The relation between potassium in body fluids and total body potassium in healthy and diabetic subjects.

1. The concentration of potassium in the erythrocytes and the plasma of forty-one normal subjects and twenty-five diabetic patients was measured and the results were used to calculate the total amount of potassium in the erythrocyte mass and the total amount of potassium in the plasma. The total body potassium was measured in a whole-body monitor. 2. In normal subjects a close correlation was found between total erythrocyte potassium and total body potassium and also between total plasma potassium and total body potassium. 3. The regression relation between total body potassium and total erythrocyte potassium in normal subjects was used to predict the total body potassium in diabetic patients. There was reasonable agreement between the measured and predicted total body potassium but there was poor agreement between the measured total body potassium and that predicted from the patient's height and age or height and age or height, weight and age.

Oxygen therapy in chronic respiratory failure.

In hypercapnic chronic respiratory failure red cell volume correlates with the arterial oxygen tension during the quiescent phase of the disease. During an acute exacerbation measurement of the red cell volume can give a reasonable index of the arterial oxygen tension existing in the previous quiescent state. This level of arterial oxygen tension appears to determine the tolerance of the individual to treatment with oxygen.

The relation of total body potassium to height, weight, and age in normal adults.

Total body potassium was measured in 103 healthy adults using a shadow-shield whole-body monitor of high sensitivity. The range of height was 147 to 192 cm, of weight 43 to 92 kg, and of age 18 to 77 years. The values obtained for total body potassium were correlated with height, with weight, and with height and weight. Age was then included as an additional variable. The standard deviation from regression was smaller when total body potassium was correlated with height than with weight and was further reduced, to about 9%, in a multiple regression using height and age. The advantages of this relationship over indices involving weight are discussed. The smallest standard deviation from regression, 7.5%, was obtained when total body potassium was correlated with height, weight, and age. The usefulness of this relationship is discussed with comment on its limitations.A regression equation was derived between lean body mass (derived from height and weight) and total body potassium with a standard deviation from regression of 5.5% in males and 7.3% in females.

Relationship between total body water and surface area in normal and obese subjects.

Total body water was measured using tritium in 30 males and 30 females. It was found that total body water could be predicted from height and weight, and formulae for both males and females have been produced with multiple correlation coefficients (r) of 0.95 and 0.96 respectively. The predicted total body water was found to be very closely related to the predicted surface area giving correlation coefficients (r) for males and females of 0.997 and 0.985 respectively.

Separation of two forms of cytochrome P450 adrenal cortex mitochondria.

The separation of two distinct forms of cytochrome P450 from adrenal cortex mitochondria has been achieved by the following steps; (1) lyophilisation (2) iso-octane extraction, (3) (NH(4))(2)SO(4) fractionation in the presence of sodium cholate. The fraction precipitating between 25-35 percent (NH(4))(2)SO(4) gave a difference spectrum with 11-deoxycorticosterone (11-DOC) but not with 20alpha-hydroxycholesterol (20alpha-HOC). This fraction showed high 11beta-hydroxylase activity but low activity for side chain cleavage of cholesterol (S.C.C.). The fraction precipitating between 45-60 percent (NH(4))(2)SO(4) gave a difference spectrum with 20alpha-HOC but not with 11-DOC and exhibited high S.C.C. activity but low 11beta-hydroxylase activity. The absorption spectrum of the 45-60 percent fraction indicated a preponderance of high spin hemoprotein (lambda(max) 395 nm).

Red cell survival in biliary cirrhosis.

Studies were carried out on eight patients with primary biliary cirrhosis. Four patients were found to have a haemolytic anaemia; one had Coombs red cell antibodies. Two patients had evidence of splenic sequestration of red cells using the radio-chromium technique. There was a significant correlation between the red cell survival and the bromsulphthalein retention test and the red cell survival and the serum level of bilirubin. It was concluded that the anaemia of cirrhosis of the liver, including primary biliary cirrhosis, may be due to a number of mechanisms and a unifying hypothesis based on the degree of liver dysfunction is suggested.
