Hepatocellular dysplasia: immunohistochemical and morphometrical evaluation.

Hepatocellular dysplasia, first described by Anthony et al. [J. clin. Path. 26: 217-223, 1973], is considered a peculiar pattern of proliferation process mainly observable in cirrhotic nodules in patients with hepatocellular carcinoma. Its precancerous meaning has been variously evaluated in the past. In the present study, immunohistochemical data concerning the presence of alpha-fetoprotein, alpha 1-antitrypsin, carcinoembryonic antigen, hepatitis B surface antigen and hepatitis B core antigen did not show meaningful differences between carcinomatous cells and normal and dysplastic hepatocytes. On the contrary, morphometric analysis seems to be useful to discriminate dysplastic cells by means of parametrical indexes of shape and symmetry of the nuclei and could probably offer in the future an objective evaluation of hepatocellular dysplasia.

[Effects of tubulo-deferens stenosis secondary to intraserosal proximal hemideferentectomy and end-to-end tubulo-vasostomy on the testicle of the rat. Histopathological aspects].

The histopathological aspects of the testis in rat after intra-serous proximal hemideferentectomy and end-to-end tubulo-vasostomy are evaluated. The histomorphometric variations of the convoluted seminiferous tubules and the quantitative modifications of the stages of spermatogenesis are correlated with the obstruction's degree of the tubulo-deferential anastomosis.

[Parameters of analytic morphometry: proposal for a symmetry evaluator].

A morphometrical analytical evaluator of asymmetries of biological structures bonded by a closed curve was suggested. A second degree equation couple was found in order to detect, respectively according X and Y axis, first main asymmetry whereas third degree equation revealed first and second asymmetry for both abscissa and ordinate values. Final evaluator, named S.A.E. (Shape Analytical Evaluator), was computed as arch/cord ratio between function curve and its cord, it was equal to 1 for circles and any ellipse and symmetrical shapes whereas it was of different value for asymmetrical shapes.

Morphometric analytical evaluators of lymphoid populations in nonneoplastic lymph nodes.

A morphometric analysis using an Apple II microcomputer and shape analytical morphometry (SAM) software was performed of the nuclear shape in the lymphoid population of normal (nonneoplastic) lymph nodes to provide baseline parameters useful for distinguishing between different subtypes of lymphomas. Fourier harmonic analysis and two indices, a contour asymmetric evaluator (CAE) and a shape asymmetric evaluator (SAE), are proposed as non-dimensional shape parameters. Results obtained with the CAE and SAE indices indicate that they represent sensitive shape descriptors able to reveal even subtle irregularities of nuclear outline. With these parameters, centrocyte nuclei were easily differentiated from those of all other lymphoid cells while an interesting close relationship was found between lymphocytes and centroblasts. Lymphocyte nuclear shape was shown to be less regular than generally reported, suggesting the existence of subtle differences between stimulated and unstimulated nuclei. Immunoblast nuclei were shown to have the most regular shapes.

[Changes in the transparency of dentin: computerized densitometric analysis].

Computerized densitometry was utilized to carry out a quantitative and distributive study of dentin transparency in dental roots from different age individuals. By means of original software S.A.M. (Shape Analytical Morphometry) a densitometric reading of transilluminated whole dental root was performed and so obtained densitometric matrixes were worked for grey level classification and correspondent areas evaluation, false colour restitution, pseudotridimensional and row/column barr graphs, isodensity outlines and density inclination maps. Results are discussed for both age evaluation in forensic application and for altered dentin recognition in pathological conditions.

[Definition of the analytical relation between the surface and internal perimeter of seminiferous tubules from subjects with normal spermatogenesis].

Luminal tubular measurements are executed in testis with normal spermatogenesis to individuate, by computerized morphometric method, the ratio between these two values. Statistical evaluations and calculation of the regression curves demonstrate that this ratio is well described by means of an exponential equation which coefficients are determined. This parameter results very useful in the diagnosis of Sertoli cell only syndrome and maturation arrest both characterized by tubular lumen modification.

[Description of the shape of carcinomatous nodules of the breast by means of high degree polynomial equations and Fourier harmonic analysis].

Breast cancer nodule shape has been investigated by two analytical procedures: upper degree polynomials and harmonic Fourier analysis. An automized method (TV camera interfaced with a computer) has been employed to find the coefficients of kth order equation giving the best fit. Then the difference between real node contour and function curve has been calculated and submitted to harmonic analysis. This gives the sine/cosine coefficients of the two series and the amplitude of the harmonics. The results point out that no difference exists between metastatic and non metastatic tumors and suggest that parametrized descriptors calculated may represent a peculiar behaviour in the growth of the tumor.

