Synchronization of estrus in embryo transfer recipients after using a combination of PRID or CIDR-B plus PGF2alpha.

The use of CIDR-B or PRID in combination with prostaglandin F2alpha (PGF2alpha) for synchronizing estrus in embryo transfer recipients was evaluated in 2 experiments. In Experiment 1, virgin heifers (n=263) were synchronized using either a PRID (including estradiol benzoate capsule) or a CIDR-B in a combined program in which devices were inserted on Day 1, an injection of prostaglandin was given on Day 6, and devices were withdrawn on Day 7. The interval from device removal to the onset of estrus was significantly shorter for CIDR-B than for PRID-treated animals (50.44 vs 55.50 hours; P<0.003). The CIDR-B treatment resulted in a similar degree of synchrony to the PRID treatment (74.0 vs 70.4%; P=0.68). InExperiment 2, cows (n=95) and heifers (n=93) were allocated at random to be synchronized using a PRID (excluding estradiol benzoate capsule) plus PGF2alpha or a CIDR-B device plus PGF2alpha. The devices were inserted on Day 1, an injection of prostaglandin was given on Day 10 and the devices were removed on Day 12. Estrus was observed earlier following the CIDR-B treatment (43.50 vs 47.04 hours; P=0.01), but the degree of synchrony was similar (76.2 vs 76.3%; P>0.10) for the CIDR-B and PRID-treated animals. In both experiments, there were no significant differences in the proportions of animals observed in estrus, selected as embryo transfer recipients, or which achieved pregnancy consequent on embryo transfer between those synchronized using CIDR-B or PRID regimens. We conclude that the CIDR-B is a suitable device for synchronizing estrus in embryo transfer recipients.

Potentiation of aggregation and inhibition of adenylate cyclase in human platelets by prostaglandin E analogues.

1. The 16-phenoxy prostaglandin E analogue sulprostone consistently potentiates primary aggregation waves induced by adenosine 5'-diphosphate (ADP), PAF and 11,9-epoxymethano PGH2 (U-46619) in platelet-rich plasma from human donors. The effect is not blocked by the TP-receptor antagonists, EP 092 and GR 32191. The high potency of sulprostone (threshold concentration = 4-10 nM) and the weak block of sulprostone potentiation by the EP1-receptor antagonist, AH 6809 (pA2 = 4.3) suggest the involvement of EP3-receptors as opposed to EP1- or EP2-subtypes. 2. Eight prostaglandin E (PGE) analogues were compared against sulprostone for their effects on PAF-induced aggregation in human platelet-rich plasma (PRP) in the presence of GR 32191 and the DP-receptor antagonist, BW A868C. PGE2 and 11-deoxy PGE2-1-alcohol showed evidence of both potentiating and inhibitory actions and butaprost showed only inhibitory activity at high concentrations. The remaining analogues always elicited potentiation, with the following potency ranking: sulprostone = 16,16-dimethyl PGE2 > MB 28767 > misoprostol > GR 63779X = 17-phenyl-omega-trinor PGE2. The results again indicate that EP3- rather than EP1- or EP2-receptors are involved. However, relative potentiating potency could be affected by differences in plasma protein binding and the very high sensitivity of the human platelet to prostacyclin (IP)-receptor-mediated inhibition (IC50 for the specific IP-receptor agonist cicaprost = 0.8 nM). 3. On human washed platelet suspensions the PGE analogues, with the exception of butaprost, inhibited the rise in adenosine 3':5'-cyclic monophosphate (cyclic AMP) induced by cicaprost (8 nM).PGE2 produced a monophasic inhibition curve (IC50 = 5.4 nM, 92% inhibition at 600 nM). The potency ranking was 16,16-dimethyl PGE2> sulprostone>MB 28767 = PGE2> misoprostol> GR 63778X>17-phenyl-w-trinor PGE2> 1 1-deoxy PGE2-1-alcohol. AH 6809 inhibited the effect of sulprostone and 17-phenyl-c-trinor PGE2 with pA2 values of 5.75 and 5.32 respectively; these values are at least one log unit lower than those found for EP1-receptor block in smooth muscle.4. There is a statistically significant correlation between IC50 values for the PGE analogues on the human platelet cyclic AMP assay and the guinea-pig vas deferens (standard EP3 preparation): slope =1.00, r = 0.80, P <0.05. However the correlation is far from ideal and GR 63779X in particular has a lower potency in the cyclic AMP assay. At this time we suggest that it is prudent to describe the human platelet receptor as 'EP3-like'.5. We believe that our results provide further evidence for linking PGE-induced potentiation of aggregation to inhibition of adenylate cyclase. Sulprostone is a suitable agonist for further study of this system and in particular the nature of the G-protein linkage(s) involved. In addition the necessity to consider potentiation of platelet aggregation in -relation to the clinical use of PGE analogues in man is emphasised.

