Method for reproducible large-volume production and purification of Rauscher murine leukemia virus.

Rauscher murine leukemia virus was produced in roller-bottle cultures of chronically infected JLS-V9 cells. Virus from this culture fluid was concentrated and purified by two semi-isopycnic bandings in sucrose gradients. Virus material obtained from young, nonconfluent cultures (early-harvest virus) yielded products characteristically containing endogenous ribonucleic acid-dependent deoxyribonucleic acid polymerase with high specific activity (400 to 1,000 pmol of [3H]thymidine 5'-triphosphate incorporated per milligram of protein per hour). Fluids obtained from older confluent cultures (late-harvest virus) yielded products with endogenous ribonucleic acid-dependent deoxyribonucleic acid polymerase with little or no specific activity (200 pmol or less of [3H]thymidine 5'-triphosphate incorporated per milligram of protein per hour), but with higher virus particle counts and greater amounts of protein and gs antigen than the early-harvest products.

Improved patient survival in renal transplantation.

Patient and graft survival in 655 consecutive renal transplants performed at the University of California, San Francisco, was analyzed in two separate groups to assess the results of the low-dose immunosuppressive regimen established in September, 1972. These results show that graft survival is not jeopardized by adopting a policy of low-dose immunosuppressive therapy, but, in fact, that patient survival is improved significantly. This study also shows that cadaver renal transplantation can be performed with a mortality rate comparable to or better than that in patients on chronic hemodialysis.

Introversion-extraversion and psychiatric diagnoses: a test of Eysenck's hypothesis.

Eysenck has stated that dysthymics were introverted and psychopaths were extraverted This study tested this hypothesis. The Ss were 28 male and 7 female inpatients from a state mental hospital. Twenty were diagnosed as neurotics, 15 as psychopaths. The sample was divided into extravert-introvert groups on the basis of scores on the Meyers-Briggs Type Indicator. Chi-square analysis revealed no significant differences in patient distribution by type and diagnosis. It was concluded that extraversion-introversion scores would not aid in the diagnosis of neurosis or psychopathy.

Prediction of kidney viability before transplantation.

A patient who receives a kidney transplant which fails suffers a physical and psychological disaster. Almost one-third of the kidneys transplanted in the United Kingdom are primary failures; many others function only after a delay, and the long term prognosis for these kidneys is poor. There is a need to identify kidneys which will function immediately after transplantation, those which will function after a delay and those which will never function. Such identification is seldom possible from the history. By continuous hypothermic perfusion we can identify a group of kidneys which will probably never function. The perfusate lactate level predicts which of the remaining kidneys will function immediately and which will not. Kidneys which function immediately after transplantation have a very much better long term prognosis after transplantation than those with delayed onset of function.
