Pathogenesis of Staphylococcus aureus mastitis: bacteriologic, histologic, and ultrastructural pathologic findings.

Three lactating cows were experimentally infected with Staphylococcus aureus ATCC 29740 (Newbould 305 strain). Cows were euthanatized 2 to 216 hours after inoculation. Bacteriologic and microscopic examinations showed that S aureus attached to epithelial cells of the mammary gland in vivo. The histopathologic changes observed were progressive swelling, vacuolar degeneration of epithelial layers, and multiple foci of epithelial erosions and ulcers throughout the ductal system. The cellular response of the infected glands was demonstrated by a rapid increase in the number of somatic cells in the secretion and by accumulation of neutrophils below, within, and on the epithelium of the teat and lactiferous sinuses. The inflammatory response did not prevent infection nor subsequent pathologic changes in the inoculated glands.

Differentiation of staphylococci on the basis of nuclease properties.

The quantity, thermostability, and serological pattern of nucleases produced by different staphylococci were studied. Staphylococcal strains were isolated from nine different species of animals or from humans. Staphylococcus aureus, Staphylococcus intermedius, and Staphylococcus hyicus subsp. hyicus were vigorous producers of nuclease, whereas the coagulase-negative staphylococci, except S. hyicus subsp. hyicus, produced significantly less nuclease. The nucleases of all strains were found to be thermostable. S. aureus, S. intermedius, and S. hyicus subsp. hyicus could be distinguished from each other and from coagulase-negative staphylococci on the basis of inhibition of nuclease activity by specific antibodies.

The comparative effect of two teat dips on udder health and teat skin quality.

The effect of two teat dips on udder health and teat skin appearance was examined. Ewodip N contained urea and a methylglycoside with a final iodine concentration of 0.25%. Ewodip with lanolin as emolient had a final iodine concentration of 0.4%. Ewodip N had a more favourable effect on teat skin quality than Ewodip. The percentage of cows with elevated cell count and without udder pathogenic microorganism (non-specific mastitis), was significantly lower in the group dipped with Ewodip N. The level of new infections of Staphylococcus aureus was lowest in the Ewodip group, but the difference was not statistically significant.

In vitro antibiotic sensitivity test of Staphylococcus aureus isolated from mastitic milk.

The antibiotic susceptibility of 2297 strains of Staphylococci aureus isolated from mastitic milk has been tested. The percentage of strains of S. aureus "resistant" to penicillin was 16.5%. Approximately 97% of these strains were susceptible both to tetracyclines and sulfonamides. The frequency of "resistant" strains is in good agreement with the results from a comparable survey done ten years ago. In this investigation, however, the strains of S. aureus showed great variation in their sensitivity to penicillin according to their geographical origin. Thus, the percentage "resistant" strains within counties varied from a minimum of 9.26 to a maximum of 53.05.