[Simulation of the metastatic spread of mammary carcinoma by evaluation of the shortest routes in a computerized model of the lymphatic system represented in a three-dimensional map].

A simulation model of mammary lymphatics has been realized by a computerized system to draw a tridimensional map of all lymphnode stations (axillary, supraclavear and internal mammary lymphnodes) draining from the breast. The purpose was to identify the minimal routes between tumor and lymphnodes and among different nodal points that represent lymphnodes of the lymphatic net situated at various distance from tumor. A simulation of node metastatization has been performed giving to every route of the model a different weight represented by the distance between the nodes.

The histologic typing of tonsillar carcinoma.

In a retrospective review of 170 cases of malignancies of the tonsil the authors report on 81 malignant epithelial tumors. In 56 cases (69.1%), the tumor (squamous cell carcinoma) was composed of solid epithelial nests with different degrees of keratinization. In 24 cases (29.6%) the tumor (undifferentiated carcinoma of nasopharyngeal type) had histologic characteristics similar to the undifferentiated carcinomas observed in the nasopharynx. Only one case was unclassifiable. Squamous cell carcinomas occurred more frequently in males (6:1), in older age, with more frequent extratonsillar and nodal extension and a poorer prognosis. Undifferentiated carcinoma had a similar incidence in both sexes, occurred also at an early age, and more often seemed to be limited to the tonsil. The 10-year disease-free survival rate was 30% for undifferentiated carcinoma; all the patients affected by squamous cell carcinoma had a recurrence within 6 years of the diagnosis.

[Analytical morphometry of seminiferous tubules in maturation arrest: Fourier analysis of the internal contours of tubules].

Shape of seminiferous tubuli in maturation arrest, is investigated by harmonic Fourier analysis and upper degree polynomials as attempt to find morphometric parameters useful in morphological diagnosis. Inner luminal contour is considered as a closed curve in a reference system and scattered in a series of points whose coordinates are automatically calculated. Then a software named S.A.M. (Shape Analytical Morphometry) is utilized to find the Kth-order equation and calculate the best-fit curve. Moreover an ellypse is drawn with the same barycenter and axes of original curve. These three curves (real curve, best-fit curve and ellypse) are considered as periodic functions and submitted to Fourier analysis to evaluate the coefficients of the series and the spectrum of harmonics. Among 20 contributors, the subsets of greatest amplitude are selected for comparison and classification in terms of fundamental shape and its perturbations.

[Morphologico-morphometric relief of the testis in Down's syndrome].

Four cases of Down's syndrome in new-born have been thoroughly studied on the histological and morphometrical point of view. It is outlined the presence in the seminiferous tubules of abnormal spermatogonia, sometimes multinucleated and characterized by a prominent nucleolus. The Authors emphasize the conspicuous number of these hypertrophic (H) spermatogonia (usually very scanty in normal new-born) never reported in the Down's syndrome before, whose significance is not easy to explain: if they represent anomalous forms of spermatogonial cells, related to the malformative condition, or a premalignant aspect is then discussed. Morphometrical measurements of the average tubular diameter (D.T.M.) and spermatogonia content have been calculated; furthermore a microprocessor system is employed for evaluating nuclear diameter, perimeter and area not only in H spermatogonia, but also in the other germ cells observed (A and P).

Morphologic and classificatory considerations about 140 cases of carcinoma of the nasopharynx.

The morphologic aspects are reported for 140 cases of nasopharyngeal carcinoma collected from a retrospective review of malignant tumors of this region. Of 97 cases of undifferentiated carcinomas of nasopharyngeal type, 63 were composed of solid cords of epithelial cells (Régaud type), 22 showed isolated cells in a lymphoid stroma (Schmincke type), and in 12 the neoplastic cells were aligned with a fibrosarcoma-like aspect (spindle-cell type). The 36 cases of squamous cell carcinoma showed various degrees of differentiation. Of the remaining 7 cases, 3 were transitional cell carcinomas, 1 was an adenoid cystic carcinoma, and 3 were unclassifiable carcinomas. The undifferentiated carcinoma was present in all ages, with a male : female rate of 1.27 : 1. The squamous cell carcinomas were prevalent in males (3.5 : 1) and older patients. The 10-year survival rate was 30% in the group of patients with undifferentiated carcinoma, whereas patients with squamous cell carcinoma died within 4 years of the diagnosis.
