Relationships of bilirubin binding parameters.

The apparent unbound bilirubin concentration by the "peroxidase" method (U) and the total unconjugated bilirubin in blood (T), albumin-bound bilirubin (B), and reserve bilirubin binding capacity (R) by the bilirubin hematofluorometer were measured in 164 specimens from 98 neonates and in a series of artifactual specimens, made by adding bilirubin to the blood of a single adult donor. Linear correlations between U and (B/R) were found for both the prepared specimens (r = 0.99) and the clinical specimens (r = 0.87), with the slopes of the regression lines being close to the reciprocal of the albumin-bilirubin binding constant, a prediction of the mass action law. An excellent linear correlation was observed for the prepared specimens (r = 0.96) between U and (T--B), the concentration of bilirubin bound to low-affinity secondary sites ("loosely bound bilirubin"). A simple model for low-affinity binding of bilirubin in blood predicts this simple relation. A significant linear correlation between U and (T--B) was found for the clinical specimens, although the correlation was less good (r = 0.72), as one would expect. The demonstrated simple linear relationships between U, (B/R), and (T--B) support the hypothesis that both the hematofluorometer and peroxidase methods provide valid measurements of bilirubin binding status.

Bilirubin binding capacity and free bilirubin concentration: fluorescence quenching compared with peroxidase oxidation and sephadex column elution techniques.

Fluorescence quenching was compared with the techniques of peroxidase oxidation and Sephadex column elution for determining free bilirubin concentrations and the capacity of albumin to bind bilirubin and assessed as to its suitability for use as a routine method in a clinical laboratory. The poor reproducibility of the fluorescence quenching method made it unacceptable and peroxidase oxidation was found to be the most satisfactory technique. The Sephadex column elution technique did not measure free bilirubin concentration but gave a good estimate of binding capacity. This method is, however, limited by the sample size required for performing the determinations.

The description of structural and temporal characteristics of tonic electrophysiological activity during sleep.

The structural and temporal characteristics of tonic variation among electrophysiological measures during sleep are examined from the point of view of sleep stage scoring procedures and the digital quantification and statistical analysis of these measures. Sleep stage scoring procedures are nominally scaled and therefore less able to index structural and temporal characteristics than are measures derived from digital methods of quantification having the properties of interval or ratio scales. These arguments are illustrated using principal components analysis to describe the structure of tonic covariation among 36 variables derived from the computer analysis of EEG, EMG, and EOG. Three components resulting from the analysis are tentatively named 'slow-fast', 'hemispheric-shift' and 'eyes-activation'. They appear across variations in experimental manipulations such as night time awakenings and also across differences in age and species. The periodic characteristics of the vectors associated with the three components are specified by means of the Fast Fourier transform in combination with digital low pass and band pass filtering. Cycles are found which are both slower and faster than the paradigmatic 90 min ultradian rhythm. These results indicate that currently available techniques of digital and statistical analysis provide new possibilities for research on the electrophysiology and psychophysiology of sleep.

Effect of tocopherol on photooxidation rate of human erythrocyte membrane in vitro.

Study indicates that tocopherol's ability to prevent erythrocyte photohemolysis is a result of selective chemical trapping of the photooxidation-generated oxidant, singlet oxygen, before it can cause peroxidation of the essential unsaturated lipids in the erythrocyte membrane. The data provide fundamental information that support the purported clinical efficacy of tocopherol in the treatment of congenital erythrocyte membrane oxyhemolysis.

Non Xenopus-like DNA sequence organization in the Chironomus tentans genome.

We have examined the sequence organization of Chironomus tentans DNA by means of optical and hydroxyapatite renaturation kinetics of total DNA fragment sizes of 0.36, 2.6 and 13.5 kilobases (kb) as well as isolated middle repeat DNA at sizes of 0.36 and 13.5 kb. 90% of the DNA renatured as unique sequences of a genome of 0.20 pg with the balance of DNA renaturing as middle repetitive sequences present on averge 90 times per haploid genome. At a DNA fragment length of 13.5 kb, 35% of the DNA was trapped on the hydroxyapatite as middle repetiitive fraction. We concluded C. tentans DNA to have a mean repeat length of about 4.3 kb distributed throuth out at least 35% of the genome with an inter repeat spacing of at least 13.5 kb but possibly being distributed throughout the whole genome with an inter repeat spacing of 36 kb. This shows C. tentans DNA organization not to follow the most ubiquitous Xenopus model but to be similar to the organization of Drosophila melanogaster DNA.
