Oxytocin levels and disappearance rate and plasma follicle-stimulating hormone and luteinizing hormone after oxytocin infusion in men.

Plasma oxytocin (OT), FSH, and LH were measured by specific RIA in eight healthy adult males before, during, and after stopping iv infusions of OT. With a constant infusion of 132 mU OT/min for 60 min, plasma OT reached a steady state concentration of 228-241 pg/ml at 30-60 min. When the dose of oxytocin infused was doubled every 15 min, plasma OT increased from 81.0 +/- 17.9 pg/ml (mean +/- SE) with 32 mU/min to reach a steady state concentration of 378 +/- 73.4 pg/ml with 256 mU/min (1 muU = 2 pg OT). The curve of disappearance of plasma OT could be resolved into a single exponential curve in all of the subjects, with a mean calculated half-life of 10.3 +/- 1.6 min (range, 5.3-17.3 min). The mean MCR of OT was 21.5 +/- 3.3 ml/kg.min, and the mean apparent volume of distribution was 305 +/- 46 ml/kg. Plasma FSH and LH showed no significant change throughout OT infusion and for up to 60 min after stopping the OT infusion. The findings demonstrate that in man 1) plasma OT concentration achieved is closely related to the infusion rate, 2) OT infusion does not affect plasma FSH and LH, and 3) the apparent volume of distribution of OT suggests that infused OT is distributed into space or spaces other than the circulating plasma volume.

Effect of radiation on myocardial metabolism and structure.

The effects of gamma radiation on structure, function, and metabolism of rat heart were studied. The precordial region of male albino rats (weighing 150-200 g) was exposed to 4000, 6000, and 8000 rads of gamma rays, using a cesium-137. Seventy-two hours after exposure, ECG lead II was taken, and animals were killed. Blood, heart, and adrenals were collected for the study of serum lactic dehydrogenase (SLD), serum glutamic oxaloacetic transaminase (SGOT), serum lactate, cardiac glycogen, and adrenal ascorbic acid. Histopathology of the heart was done using eosine and hematoxyline stains. The structural, functional, and biochemical changes observed were directly proportional to the intensity of gamma radiation.

Necrotizing sarcoid granulomatosis.

Twelve cases of necrotizing sarcoid granulomatosis are presented as a retrospective study. The population of patients consisted of ten women and two men, with an average age of 50 years. Nine patients initially had a variety of pulmonary and nonpulmonary complaints, while three were asymptomatic. Chest roentgenograms demonstrated bilateral nodules in seven patients, solitary nodules in four, and a miliary pattern progressing to nodules in one. Enlarged hilar nodes were found in six. Histologically, all biopsies showed a combination of individual granulomas, confluent masses of granulomas which formed the nodular masses seen on the roentgenograms, a variable amount of necrosis of tissue and hyalinization, and a granulomatous vasculitis. Follow-up periods ranged from four months to 11 years. Eleven patients are alive and asymptomatic. The sole death occurred in a patient treated with an immunosuppressive agent (cyclophosphamide). We conclude that the clinical behavior of necrotizing sarcoid granulomatosis is not similar to that of the other angiocentric granulomatoses and that most patients with this disease can be left untreated or be treated with steroids alone. We suggest the possibility that necrotizing sarcoid granulomatosis may be the histologic counterpart of so-called nodular sarcoid.

Fetal contribution to oxytocin in human labor.

Fetal oxytocin contribution to the mother during spontaneous labor was investigated using a specific and sensitive radioimmunoassay to measure oxytocin in blood, amniotic fluid, and urine. In 26 subjects with spontaneous labor and vaginal delivery (Group I) and 18 subjects with cesarean section after labor (Group II), umbilical arterial plasma (UA) oxytocin concentrations were significantly higher than umbilical venous plasma (UV) ocytocin concentrations. With elective cesarean section (Group III), UA oxytocin concentration was 29.8 +/- 7.5 pg/ml and UV oxytocin concentration was 16.1 +/- 5.9 pg/ml (n = 14). In contrast, the mean UV oxytocin concentration was higher than the mean UA oxytocin concentration, when oxytocin was given to the mothen concentration in Groups I and II was significantly higher than in Group III. Amniotic fluid oxytocin concentrations in Group I and II patients were higher than in Group III. Oxytocin was also present in fetal urine. The findings indicate that during spontaneous labor, oxytocin is produced by the fetus and flows toward the maternal circulation.

Antidepressant evaluation and the pharmacological actions of FG4963 in depressive patients.

FG4963 a phenylpiperidine derivative and potent 5-HT reuptake inhibitor at neuronal sites was investigated in a group of 10 depressive patients, and its effect was compared with that of amitryptyline in a group of 10 depressive patients treated with amitriptyline. FG4963 was found to be significantly inferior as an antidepressant compared to amitryptyline over a 6-week period. FG4963 did not appear to be anticholinergic as judged by its lack of effect on salivary flow. Its effect (in the dosage used) on the tyramine dose--pressor response and NA dose--pressor response tests were less than those of amitryptyline.
