[Glucose disposal in very low birth weight infants ].

Glucose disposal was studied in six very low birth weight infants (birth weight 1093 +/- 67 g (M +/- SEM), gestational age 29.8 +/- 0.6 weeks, postnatal age 5.2 +/- 0.9 days) by constant infusion steady state technique. All babies required artificial ventilation because of respiratory distress syndrome (four) or aspiration pneumonia (two). Nine healthy prematures of comparable birth weight, gestational age, and postnatal age formed the control group. During a constant glucose infusion at a rate of 8 mg/kg/min (7.7 +/- 0.2) mean blood glucose levels of the sick infants were higher than the corresponding control values. All sick babies had glycosuria (glucose excretion 34 +/- 19 mg/kg/hr compared to none of the controls 0.8 +/- 0.2, p less than 0.01). Glucose disposal in the sick prematures was significantly decreased in comparison to the healthy infants [92.6% (78.7 to 99.3%) of input rate vs. 99.8% (99.7 to 99.9%), p less than 0.01]. It is concluded that sick very low birth weight infants tend to develop hyperglycemia, glucosuria, impaired glucose disposal, and renal caloric wasting when exogenous glucose is supplied at a rate of 8 mg/kg/min, which is usually well tolerated by healthy prematures.

Defective expression of mouse adenovirus Fl in human cells.

HeLa cells infected with mouse adenovirus strain Fl ( AdFl ) produce at least 2000 times less virus than permissive mouse 3T3 cells. Viral DNA synthesis, however, proceeds unimpaired. The defect in virion production was linked to a dramatic reduction in the synthesis of AdFl structural proteins, in particular the hexon. The identity of the AdFl hexon gene product was recognized through its immunogenic reactivity towards an antiserum raised against the human adenovirus type 2 (Ad2) hexon gene product. This cross-reactivity is reflected in an extensive DNA sequence homology between AdFl and Ad2 DNA at position 53-60, the locus of the hexon gene, on the Ad2 physical map. Through hybridization at different formamide concentrations, the present study identifies one additional, highly conserved region at map positions 14-15 on the Ad2 genome.

[Phosphorus and calcium metabolism of premature infants fed human milk and formulated milk].

Development of rickets in vitamin-D supplemented very-low-birthweight (VLBW) infants can be associated with low Phosphorus (P) and Calcium (Ca) intake. Therefore, we estimated P and Ca intake of three VLBW-infants who were fed pooled human milk after birth and then switched to an adapted formula. We measured P, Ca, activity of alkaline phosphatase and parathormone in serum and P and Ca excretion in urine. P and Ca intake was below the estimated requirement for prematures with pooled human milk as well as with the adapted formula, on the other hand, Ca and especially P intake increased when infants were switched to the adapted formula. When fed human milk we found evidence for a P deficiency in the body with signs of inadequate bone mineralisation. On the adapted formula infants showed Ca deficiency but bone mineralisation improved. From our findings we conclude that on pooled human milk VLBW-infants do not receive the required amount of P and Ca. The signs of Ca deficiency on the adapted formula might be related to low Ca intake or to the P: Ca ratio of the formula which could be inadequate for VLBW-infants.

Incidence of plant poisonings in Philadelphia noted as poison information calls.

During a one year period, plant calls were 5.3% of the total number of poison information calls. Of all confirmed plant exposures, 6.8% developed symptoms. Of all suspected plant exposures, 3.2% developed symptoms. Most (75%) of plant exposures occurred in children 3 years old and under. There was no difference between the sexes in exposures or symptoms.

In vitro translation of adenovirus type 12-specific mRNA isolated from infected and transformed cells.

The early and late gene products of human adenovirus type 12 (Ad12), as well as the viral proteins synthesized in an Ad12-transformed cell line, were identified by translation of viral mRNA in an in vitro protein-synthesizing system. Cytoplasmic RNA was isolated from permissive KB or nonpermissive BHK cells infected with Ad12 and from Ad12-transformed HA12/7 cells. Virus-specific RNA was selected by hybridization to Ad12 DNA covalently bound to cellulose. Viral RNA was then translated in a fractionated rabbit reticulocyte cell-free system or in wheat germ S-30 extracts. The proteins synthesized were characterized by immunoprecipitation and subsequent electrophoresis on sodium dodecyl sulfate-polyacrylamide gels. RNA prepared from KB cells late after infection with Ad12 elicited the synthesis of most of the structural polypeptides of the virion and at least two presumably nonstructural Ad12 proteins. When viral RNA isolated early after infection of KB cells with Ad12 was translated in vitro, 10 polypeptides were observed: E-68K, E-50K, E-42K, E-39K, E-34K, E-21K, E-19K, E-13K, E-12K, and E-10K. Ad12-specific RNA was also isolated from the Ad12-transformed hamster cell line HA12/7, which contains several copies of the Ad12 genome integrated in the host genome. The RNA codes for at least seven polypeptides with molecular weights very similar to those of the early viral proteins.

Adenovirus type 2 DNA replication. II. Termini of DNA replication.

Complete, mature adenovirus type 2 DNA molecules were isolated from virus-infected HeLa cells, pulse-labeled at 20 h postinfection in [3H]thymidine pulses shorter than the time necessary for one round of viral DNA replication. After digestion with the restriction endonucleases Eco RI, Hpa I, and Hind III, a temporal order of synthesis of different regions of the viral genome was established from the relative specific radioactivities in the restriction enzyme fragments. A comparison with the physical order of these fragments revealed the existence of two termini of DNA replication towards both the molecular right and left ends, respectively, of the viral chromosome.

[Reduction of body height in tall girls by estrogen treatment].

Twenty-six girls with constitutional tall stature were treated with 0.5 mg ethinyloestradiol daily po and 10 mg norethisteronacetate from day 21 to 25 of the cycle for 1.4 +/- 0.4 years. Height, weight, bone age according to Greulich-Pyle (GP) and Tanner (RUS), height predictions according to Bayley Pinneau (BP) and Tanner Whitehouse (TW) were recorded at the onset of therapy, at the end of treatment and 7.4 years later. The mean chronological age at start of treatment was 12.6 +/- 1.1 years, bone age (GP) 12.4 +/- 0.7 years, (RUS) 13.5 +/- 0.7 years. The initial height prediction according to BP was 186.2 +/- 4.1 cm, which was 3.5 cm higher than the prognosis according to Tanner Whitehouse. Depending on the method used for height prediction, the height reduction by treatment was 4.6 +/- 3.0 cm (BP) or 1.4 +/- 2.2 cm (TW), respectively. The reduction of predicted height was more pronounced in girls with a bone age (GP) < 12.5 years at the initiation of therapy (n = 13), than in girls with a bone age > or = 12.5 years (n = 13). Final height was measured 7.4 years after the end of treatment. Chronological age: 21.3 +/- 3.7 years, final height 181.8 +/- 3.7 cm. Mean final height after therapy was 7.7 cm above target height, or, after allowance for the secular trend, 3.8 cm above target height. Body weight, recorded in centiles of body mass index, increased from the 38.5 +/- 26 to the 58.1 +/- 22 centile, weight gain was more pronounced in younger girls. At follow up investigation 18/19 girls, who answered a questionnaire, had a positive view of the previous treatment.
