Alloantiserum recognizing a DQw2 split which is associated with DR3.

A typing serum MUE 38539 II, was found to recognize a DR3-associated split of DQw2. In cytotoxicity tests, MUE 38539 II yielded positive test results with B lymphocytes but not with monocytes of DR3-positive cell donors. This was in contrast to other typing reagents for DR3 that react with B lymphocytes as well as monocytes. Lymphocytotoxicity tests using MUE 38539 II were negative with DR7- and DQw2-positive cells. The assumption that the serum recognizes a DR3-associated split of DQw2, and not DR3 itself, was confirmed by the lack of reactivity with a DQw4- and DR3-positive lymphoblastoid cell line (RSH). The assumption was also corroborated using reagents from a family in which DR3 and DQw2 were not found in the usually described linkage. In two lines, DR3 was associated with DQw- (2707 and 2710), and in the cell line 2704, DQw2 was associated with DRw-. The serum MUE 38539 II was exclusively cytotoxic with lymphoblastoid cell lines from those family members who were positive for DQw2, independently of the DR3 antigens of the cells.

[The microbial colonization pattern of newborn infants. What is its significance? Results of a study at a district hospital in the Gera district].

A microbiological investigation was performed under standardized conditions both on mother and child for total of 151 births, 112 of which being classified as normal, the remainder of 39 as risk births. Significant differences were found between the microbial colonization patterns for normal and risk births, respectively, as judged from cultures of smears from maternal cervices and from their neonates. In addition, colonization patterns of the newborn children turned out to be age-dependent. No correlation between the colonization pattern of the neonatal surface and infections of the newborn infants has been found.

Hyperzincuria and hypozincemia in patients treated with cisplatin.

The effect of cis-diamminedichloroplatinum (DDP) on plasma and urinary zinc was studied in fifteen patients with squamous cell carcinoma. A decrease in plasma zinc accompanied by an increase in urinary zinc excretion was observed. Pretreatment plasma or urinary zinc did not correlate with tumor size or site, nor was there a correlation between changes in these measurements and a response to therapy. The enhanced excretion of urinary zinc may be related to a decrease in amino acid resorption in the proximal tubule. It is concluded that DDP administration may result in hyperzincuria and could potentially precipitate a symptomatic zinc deficiency state.

A clinical study of maxillary canine retraction with a retraction spring and with sliding mechanics.

The efficiency of maxillary canine retraction by means of sliding mechanics along an 0.018-inch labial arch and an AlastiK chain was compared with that using the canine retraction spring designed by Gjessing. The rate of canine retraction and degree of tipping, and rotation of the canines were studied in 21 subjects by one of these two methods on either side of the dental arch. Measurements were made in the mouth and on photographs of dental casts. The canine was retracted faster and with less distal tipping with the spring than with the sliding mechanics. The canine retraction spring was not superior to the sliding mechanics in controlling canine rotation during the retraction.

Partial purification and characterization of the major endoamylase of mature pea leaves.

An endoamylase from leaves of pea (Pisum sativum) was purified to near homogeneity by affinity chromatography and ultrafiltration with a yield of about 20%. The purified protein had a specific activity of 686 to 1300 units per milligram protein. Molecular weights of 45 and 41 kilodalton were determined by SDS-PAGE and molecular sieve chromatography, respectively. The purified protein exhibited an action pattern commensurate with that of an endoamylase and exhibited properties indicating it to be very similar to cereal grain alpha-amylases (calcium requirement, stability to heat, lability to low pH-values, insensitivity to sulfhydryl reagents). Leaf frationation studies indicated that the enzyme was not primarily located in assimilatory mesophyll cells. Chloroplasts isolated from the leaves were found to contain endoamylases, but their activities represented only a small proportion of the total amylolytic potential of the leaf and reflected for the most part properties quite different from those exhibited by the purified enzyme.

Urinary excretion of N-nitrosamino acids and nitrate by inhabitants in high- and low-risk areas for stomach cancer in northern Japan.

Three samples of 24-h urine were collected from each of 104 inhabitants of high-risk (Akita) and low-risk (Iwate) areas for stomach cancer in northern Japan, according to the following protocols: (i) when they were undosed, (ii) after ingestion of proline three times a day and (iii) after ingestion of proline together with vitamin C three times a day. These samples were analysed for N-nitrosamino acids, nitrate and chloride ion as indices of the exposure. The median values of N-nitrosoproline (NPRO) and N-nitroso-2-methylthiazolidine 4-carboxylic acid (NMTCA) excreted in the urine of undosed subjects were not different between the two areas; however, that of N-nitrosothiazolidine 4-carboxylic acid (NTCA) was significantly higher in subjects of the high-risk area. Salt intake, estimated from the level of chloride ion in the urine, did not differ in two areas. After intake of proline, the NPRO level increased significantly only in subjects of the high-risk area, but not in those of the low-risk area; intake of vitamin C inhibited this increase of NPRO and lowered the levels of other nitrosamino acids only in the high-risk subjects. In contrast, the urinary level of nitrate was higher in subjects of the low-risk area than in those of the high-risk area; nitrate levels were found to correlate well with the amounts of vegetables consumed.(ABSTRACT TRUNCATED AT 250 WORDS)

Induction therapy in head and neck cancer. A comparison of two regimens.

