In vivo response of mast cells to lepromatous serum.

Adult healthly mice of pure line Institute strain were given, into the foot pad, an intradermal injection of normal human serum in group A and lepromatous serum in group B. In both groups, the animals were killed at varying time intervals. Serial paraffin sections of the foot pad skin 5--7 mu thick, were cut and stained with haematoxylin and eosin, toludine blue, acridine orange and para-bromaniline. On comparing the two groups, in group B the response of mast cells was more prompt, pronounced and persistant. The increase in the number of cells was accompanied by explosive degranulation around nerves, blood vessels and muscles. This response became less by 15 days but did not disappear completely. There were still few intact cells in the dermis. Histamine, heparin and 5-Ht showed a corresponding rise. The importance of the findings is discussed.

Circulating immune complexes in carcinoma of uterine cervix.

Circulating immune complexes were determined in the sera of forty-eight patients wital status by inhibition of rheumatoid factor (RF) maediated agglutination of IgG coated latex particles. The inhibitory activity was present in 63% of the sera from cervical cancer patients but only 13% of sera from control women contained this activity. Similarly, anticomplementary activity suggestive of the presence of immune complexes was present in 71% of sera from cancer patients and 13% of sera from control women. The incidence as well as the levels of immune complexes (IC) were higher in sera of patients with advanced stages of cancer as compared to those with Stage I cervical cancer. In addition, 95% of sera from cervical cancer patients had cryoblobulins, whereas only 20.8% sera from matched controls were positive for cryoprecipitates. All the cryoglobulins contained IgG type of immunoglobulin except one which had IgA immunoglobulins also. The complexes have not been characterized with regard to the nature of the antigen(s) present.

Rat intestinal 25-hydroxyvitamin D3- and 1alpha,25-dihydroxyvitamin D3-24-hydroxylase.

The 24-hydroxylation of 25-hydroxyvitamin D3 and 1alpha,25-dihydroxyvitamin D3 occurs in vitro in rat intestinal homogenates. 1alpha,24,25-Trihydroxyvitamin D3 obtained by incubating 1alpha,25-dihydroxyvitamin D3 with rat intestinal homogenates was isolated and then identified by co-chromatography of the biologically generated material with synthetic 1alpha,24R,25-trihydroxyvitamin D3 on a high pressure liquid chromatography system and by ultraviolet spectroscopy, periodate cleavage, and mass spectroscopy. 24,25-Dihydroxyvitamin D3 obtained by incubating 25-hydroxyvitamin D3 with intestinal homogenates was identified by co-chromatography of the biologically generated material with authentic 24R,25-dihydroxyvitamin D3 on a high pressure liquid chromatography system and by periodate cleavage. Tissue distribution studies indicate that the process occurs in rat intestinal homogenates but not in rat muscle, liver, or fetal calvaria homogenates. Nephrectomy does not diminish the production of 1alpha,24,25-trihydroxyvitamin D3 in vivo while it reduces markedly, but does not eliminate, production of 24,25-dihydroxyvitamin D3.

Previous induced abortion and ante-natal depression in primiparae: preliminary report of a survey of mental health in pregnancy.

One hundred and nineteen primiparae, who were routinely attending ante-natal clinics, were interviewed repeatedly between the 12th and 36th weeks of their pregnancies. The incidence of depression was highest in the first trimester and, overall, about a fifth of the sample was found to be suffering from clinically significant neurotic disturbances. In a proportion of these expectant mothers there was an association between depression and anxiety early in pregnancy and a previous history of induced abortion; this phenomenon may reflect a reactivation of mourning which was previously suppressed.

Clinical trial of a new bleeding-time device.

The authors report a clinical trial comparing a new bleeding time device (Simplate II) with the Mielke Template. Bleeding times were determined at the same time on the same arm using the two devices. Subjects of the study were 24 healthy volunteers, before and two hours after ingestion of 975 mg aspirin, and 28 patients. For the normal subjects the mean pre-aspirin bleeding times were 4.75 +/- 1.42 minutes (1 SD) with the Simplate II and 3.65 +/- 1.22 minutes with the Mielke Template. The mean bleeding times two hours after ingestion of aspirin were 7.86 +/- 2.76 minutes with the Simplate II and 7.84 +/- 2.94 minutes with the Template. The pre- and the post-aspirin values with the two devices were not significantly different from each other, nor were the bleeding times obtained in the patients with the two devices. The extents of scarring were similar with the two devices. The results were highly reproducible by both methods. The new device was simpler and more rapid to use.

Gastric ulceration and the concentration of salicylate in plasma in rats after administration of 14C-labelled aspirin and its synthetic triglyceride, 1,3-dipalmitoyl-2(2'-acetoxy-[14C]carboxylbenzoyl) glycerol.

Triglycerides containing aspirin in place of one or more fatty acid residues of the molecule have been synthesized. Metabolism of the compound with the labelled (14C) drug residue introduced specifically into the 2-position of the triglyceride is reported. Plasma salicylate concentrations with this synthetic glyceride were determined and compared with those obtained with commercially available aspirin labelled with the 14C-isotope. Both compounds gave a therapeutic concentration of salicylate in the plasma after ingestion. The 1,3-di-fatty acyl-2-aspirin glyceride was absorbed through the intestine as 2-aspirin monoglyceride, some 20% of which was transported through the thoracic-duct chyle and about 30% through the portal system. Whereas pronounced ulceration of the rat stomach occurred with free aspirin, the above fatty acyl glyceride of aspirin produced no ulceration.

Human erythrocyte proteins associated with adenosine 3',5'-cyclic monophosphate action.

Two adenosine 3',5'-cyclic monophosphate (cyclic-AMP)-binding protein factors (molecular weight 230,000) have been partially purified from human erythrocytes. One of these proteins seems to be different from the cyclic-AMP-binding component of the cyclic-AMP-dependent protein kinases. These protein factors are also capable of binding adenosine. We present data also on two forms of cyclic-AMP-dependent protein kinases (ATP: protein phosphotransferase, EC 2.7.1.37) partially purified from the cytosol of normal human erythrocytes. Kinase I has been classified as type I enzyme on the basis of its activation when preincubated with protamine, histone or NaCl. The substrate specificities of the two kinases and many of their kinetic parameters are rather similar. Their subunit structure is reminiscent of that of kinases obtained from other sources. The catalytic subunit of both enzymes reversibly cross-react with the regulatory subunit of kinase I from the rabbit red blood cell.
