phosphodiesterase inhibitors IBMX and theophylline, cyanoketone, and cycloheximide on the production of estradiol-17 beta by isolated ovarian follicles of vitellogenic goldfish (Carassius auratus) were examined using 18-hr incubations. HCG and all test agents which are known to increase intracellular concentrations of cAMP significantly stimulated the production of estradiol-17 beta. However, dibutyryl cGMP was unable to stimulate estradiol-17 beta production at any concentration used (1-10 mM). Cyanoketone at a concentration of 1 micrograms/ml completely blocked forskolin-induced estradiol-17 beta production. Even in the presence of cyanoketone, however, forskolin stimulated conversion of exogenous testosterone to estradiol-17 beta in a dose-dependent manner, suggesting the involvement of an adenylate-cyclase system in the induction of aromatase activation by vitellogenic follicles of goldfish. Cycloheximide also completely abolished HCG-induced estradiol-17 beta production when this inhibitor was added within the first 1 hr after the addition of HCG. These results provide evidence that the stimulation of estradiol-17 beta by goldfish vitellogenic follicles in response to HCG is dependent upon the synthesis of new protein.
The success of fasciocutaneous flaps is based on the existence of epifascial vascular networks and reliable blood supply. However, there has been no thorough classification of the vascular anatomy of the fascia and skin and there is some confusion in regard to cutaneous vascular nomenclature. We divide the vascular systems involved in the cutaneous circulation into four categories. This permits classification of skin flaps into five types (cutaneous, fasciocutaneous, adipofascial, septocutaneous, and musculocutaneous flaps). Fasciocutaneous flaps can be further divided into six types, according to the patterns of the vascular input to the fasciocutaneous plexus. This classification has been demonstrated to be related to clinical effects. Nine new free and island flaps are discussed.

In order to determine the effect on the respiratory system of long-term inhalation of chromic acid in electroplating, histopathological examination was performed on mice (ICR, female, n = 50) exposed to chromic acid mist for 12 months (30 min/d, 2 d/wk), by using the authors' miniaturized chromium electroplating system. After 12 months' exposure, the mice were kept for 6 months without exposure. In the upper respiratory tract, round perforations were found in the nasal septum in 6 mice that were sacrificed or died after 10 months' exposure. The perforations were located near the point of the septum, the thinnest part of the nasal septum, and were of pin-hole sige. On the epithelium of the trachea and bronchus, loss of cilia, proliferation of goblet cells or basal cells, and squamous metaplasia were observed in almost all of the exposed mice. These changes appeared to be more advanced in mice exposed for a longer period. In the lung, development of benign adenoma was observed in one mouse in the 6- to 9-months' exposure group, in three in the 10- to 14-months' exposure group and in one in the group examined after 15 to 18 months. Furthermore, adenocarcinomas were found in two mice, one of which died at 17 months and the other was sacrificed 18 months after initiation of exposure. In conclusion, the finding that adenomas and adenocarcinomas were observed in mice exposed to chromic acid mist suggests the need to give careful attention to the possibility of respiratory cancers in chromium electroplating workers.

The cell lines designated KKNS-I and KKNS-II were derived from the metastatic lesions in the lymph nodes of endometrial adenocarcinoma. The KKNS-I cells were small spindle or polygonal in shape and showed a jig-saw puzzle-like arrangement with a tendency to pile up. The KKNS-II cells were polygonal in shape, and larger than the KKNS-I. They appeared to be arranged like a pavement. The modal number of the two cell lines was 46 without marker chromosome. Calculating from the growth curves, the doubling times for the KKNS-I and the KKNS-II cells were 35 hours and 60 hours, respectively. The tumors obtained from the nude mouse inoculated with the KKNS-I and the KKNS-II were a poorly differentiated endometrial adenocarcinoma and a well differentiated endometrial adenocarcinoma in histology, respectively. The volume of estrogen receptor and progesterone receptor in the KKNS-II were more than that in the KKNS-I cells. HLA-ABC antigens were detected in both the cells, but HLA-DR antigens were detected in only some populations of KKNS-II cells. These data suggest a relationship between the HLA-DR antigens and the degree of differentiation of the endometrial adenocarcinoma.
The study dealt with anti-tumor and adverse effects of etoposide and cisplatin on human choriocarcinoma cell line (GCH-1) transplantable in nude mice in relation to the rate of their uptake into tumor tissue. Nude mice were divided into 3 groups (etoposide, cisplatin and MTX-group), 7 to 9 per group and received drug treatment which started when their tumor grew to 100 approximately 300 mm3 in volume. All drugs were intraperitoneally administrated to nude mice (each drug: 1/4 mouse LD50 dose X3, weekly). The inhibiting action on the tumor was observed to be etoposide greater than cisplatin greater than MTX. The body weight loss of nude mice was observed to at its maximum in the cisplatin-treated group. The serum beta-HCG level did not rise in the drug-treated groups but rose in the non-treated control group. No histopathological changes characteristic of each drug were found. A concentration of etoposide and cisplatin in various tissues was serially measured after one shot intraperitoneal injection of etoposide 25 mg/kg or cisplatin 5 mg/kg. Etoposide reached its peak concentration after 30 minutes in tumor, liver and kidney and, after 4 hours, was left with 37.1%, 10.4% and 2.3% concentrations after 30 minutes in tumor, liver and kidney, respectively. Cisplatin showed similar kinetics, but was found to remain at a comparatively high concentration in both liver and kidney. Thus, it was demonstrated that etoposide and cisplatin were active against the human choriocarcinoma cell line. Furthermore, the less adverse effect of etoposide was felt to be partially due to its lower residue in liver and kidney.