Seventy-two patients with advanced resectable head and cancer received two courses of induction chemotherapy before definitive therapy. Forty-six patients were treated with platinol, Oncovin (vincristine), and bleomycin. Twenty-six received Platinol (cisplatin), Velban (vinblastine), and 5-fluorouracil (5-FU). Although both regimens had an overall response rate of 80% or more, the bleomycin-containing regimen had a higher complete response rate and better long-term disease control, with greater than 60% probability of remaining disease-free (36 month minimum follow-up). As given in this regimen, the 5-FU regimen was well tolerated but had a higher incidence of stomatitis and a low rate of complete responses. When the two regimens were compared to a historical control, the regimen with Platinol, Oncovin, and bleomycin was significantly better.

Exoamylase activity in vacuoles isolated from pea and wheat leaf protoplasts.

Vacuoles isolated from pea (Pisum sativum), and wheat (Triticum aestivum) leaf protoplasts contained considerable activities of electrophoretically highly mobile exoamylases. Vacuoles from spinach (Spinacia oleracea) leaf and photoautotrophic Chenopodium rubrum suspension culture cell protoplasts were devoid of amylolytic activity. Endoamylase activity was in all cases associated primarily with the chloroplast.

Purification and properties of spinach leaf debranching enzyme.

Starch debranching enzyme was purified from intact spinach (Spinacia oleracea L. cv Vital) chloroplasts and from a spinach leaf extract using affinity chromatography on Sepharose 6B-bound cycloheptaamylose (Schardinger beta-dextrin). The enzyme from both sources was homogeneous upon sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Spinach leaf debranching enzyme appears to consist of a single polypeptide chain, since the molecular weight of the native protein (110,000 daltons) was not changed by treatment with sodium dodecyl sulfate. Only one spinach leaf debranching enzyme band could be detected after electrophoresis of a leaf extract on amylopectin-containing polyacrylamide gel, the retardation factor of which coincided with that of the single band seen with the chloroplast enzyme. The purified enzyme exhibited strong pullulanase activity, the specific activity being 69 units per milligram protein with pullulan and 22 units per milligram protein with amylopectin. Cycloheptaamylose is a potent competitive inhibitor of spinach leaf debranching enzyme. The pH optimum of the enzyme was found to be 5.5. The purified enzyme is rather unstable at both 20 degrees and 0 degrees C. Part of the activity lost under storage or at a suboptimal pH could immediately be restored by the addition of thiols. The reactivatable protein, being of the same molecular weight as the native enzyme, exhibited a somewhat altered electrophoretic mobility resulting in one or two minor bands on a zymogram.

Development of Nitrogen Assimilation Enzymes during Photoautotrophic Growth of Chenopodium rubrum Suspension Cultures.

Chenopodium rubrum cells were grown in suspension as a photoautotrophic culture with a 16 hour day. Cell growth had three phases: a 3-day lag, a 3-week logarithmic phase, and a 10-day stationary phase. Chlorophyll content increased steadily during log phase and reached a level of 0.5 to 0.6 mg Chl g(-1) fresh weight. Soluble protein of the cells increased more rapidly from day 4 to day 12 than during midlog phase. Initially, ammonium was taken up in preference to nitrate. However, during the second two weeks of growth, ammonium and nitrate were taken up simultaneously; this period of growth was the time of highest rates of N uptake by the cultured cells. Glutamine synthetase had a high specific activity (17 mumol.hour(-1) mg(-1) protein) in day 1 cells, and this level was sustained until midlog phase when it increased by 20%. Methyl viologen-dependent glutamate synthase specific activity increased rapidly in lag phase cells (day 4 = 10 mumol.hour(-1) mg(-1) protein), but decreased by day 9 to about 50% of the peak and remained constant. NADH:nitrate reductase specific activity increased rapidly in lag phase cells and reached a plateau that lasted from day 4 to 14 (1 mumol.hour(-1) mg(-1) protein). Methyl viologen-dependent nitrite reductase specific activity was high when assayed on day 5 and increased to a maximum on day 15 to 16 (12 mumol.hour(-1) mg(-1) protein). NADPH- and NADH-dependent glutamate dehydrogenase specific activities remained rather constant throughout the growth cycle. The cells appeared to have developed photosynthetic competence and to have leaf-like activities of nitrogen assimilation enzymes.

[Thoracic surgical interventions].

Thoracic surgery necessitates more intensive care postoperatively than general surgery. For elective lung resections mortality was significantly higher in patients older than 70 years. Operative risk has to be considered very thoroughly versus natural history of the diseases involved. Vascular reconstructions in the chest, however, were performed mainly in symptomatic patients, when increased operative risk must be accepted because of an increased threat by symptomatic disease. For surgery of the trachea or esophagus techniques are preferred which avoid thoracotomy.
