High and low Fc IgG-receptor expression in human chronic granulocytic leukaemia cells.

Discontinuous density-gradient centrifugation was used to separate chronic granulocytic leukaemia (CGL) cells in the chronic phase and blast crisis (BC) into fractions containing granulocytes in individual stages of maturation. The occurrence of the Fc IgG (FcR) and complement-component receptors (CR1 and CR2) in each fraction was estimated. It was established that, with increasing yields of mature granulocytes, the proportion of cells bearing Fc and C3 receptors increased. The most important finding was that the high- and low-receptor categories of CGL cells in chronic phase depended on the percentage of FcR+ cells. In the high-receptor CGL group, in addition to FcR, the proportion of CR1+ and CR2+ cells was also greater than in the low-receptor CGL group. Some differences in clinical course of both immunological CGL groups were observed.

[The patellar focus, a rare form of localization of bone tuberculosis].

Patellar foci of any origin are very rare. Form 1955 to the end of 1979, 9 patients underwent patellar surgery in our hospital. 7 of these patients had tuberculosis, whereas in one case the patient had chronic nonspecific osteomyelitis, and in one case we were unable to clarify the underlying cause of the focus, neither from the histological nor from the bacteriological aspect. All tuberculous foci, one of which had penetrated into the knee joint, were situated in the corpus of the patellar. In a 10-year old girl, the entire patellar was infected with tuberculous foci. In the anterior-posterior x-ray film, the processes mainly presented a circular appearance. Their diameter different between 6 and 25 mm. In one of the foci, two sequesters were conspicuous, and one in another patient. 6 of the patients had an active organ tuberculosis at the same time, or reported on a specific disease during anamnesis. The period from the onset of complaints until diagnosis had been established, was on the average 14.4 months with the 7 patients under investigation (average age 29 years, 4 female/3 male). In view of the fact that all the patients reported here were late cases, the results of the combined drug and surgical removal of foci (filling of the surgical cavities with spongiosa) must be classified as good. One knee joint had to be stiffened by operation. There were no reactivations within an average followup period of 3 years.

[Isolated ilial tuberculosis and its differential diagnosis].

Isolated tuberculous foci in the ilium are particularly rare. Over a period of 26 years (1955 to 1981) we have treated ten lesions in the ilium in our Clinic. Only two (one male, one female) were of tuberculous origin, the other consisted of non-specific osteomyelitis, one sarcoma and one lesion in a Hand-Schüller-Christian syndrome. In the discussion, the literature is reviewed concerning the frequency, features, treatment, prognosis and differential diagnosis of specific infections of the ilium.

Poly(adenylate) polymerase activity of rat brain.

The poly(adenylate)[poly(A)] polymerase of rat brain, as in rat liver, is located primarily in the nuclear sap when nuclei are prepared under hypertonic conditions. The enzyme can be released from nuclei in two forms. Form I is prepared by gentle incubation of nuclei at 0 degrees C in hypotonic buffer. It has a Mn optimum of 0.6 mM and a pH optimum between 8 and 9. The ATP concentration curve plateaus at 0.2 mM. The optimal poly(A) primer concentration is 600 micrograms/ml, which is three times higher than that for the enzyme similarly prepared from liver. The time course of the reaction for the form I enzyme is increasing over the first 40 min and becomes nearly linear thereafter. Form I is not stimulated by either calcium or cyclic nucleotides, but is inhibited by polyamines, pyrophosphate, and high concentrations of GTP. Form II enzyme is prepared by homogenization of nuclei in hypotonic buffer. It has the same ATP and poly(A) optima as the form I enzyme but displays linear kinetics over a 60-min time course. It is slightly stimulated by cGMP and cAMP and strongly inhibited by spermine, sodium pyrophosphate, and high concentrations of GTP.

[Concept for the total care of families of children with leukemia and tumors based on conversations with the parents].

In order to create a foundation for prophylactic and therapeutic measures the problems of children with leucaemia and other malignancies and of their families were assessed in detail with parents of these children in single or group discussions. The necessity for integrated therapy including families of children with leucaemia or tumours became apparent. Care should include extensive talks with all family members and also with teachers of patients and of healthy siblings, the use of aids such a painting or puppet theatre for recognition of problems and needs, particularly in small children, home visits to explore problems of health siblings, involvement of social workers, aftercare for the whole family after the death of a child, and, finally the help for overcoming problems of the treating team. Care to such an extent has been found to be necessary and requires an increase in posts in oncological units. Lack of time for the patients as human beings is unacceptable even from an economical point of view. A psychotherapeutically versed medical doctor or a clinical psychologist and a social worker should be permanent members of any oncological team.