Normal values for the hypercapnic ventilation response: effects of age and the ability to ventilate.

Normal values reported for the hypercapnic ventilation response (HCVR) vary considerably, but the reported normal values have come from studies containing small sample sizes and/or the subjects were young or of unidentified age. We speculated that age has a major effect on HCVR due to the generally lower maximal ventilation (VEmax) of elderly subjects. Therefore, we performed a large study to more accurately define the normal range and to reveal any effects which age and sex might have on HCVR. We studied 181 normal subjects (69 males, 112 females) between the ages of 20 and 93 years. Prior to measuring HCVR we measured forced expired volume in 1 s (FEV1) and forced vital capacity to establish whether lung function was normal and to obtain an estimate of VEmax. Results for the entire group revealed a significant correlation between FEV1 and HCVR [HCVR = 0.51 + (0.33 FEV1), r = 0.43, p < 0.001], so it is clear that the ability to ventilate can influence HCVR. We also found a significant correlation between age and HCVR [HCVR = 2.08-(0.01.age), r = 0.34, p < 0.001]. The mean HCVR for our male group (1.86 +/- 0.54 l/min.mm Hg) was significantly higher (p < 0.001) than that for the females (1.37 +/- 0.60) even though their mean ages were similar (41.3 vs. 41.7 years, respectively). We expect that this difference in HCVR was due to the higher FEV1 in males compared to females (3.82 vs. 2.83 liter, respectively).(ABSTRACT TRUNCATED AT 250 WORDS)

Gibberellic acid and abscisic acid coordinately regulate cytoplasmic calcium and secretory activity in barley aleurone protoplasts.

The effects of gibberellic acid (GA3) and abscisic acid (ABA) on the temporal and spatial dynamics of cytosolic calcium concentration ([Ca2+]i) in aleurone protoplasts of barley (Hordeum vulgare L. cv. Himalaya) were measured by using fluorescence ratio analysis and confocal microscopy. After 4-6 h of treatment, GA3 induced a sustained increase in [Ca2+]i from 50 to 150 nM in aleurone protoplasts. The increase in [Ca2+]i preceded the GA3-induced increase in alpha-amylase synthesis and secretion by 4 h. The elevation of [Ca2+]i was highest in the peripheral cytoplasm and may play a role in coordinating the secretory events there. Reducing Ca2+ levels in the incubation medium to below 500 microM inhibited the increase in [Ca2+]i and the GA3-induced stimulation of alpha-amylase synthesis and secretion. These data suggest that GA3 may increase [Ca2+]i by increasing the influx of Ca2+ at the plasma membrane. ABA reversed the effect of GA3 on [Ca2+]i within 3 h, 2 h before its effect on alpha-amylase production could be detected. Thus changes in [Ca2+]i may play a role in mediating the effects of GA3 and ABA on the synthesis and secretion of alpha-amylase in the aleurone cell.

Overall and regional lung function in Andean natives after descent to low altitude.

Overall lung volumes, regional residual volume to total lung capacity ratio (RVr/TLCr), regional ventilation (V/V) and perfusion (Q/V) were measured at 670 m in six Quechua Indians on days 2 and 37 after leaving their high-altitude homes (3500-4500 m). On day 2 the lung volumes averaged between 124 and 137% of those predicted for low-altitude residents (LAR) and there were no significant changes on day 37. Although overall RV/TLC was not different from the predicted value for LAR, RVr/TLCr on day 2 was higher at the top and lower at the bottom of the lungs compared to LAR. Regional Q/V and V/V were not different from LAR on day 2, or on day 37. However, the ratio of Q/V at the bottom to Q/V at the top was 2.36 on day 2 and 2.84 on day 37 (P less than 0.05). On day 2 hemoglobin- and volume-corrected diffusing capacity was 145% of the value predicted for LAR and this fell to 135% predicted on day 37 (P less than 0.05). Natives of high altitude reportedly have more alveoli that LAR and this could explain the greater vital capacity toward the bottom of the lung if the alveolar proliferation is concentrated there. This might also lower pulmonary vascular resistance at the bottom which would explain the normal Q/V distribution, even though pulmonary artery pressure may be increased.

Detection of vincristine-induced hyperploidy in meiotic II metaphases of male Chinese hamsters.

Induction of hyperploidy in germ cells of male Chinese hamsters treated with vincristine at dose levels of 0.25, 0.50 or 0.75 mg/kg of body weight was investigated. Animals were killed at 6, 24, 48, 72 and 96 h after administration of the chemical by a single intraperitoneal injection. The testes were removed and processed for spermatogonial, meiotic I, and meiotic II metaphases. Significantly increased frequencies of hyperploidy were obtained in meiotic II cells harvested 6, 24 and 48 h but not 72 and 96 h after treatment, indicating the importance of multiple sampling times. Analysis of spermatogonial cells shows that the frequencies of hyperploidy in the treated samples were comparable to those of controls. Limited sampling times used in the present study as well as small sample size or possible loss of hyperploid cells may be responsible for the negative findings for spermatogonial cells. Examination of meiotic I cells from 53 animals reveals the presence of one animal with an elevated level of hyperploidy unrelated to the vincristine treatment.

alpha-Amylase Isoforms are Posttranslationally Modified in the Endomembrane System of the Barley Aleurone Layer.

