In chick embryo fibroblasts treated in vitro with adriamycin, the mitotic activity is strongly depressed, the nucleoli are altered, nucleolar RNA synthesis is inhibited and the RNA content is lowered. These effects can be concomitantly and spontaneously reversible, but the mitotic activity does not resume.

Chick embryo fibroblasts cultivated in vitro were treated with ethidium bromide (E.B.) or with DNA-E.B. complex (DNA-E.B.). E.B. (5 mug/ml) provokes morphological alterations and cell death, inhibits DNA synthesis and mitotic activity. DNA-E.B. (E.B. 5 mug/ml) is less toxic to the fibroblasts as far as cell structure, DNA synthesis and mitotic activity are concerned. DNA alone has no apparent effect on the fibroblasts. As shown by fluorescence microscopy, the lower toxicity of DNA-E.B. seems to be related to its mode of penetration into the cells.
Chick embryo fibroblasts cultivated in vitro and treated with cis-dichlorodiamino platinum (II) have been analysed by cytological and cytochemical methods. Morphological alterations (in the nucleoli, in the chromatin and in the cytoplasm), inhibition of DNA and RNA syntheses and of cell multiplication have been noted. Under some conditions, cells are blocked just before entering into mitosis (G2 block) and can become polyploid. A high degree of cell degeneration has also been observed.
The RNA content was measured in individual mouse Ehrlich ascites tumour cells by absorption cytophotometry after gallocyanin chrome alum staining. The cellular RNA content doubles during the preparation for mitosis and is twice as high as in an hyperdiploid line in an hypertetraploid cell lines.

Chick embryo fibroblasts cultivated in vitro and Ehrlich ascites tumor cells (in vivo or in vitro) have been treated with amphotericin B. Cell multiplication is strongly inhibited. Large clear zones appear in the fibroblast nucleoi (phase-contrast and electron-microscope observations). Many treated fibroblasts and tumour cells have a high DNA content (pre-mitotic or polyploid level; measurements by cytophotometry). However, the RNA content (cytophotometry) and the total protein content (cytophotometry and micro-interferometry) are relatively low in the tumour cells. As shown by autoradiography, DNA synthesis is active but RNA synthesis and, in some cases, protein synthesis are inhibited. Due to this unbalanced growth, the cells cannot divide.
Cordycepin strongly inhibits cell multiplication in chick fibroblasts cultivated in vitro. Many cells cannot enter into mitosis and cell separation at telophase is disturbed. However, DNA synthesis takes place. Nucleolar alterations observed in the treated cells are probably responsible for some of these effects.
