Lung water and vascular permeability-surface area in newborn lambs delivered by Caesarean section compared with the 3-5 days-old lamb and adult sheep.
Extravascular lung water and vascular permeability-surface area products were measured with a multiple indicator dilution method in five unanaesthetized newborn lambs 2 to 7 h following delivery by Caesarean section at a gestational age of 135-142 days. The indicators used were 51Cr-labelled erythrocytes, [125I] albumin, [3H] water and [14C] urea. Post-mortem determination of extravascular lung water and dry bloodless lung weight were also performed. The results were compared with previously obtained data in unanaesthetized 3-5 days-old lambs and adult sheep. Normalized to dry lung weight, extravascular lung water by both techniques was significantly higher in lambs after Caesarean section than in either the 3-5 days-old lambs or the adult sheep (P less than 0.05). (Indicator values = 3.9 +/- 0.2 (SEM) ml/g for Caesarean section lambs, 3.1 +/- 0.3 for 3-5 days-old lambs and 3.3 +/- 0.5 for adult sheep; post-mortem values were 6.12 +/- 0.28 g/g for after Caesarean section lambs, 4.07 +/- 0.26 for 3-5 days old lambs and 4.03 +/- 0.17 for sheep). Vascular permeability-surface area product for [14C] urea, computed by a mathematical model, was significantly higher (P less than 0.05) in lambs after Caesarean section than in adult sheep (0.18 +/- 0.02 compared with 0.10 +/- 0.03). In 3-5 days-old lambs the value was 0.16 +/- 0.04. It is concluded that, when normalized to dry lung weight, lung following Caesarean section delivery near the end of gestation. Adult values for lung water have been reached by the age of 3-5 days.

Computerised image analysis of split-drop micropuncture data.
A technique is described for rapid and reproducible analysis of split-drop micropuncture sequences recorded on film. Automatic identification of the droplet menisci in each cine frame, and subsequent evaluation is performed by a computerised T.V. image analysis system. In comparison with a more conventional technique, the improvement in reproducibility of the analysis is achieved without loss of accuracy and is accompanied by a three fold increase in speed.

Pulmonary vascular response to prostacyclin in fetal lambs.
Eighteen prostacyclin injections (19.4 +/- 1.5 micrograms/kg) were performed in five chronically instrumented, intact fetal lambs in order to study the effects on pulmonary blood flow. These resulted in a brief period of bradycardia followed by a more prolonged period of increased pulmonary blood flow. In this latter phase, pulmonary blood flow increased from a baseline value of 49 +/- 4 ml/(kg min) to 122 +/- 10 ml/(kg min). Systolic/diastolic pulmonary arterial pressure simultaneously fell from 73 +/- 2/48 +/- 1 to 68 +/- 2/42 +/- 1 mm Hg. Flow through the ductus arteriosus was unchanged and right ventricular output increased to account for the increased pulmonary blood flow. Thus, prostacyclin causes pulmonary vasodilation in intact fetal lambs and may participate in the control of fetal pulmonary blood flow and the circulatory adjustments to extra-uterine life.
Treatment of Peyronie's disease by dermal grafting.
We describe our experience with dermal grafting for the treatment of Peyronie's disease in 27 patients over a 3-year period. With the various nonsurgical treatments, a significant number of patients remain handicapped by the disease. For this unresponsive group, we recommend surgical excision of the penile plaque with dermal grafting. The proper selection of patients for this operation is of paramount importance, and this can be facilitated by a preoperative psychological evaluation in doubtful cases.
A new gene of Escherichia coli K-12 whose product participates in T4 bacteriophage late gene expression: interaction of lit with the T4-induced polynucleotide 5'-kinase 3'-phosphatase.
We isolated five Escherichia coli mutants deficient in their ability to support the late (replication-coupled) gene expression of T4 bacteriophage at 30 degrees C. These mutants, which we call Lit mutants, define at least one novel gene at 25 min on the E. coli map. They were selected in an attempt to obtain mutants which restrict the growth of T4 mutants deficient in polynucleotide 5'-kinase 3'-phosphatase but not that of wild-type T4 at 37 degrees C. Some of the mutants do have these phenotypes under some conditions. Studies of the block in T4 development in some of the E. coli mutants suggest that Lit mutants are affected in a gene product involved in the metabolism of deoxyribonucleic acid nicks or single-strand gaps. None of the Lit mutants is deficient in the major, bacterial, 3'-phosphatase activity in crude extracts.
Ionic requirements of proximal tubular sodium transport. III. Selective luminal anion substitution.
The effect of substitution of luminal anions on sodium and fluid absorption in rat renal proximal convoluted tubules was studied with continuous luminal microperfusion methods. Substitution of bicarbonate in the control Ringer perfusion fluid by 25 mM acetate reduced net sodium reabsorption by 40%; substitution by chloride reduced it by 25%; and substitution by cyclamate reduced it by 70%. Infusion of acetazolamide reduced net sodium and fluid transport in all cases except chloride-Ringer perfusion. Cyanide added to the perfusion fluid inhibited fluid and sodium movement completely when there was no imposed chloride concentration gradient, but only reduced fluid and solute movement by 68% when a nominal 36 mM transepithelial chloride concentration gradient existed. We conclude from these observations that passive forces for sodium reabsorption can account for a moderate amount of sodium transport, that the effects of acetazolamide in low concentrations are dependent on the presence of bicarbonate in the lumen, and that some anions alter net sodium transport either by reducing the availability of permeant anion for co-transport with sodium or by a direct effect on the sodium and/or chloride transport systems.