Improving classroom behavior in mentally retarded children through correspondence training.

Except for a few studies, most research investigating correspondence training procedures has been more analogue in nature. The purpose of the present set of studies was to examine whether a "say-do" correspondence training technique could be used with children in special education classes to improve classroom behavior. The specific behaviors targeted for change included: out-of-seat behavior (Experiment 1), sitting posture (Experiment 2), and on-task behavior (Experiment 3). The say-do procedure used in Experiment 1 resembled that of previous studies, whereas that in Experiment 2 was more elaborate in the specificity of verbal statements required from the children and the feedback given them. The training procedure in Experiment 3 used a format similar to the say-do approach, but stressed visual rather than verbal cuing because it was used with nonverbal children. All three studies used single-subject designs and examined maintenance and/or generalization questions. Experiments 2 and 3 also evaluated whether concomitant changes in performance on academic tasks occurred. The results of the three studies provide strong evidence that correspondence training can be effectively used with educationally handicapped children. Moreover, the successful modification of the "say-do" to a "show-do" procedure in Experiment 3 points out the flexibility of the correspondence training approach.

[On the role of substance P in thermoregulation of normotensive and spontaneously hypertensive rats (author's transl)].

Various peptides have been shown to alter body temperature. Intraperitoneal administration of substance P (SP) in doses of 25 and 250 micrograms/kg produces a significant hypothermic response in normotensive rats under cold exposure at 4 degrees C, whereas a dose of 2.5 micrograms/kg causes an increase in body temperature. Pretreatment with naloxone (4 mg/kg, 30 min before SP) suppresses the hypothermic effect. In spontaneously hypertensive rats (SHR), however, SP does not alter the body temperature. This result points to an altered thermoregulation in SHR based on changes in endorphin systems.

Effects of substance P and shorter analogs on body temperature in the rat.

The thermoregulatory effects of intraperitoneally administered substance P (SP1-11) (250 micrograms/kg), or equimolar quantities of SP1-4 and SP5-11 were studied in conscious rats at an ambient temperature of 4 degrees C. SP1-11 produced hypothermia, whereas SP1-4 and SP5-11 had no influence. The hypothermic effect of SP1-11 was partially antagonized by naloxone. Thus, an opiate-dependent step seems to be involved in the action of SP1-11.

Distribution of membrane potential of aortic media cells and fibroblasts in culture.

The distribution of membrane potential (Em) of aortic media cells and fibroblastoid kidney cells (both derived from calf and monkey) was investigated by using dense cell layers. Fibroblastoid kidney cells showed a symmetric distribution of Em with a mean of -10.6 +/- 0.29 mV for calf and -12.1 +/- 0.31 mV for monkey (m +/- sm). In aortic media cell lines, however, a wide bimodal distribution of Em was found (between -4 mV and -32 mV). The peaks were at about -17 mV and -27 mV in calf aortic media cells, and at about -13 mV and -22 mV in monkey aortic media cells. The bimodality of Em distribution of cultured aortic media cells may be typical for this cell type. Presumably, this pattern reflects two cell populations in different states of differentiation or maturation.

Age-associated changes in poly(adenylate) polymerase of rat liver and brain.

The activity of poly(adenylate) polymerase was measured in nuclear lysates of rat brain and liver of animals ranging from 25 to 300 days. The activity was 26 nmoles AMP incorporated per h per mg of protein in liver and 8 moles per h mg in brain for 25-day-old rats, and declined to 1.0 and 3.3 for liver and brain, respectively, in 300-day-old rats. At all ages the liver showed a linear time course and had a Km for ATP of 0.5 mM. The brain enzyme gave a non-linear time course at all ages with an ATP optimum of 0.2 mM. Mixing enzyme preparations from young and old animals produced additive results. Mixing brain and liver enzyme produced greater than additive results. No substantial increase in activity was observed when enzyme was prepared from old or young partially hepatectomized rats as compared to sham-operated controls.