The subcellular site of the posttranslational modification of alpha-amylase was investigated in aleurone layers of barley (Hordeum vulgare L. cv Himalaya). Aleurone layers of Himalaya barley synthesize and secrete two groups of alpha-amylase isoforms, referred to as low-isoelectric point (low-pl) or HAMY1 and high-pl or HAMY2, when incubated in gibberellic acid and CaCl(2). Whereas homogenates of aleurone layers contain four isoforms of HAMY1 with pls 4.90, 4.72, 4.64, and 4.56, incubation media contain predominantly isoforms 4.72 and 4.56. Microsomal membranes isolated from aleurone layers contain all four isoforms of HAMY1. Microsomal membranes can be resolved into two peaks by isopycnic density gradient centrifugation: a peak of heavy membranes with endoplasmic reticulum and Golgi apparatus (GApp) marker enzyme activities and a peak of light membranes with characteristics of the GApp. The heavy membranes contain proportionally more HAMY1 pl 4.90 and 4.64 isoforms, whereas light membranes contain a higher proportion of pl 4.72 and 4.56 isoforms. Experiments with the ionophore monensin show that membranes of the GApp as well as the endoplasmic reticulum are involved in the posttranslational modification of HAMY1 isoforms. Monensin inhibits the secretion of alpha-amylase and causes the enzyme to accumulate within the cell. Precursor forms of HAMY1 accumulate in light membranes isolated from monensin-treated aleurone layers indicating that the GApp is involved in the conversion of the precursor to the secreted forms of the enzyme.

Cell Wall Free Space of Cucumis Hypocotyls Contains NAD and a Blue Light-Regulated Peroxidase Activity.

Solutions were obtained from the cell wall free space of red light-grown cucumber (Cucumis sativus L.) hypocotyl sections by a low-speed centrifugation technique. The centrifugate contained NAD and peroxidase but no detectable cytoplasmic contamination, as indicated by the absence of the activity of glucose-6-phosphate dehydrogenase from the cell wall solution. Peroxidase activity centrifuged from the cell wall of red light-grown cucumber hypocotyl section could be resolved into at least three cathodic isoforms and two anodic isoforms by isoelectric focusing. Treatment of red light-grown cucumber seedlings with a 10-minute pulse of high-intensity blue light increased the level of cell wall peroxidase by about 60% and caused a qualitative change in the anodic isoforms of this enzyme. The increase in peroxidase activity was detectable within 25 minutes after the start of the blue light pulse, was maximal at 35 minutes, and declined to control levels by 45 minutes of irradiation. The inhibitory effect of blue light on hypocotyl elongation was more rapid than the effect of blue light on total wall peroxidase activity, leading to the conclusion that growth and peroxidase activity are not causally related.

In vitro processing of tomato proteinase inhibitor I by barley microsomal membranes: a system for analysis of cotranslational processing of plant endomembrane proteins.

A plant-derived in vitro system for the study of cotranslational processing of plant endomembrane proteins has been developed and used to investigate cotranslational proteolytic processing of tomato proteinase inhibitor I. Translation of the inhibitor I precursor in wheat germ lysate supplemented with barley aleurone microsomal membranes resulted in cotranslational import of the protein into microsomal vesicles and cleavage of the signal sequence. NH(2)-terminal sequence analysis of the translocated inhibitor I processing intermediate showed that the signal sequence was cleaved between Ala(23) and Arg(24) of the precursor protein. Parallel experiments using dog pancreas microsomal membranes indicated an identical site of cleavage, suggesting that the substrate determinants for signal sequence processing are conserved across kingdoms. The plant-derived processing system used for this study may be valuable for analysis of cotranslational processing of other plant preproteins and for characterizing the components of the cotranslational import machinery in plants.

Adaptation to dim-red light leads to a nongradient pattern of stem elongation in cucumis seedlings.

Relative growth rate determinations on 5-millimeter regions of cucumber (Cucumis sativus L.) hypocotyls show that dim-red light-grown seedlings have an even distribution of growth along the stem axis. This contrasts with the apical to basal graded decline in growth rate seen in dark-grown seedlings, including dark-grown cucumber seedlings used as controls in this study. Dark-grown seedlings convert to the nongradient pattern when transferred to dim-red light. The small amount of light required suggests that the change in developmental pattern may happen in the natural light environment.
