Noradrenaline modulation of the responses of the cerebellar Purkinje cell to afferent synaptic activity.

Noradrenaline, applied by microiontophoresis to rat cerebellar Purkinje neurones, selectively depressed spontaneous neuronal discharge. Simple spike and complex spike excitations, evoked by stimulation of the mossy and climbing fibres, were relatively preserved during the inhibition of spontaneous activity, and the number of full-sized action potentials in the complex spike increased. Inhibition mediated by the basket and stellate cells was augmented. Thus, relative to the change in spontaneous activity, noradrenaline increased the responsiveness of the Purkinje cell to afferent input.

The record of family planning programs.

In the developing countries in recent years there has been a great expansion of public programs to provide modern means of fertility control. This paper is an effort to appraise the demographic impact of such programs through a comprehensive evaluation of the record. The paper briefly reviews the criticisms of such programs and the historical and comparative background before turning directly to issues of performance. After establishing the broad range of acceptance, the paper reviews the nature of acceptors and of fertility control methods as they affect overall impact. Then follow a series of tabulations and analyses of country performance by both social setting and program effort in an attempt to discern the effect of "modernization" and "family planning" in line with the current controversy over their relative importance. That detailed analysis is followed by an extensive review of 14 country cases, similarly organized, plus brief summaries of 15 major experiments/demonstrations in this field and one example of a developed country program. After a brief section on the alternatives to family planning programs, the paper concludes with a summary of major findings-underlined in the text-and the authors' personal commentary on their implications for both study and action.

Histochemical characterization of a neocortical projection of the nucleus locus coeruleus in the squirrel monkey.

Histochemical evidence is presented for a catecholamine-containing projection from the nucleus locus coeruleus to the neocortex in the squirrel monkey. The innervation of superior temporal gyrus has been examined in particular. Glyoxylic acid-induced fluorescence shows an extensive arborization of fine, catecholamine-containing fibers with prominent varicosities in all layers of the neocortex. The nucleus locus coeruleus is identified as a source of these fibers by both ortho- and retrograde axonal tracing techniques. After injection of horseradish peroxidase into the neocortex, labelled cell bodies are localized throughout the major portions of the locus coeruleus. Conversely, after microinjection into the nucleus locus coeruleus, tritiated proline is transported into the neocortex where it appears within fibers similar in distribution to those revealed by fluorescence histochemistry. Both transport techniques indicate that cortical projections of the locus coerculeus originate from both ipsilateral and contralateral nuclei.

Effects of putative neurotransmitters on neuronal activity in monkey auditory cortex.

The effects of the putative neurotransmitters norepinephrine (NE), gamma-aminobutyric acid (BAGA), and acetylcholine (ACh) were tested on auditory cortex neurons which were activated acoustically by species-specific vocalizations in awake squirrel monkeys. Five-barrel glass electrodes were used to record the activity single neurons in the superior temporal gyrus and to apply NE, GABA, or ACh microiontophoretically. Poststimulus time histograms and raster displays of neuronal responses to the vocalizations were computed before, during, and after iontophoresis. Dose-dependent inhibition of spontaneous and vocalization-evoked discharge rates was seen with NE and GABA. Generally, excitation was observed with ACh. A given dose of NE or GABA reduced spontaneous activity by a greater proportion than it reduced activity evoked by the vocalizations. During excitatory responses, segments with lower discharge rates were reduced proportionately more than segments with higher discharge rates. Usually, response 'pattern' was not altered by iontophoresis of any of the substances. However, in some cases the differential inhibition of slow activity produced by NE or GABA did result in a 'patern' change. The demonstration that small amounts of locally applied NE and GABA substantially alter the specific neuronal activation produced by vocalizations provides additional evidence that these agents may function as neurotransmitters in this neocortical area and offers clues about their functional significance.

Phenothiazine antagonism of the noradrenergic inhibition of cerebellar Purkinje neurons.

Phenothiazine derivatives were examined as potential antagonists of the inhibitory noradrenergic synapses from the nucleus locus coeruleus to rat cerebellar Purkinje cells. Fluphenazine, and its thioxanthine analogue, flupenthixol, antagonized the inhibitory action of norepinephrine, when iontrophoretically applied to single cells. Alpha-flupenthixol was generally more active than the beta isomer. Fluphenazine had no appreciable effect on inhibitions induced by iontophoresis of GABA or cyclic AMP. Parenteral fluphenazine also blocked the inhibition of Purkinje cells produced by the stimulation of the noradrenergic pathway from locus coeruleus, but basket and stellate cell inhibitory inputs to Purkinje cells were unaffected. These data suggest that fluphenazine can specifically block a known central adrenergic inhibitory pathway.

Substituted 1-[(5-nitrofurfurylidene)amino]-4-imidazolin-2-ones.

A series of 1-[(5-nitrofurfurylidene)amino]-4-imidazolin-2-ones has been prepared. A new synthesis of 4-alkyl-1-[(5-nitrofurfurylidene)amino]-4-imidazolin-2-ones involving the oxidative ring closure of 5-nitro-2-furaldehyde 2-(2-hydroxyethylalkyl)semicarbazones is described. The in vitro testing of the compounds against a variety of bacteria is reported.

A quantitative microiontophoretic analysis of the responses of central neurones to noradrenaline: interactions with cobalt, manganese, verapamil and dichloroisoprenaline.

1. A new experimental procedure has been devised for the study of pharmacological antagonism in the central nervous sytem using automated microiontophoresis to deliver pulses of agonists and computer-generated histograms to quantify the neuronal response. The system allows study of potential antagonists having direct depressant effects and also of neurones with irregular or slow discharge rates. 2. The histogram analysis reveals the necessity for regular, periodic delivery of agonists during the assessment of agonist-antagonist interactions. Without regular repetitive delivery, many agonists, such as noradrenaline, exhibit an apparent but artifactual decrease in inhibitory potency after an interruption of agonist pulses. Examples of this phenomenon are shown, using cerebellar Punkinje cells and cerebral cortical neurones in rats anaesthetized with halothane. 3. Preliminary results with these computer-generated drug response histograms revealed manganese, cobalt, and verapamil to be generally ineffective as antagonists of noradrenaline, despite their direct depressant effects. 4. Conversely, dichloroisoprenaline (DCI), a beta-adrenoceptor antagonist, was effective in blocking noradrenaline-induced depressions of firing in the cerebral cortex at doses which caused over 50% decrease in spontaneous discharge.

Do statements about desired family size predict fertility? The case of Taiwan, 1967-1970.

The predictive accuracy of respondents' statements about their future fertility is examined, using interview data from a longitudinal study conducted in Taiwan. Two measures of preference are found to be highly intercorrelated; and regardless of which one is used, Taiwanese women are shown to predict their subsequent fertility at least as well as U.S. women. The preference measures are also predictive of rates of contraceptive use and abortion. While demographic and social characteristics are correlated with fertility in expected directions, statements about wanting more children prove to be highly predictive of subsequent fertility for both modern and less advanced segments of the population.

Specificity of fixation loci for homocytotropic antibodies.

The heat-stable, homocytotropic antibody synthesized by guinea pigs to highly purified dextran has been studied by passive cutaneous anaphylaxis (PCA). Antibody arising in single guinea pigs was found to give PCA in some homologous recipients but not in others. Data are presented which indicate that the antibodies are bound in the skin of some animals to a greater degree than in that of others. This suggests some type of variability, perhaps allotypic, in the Fc portion of these immunoglobulins which relates to specificities of the loci where Fc becomes fixed in skin.
