Neuronal and glial differentiation in reaggregation cultures.

Dissociation and reaggregation cultures from different portions of the chick embryo neural tube were made, and the resulting aggregates were fixed for electron microscopy after 1, 5, 8, 14, 16 and 22 days in vitro. All cultures (pure aggregates of telencephalon, optic lobe or neural retina, and combined aggregates made from mixtures of optic lobe plus neural retina or optic lobe plus telencephalon) show a common timing of neuronal and glial morphological differentiation. During the first week in vitro, some cells developed neuronal characteristics in the absence of morphological evidence of glial differentiation. Numerous axonic processes usually formed fascicles with all the fibers running parallel to each other. Axonic growth cones were abundant and a few immature synapses were also persent. The second week in culture was characterized by the disappearance of growth cones and the increase in number and morphological maturation of synapses. Morphologically detectable glial differentiation began by the end of this week, and during the third week almost every neuronal element, including the axonic fascicles, became associated with glial cells showing astrocytic features.

Fulminant hepatitis. A presentation of Wilson's disease.

A boy, 10 years of age, was admitted to the hospital with rapid onset of hepatic failure and died within three weeks. Laboratory and pathological data were consistent with Wilson's disease. We discuss the importance of Wilson's disease in the differential diagnosis of acute liver failure.

Rhabdomyolysis with severe hypernatremia.

Three children with severe hypernatremia presented with profound generalized weakness and biochemical evidence of rhabdomyolysis and myoglobinuria. These findings in combination have not been previously reported, to our knowledge, in children with severe hypernatremia. Unusual complications included respiratory failure in one child and cardiac arrhythmias in two children. All three children had acute renal insufficiency; one required peritoneal dialysis.

Analysis of the RNA species isolated from defective particles of vesicular stomatitis virus.

Serial high multiplicity passage of a cloned stock of vesicular stomatitis virus was found to generate defective interfering particles containing three size classes of RNA, with sedimentaiton coefficients of 31 S, 23 S and 19 S. The 31 S and 23 S RNA species were found to be complementary to both the 12 to 18 S and 31 S size classes of VSV mRNAs. The 19 S class of RNA was found to be partially base-paired. All three RNA species were found to contain ppAp at their 5' termini.

Practical problems in the treatment of pain.

According to modern pain concepts, "pain" is the result of multifactorial influences. Planning of therapy therefore requires the diagnostic evaluation of a number of factors including delineation of an organic lesion, evaluation of the influence of pain on the individual's psychological development, situational anxiety, psychological coping style etc. The understanding and management of psychological factors is crucial in every pain syndrome as the idea the pain is either psychogenic or organic is outdated. For the treatment of mild pain salicylic acid is still unsurpassed. The rationale of its combination with a tranquillizer, and with codeine or benzomorphane for moderately severe pain, can be understood on the basis of modern pain concepts. Before strong narcotics are used a trial with combination of a phenothiazine with a tricyclic antidepressant drug is warranted. Methadone seems to have advantages over morphine for very distressing pain treated with narcotics. Biofeedback for pain due to muscle spasms, electrical stimulation of the dorsal column for intense chronic pain due to a well-delineated organic lesion, and operant conditioning applied by a well-trained team of physicians and staff are promising new treatments for pain. The place of acupuncture in future pain therapy cannot yet be judged. It must first be freed of cult-type opinions and evaluated further by studies meeting modern western standards of pain research.

Methylated and blocked 5' termini in vesicular stomatitis virus in vivo mRNAs.

Methyl groups derived from 3H-methyl methionine were incorporated into vesicular stomatitis virus (VSV) MRNAs isolated from infected cells. Sequential degradation of the 12-18S viral mRNA species with ribonuclease T2, penicillium nuclease, and alkaline phosphatase yielded a single 3H-labeled dinucleotide. A similar resistant 32P-labeled fragment was obtained by digesting VSV mRNA uniformly labeled with 32P. This methylated and blocked oligomer was further cleaved with nucleotide pyrophosphatase, yielding two methylated 5' nucleotides. We postulate that the 5' terminal structure of the vivo 12-18S VSV mRNA contains 7-methylguanosine linked by a 5'-5' pyrophosphate bond to a methylated derivative of adenosine. In contrast to the mRNAs (+ strand), the VSV genome RNA ( MINUS STRAND) IS NOT BLOCKED.

Transformation of cells by herpes simplex virus--fact or fantasy?

The replication of herpes simplex virus types 1 and 2 is defective in XC cells. One lesion is the failure of viral proteins to be transported to the cell nucleus. The resistance of XC cells to the toxic effects of herpes simplex virus is apparently not due directly to the presence of RNA tumor virus in these cells. Other cell lines transformed by RNA tumor viruses exhibited no comparable resistance. The nature of the association between HSV and cell for a number of generations after first selecting "transformants" is not yet established; on prolonged subculture, much of the original HSV information fails to be expressed. Not more than approximately to 0.1 viral genomes per cell remains permanently associated with the host DNA. The "transformants" retain the capacity to synthesize HSV-specific polypeptides, as determined by radioimmunoprecipitation. The "transformants" have a number of characteristics which distinguish them from parental cells, and such characteristics in the light of the immunological data suggest that XC cells exposed to HSV can permanently retain HSV-specific information.

Various parameters of diabetic retinopathy among clinic populations.

A study of 402 patients was undertaken in a diabetic clinic at a county hospital. Diabetes was more common in females. Diabetic retinopathy varied directly with the duration of diabetes. Retinopathy occurred earlier with late onset diabetes as compared with diabetes occurring in younger patients. The incidence of raised intraocular pressure was 13.1%. Raised intraocular pressure did not seem to protect the eye against diabetic retinopathy. Good control of diabetes did not seem to affect the nature of established retinopathy. The incidence of retinopathy was higher if hypertension was present.
