The effect of chronic ethanol feeding on ornithine decarboxylase activity and liver regeneration.

The effects of ethanol on liver regeneration are poorly understood. Acute and chronic exposure to ethanol have been found to exert opposite effects on the induction of ornithine decarboxylase, the rate-limiting enzyme for polyamine biosynthesis. Polyamines are necessary for DNA synthesis and liver regeneration after chemical or surgical liver injury. Short-term exposure to ethanol, which inhibits ornithine decarboxylase has been shown to inhibit DNA synthesis and liver regeneration, whereas more chronic exposure to ethanol increases ornithine decarboxylase activity and therefore could conceivably stimulate DNA synthesis and regeneration. To explore this later possibility, the effects of chronic ethanol consumption on ornithine decarboxylase activity, DNA synthesis and liver regeneration were studied in rats after sham laparotomy and partial hepatectomy. Chronic ethanol feeding failed to inhibit the induction of ornithine decarboxylase that occurred after partial hepatectomy and yet significantly inhibited posthepatectomy DNA synthesis and restitution of liver mass. These data suggest that the induction of hepatic polyamine biosynthesis is dissociated from DNA synthesis and liver regeneration after chronic consumption of ethanol.

A replication study on P300 single trial analysis in schizophrenia: confirmation of a reduced number of 'true positive' P300 waves.

A single trial analysis of event-related potentials (auditory odd-ball paradigm) of 20 schizophrenics was performed in comparison to matched healthy controls. The purpose of the study was to test the hypothesis that in schizophrenia the well-known P300 amplitude reduction of averaged event-related potentials is due to fewer elicited single trial P300 waves. The results of the present study support this finding of our previous exploratory investigation and point to the view that schizophrenics reveal basal disturbances in information processing due to inadequately elicited electrophysiological responses to target stimuli.

Electrophysiological evidence for an inverse benzodiazepine receptor agonist in panic disorder.

Inverse agonists of the GABA(A) receptor clearly decrease the amplitudes of the spontaneous EEG in the beta-frequency range. Therefore, we tested the hypothesis that panic patients exhibit a reduction of the EEG's spectral power in the beta-frequency band. Ten unmedicated patients with panic disorder and agoraphobia according to DSM-III-R criteria and 10 matched controls were investigated under baseline conditions, after hyperventilation and 30 min after hyperventilation. EEG recordings from the position Pz and Cz were performed under eyes closed conditions. At baseline conditions the patients suffering from panic disorder depicted a reduced beta-power reaching statistically significance for lead position Pz. Immediately after hyperventilation for both channels we observed a decreased beta-power. After hyperventilation we observed the same situation as under baseline conditions. Taken together, our results point to the view that in panic disorder an endogenous inverse agonist of the GABA(A)-benzodiazepine receptor could be hypothesized.

The benefit from whole body acupuncture in major depression.

BACKGROUND: In a single-blind placebo-controlled study design we investigated the efficacy of acupuncture additionally applied to drug treatment in major depression. METHODS: We randomly included 70 inpatients with a major depressive episode in three different treatment groups: verum acupuncture, placebo acupuncture and a control group. All three groups were pharmacologically treated with the antidepressant mianserin. The verum group received acupuncture at specific points considered effective in the treatment of depression. The placebo group was treated with acupuncture at non-specific locations and the control group received pharmacological treatment plus clinical management. Acupuncture was applied three times a week over a period of 4 weeks. Psychopathology was rated by judges blind to verum/placebo conditions twice a week over 8 weeks. RESULTS: Patients who experienced acupuncture improved slightly more than patients treated with mianserin alone. CONCLUSIONS: Additionally applied acupuncture improved the course of depression more than pharmacological treatment with mianserin alone. However, we could not detect any differences between placebo and verum acupuncture.

[Bilateral necrobiotic xanthogranuloma of the eyelids with associated paraproteinemia: Case report and review of literature].

BACKGROUND: Necrobiotic xanthogranuloma is a rare disorder which predominantly affects the periorbital region and is frequently associated with paraproteinemia and lymphoproliferative diseases. PATIENT AND METHODS: A 49-year old woman presented with bilateral, yellowish, subcutaneous lesions involving all four eyelids. These lesions have developed over the last year. Additionally, similar skin lesions developed on the trunk and the lower extremities. Two years ago, the patient had a bilateral episcleritis. An incisional biopsy was taken form the lesion. In addition, we reviewed the literature concerning similar cases. RESULTS: Histologic examination revealed a granulomatous process consisting of histiocytes, foamy cells, multinucleated giant cells (touton- and foreign body-type) and associated with necrobiotic collagen and cholesterol clefts. General examination revealed the presence of a paraproteinemia of the IgG-typ associated with a hyperlipidemia. The patient was treated with a low dose chemotherapy which resulted in improvement of signs and symptoms. We could identify in the literature 75 similar cases. The mean age of those patients was 53 years. CONCLUSION: The clinical and histologic findings in our patient were typical for a necrobiotic xanthogranuloma with associated paraproteinemia. The most favorable treatment response has been reported with low doses of systemic administered chemotherapeutic agents in combination with radiation therapy; however, the mortality due to underlying diseases is high.

[Functional evaluation of shoes for children based on gait analysis of children in the learning to walk stage].

RESEARCH QUESTION: Although gait patterns of children are well described in gait analysis studies for barefoot gait, loading patterns in gait with shoes have not been analysed compared to barefoot gait. METHODS: 30 children (age 1.8 to 4.8 years) were examined. Ground reaction forces were recorded using two series connected Kistler plates, taking the 3D coordinates and the time coordinate. The gait aspect was recorded using a video system. Three shoe types were tested against each other and against barefoot gait. RESULTS: 1399 gait cycles, 739 barefoot, 660 with shoes were usable for calculation. Statistic analyses comparing barefoot gait to gait with shoes showed: A significant increase in ground contact duration, a shifting of maximal load from the rear towards the midfoot area, a significant increased maximal load, a cushioning of the initial impact at primary ground contact. A comparison between the shoes showed no significant influence of a medial subtalar orthotic support. CONCLUSION: Shoes cause a significant change of loading patterns. But there are also significant differences between different shoe types. CLINICAL RELEVANCE: Shoes for children must provide a stabilisation for the hindfoot, and must not restrict motion in the areas of the ankle and the metatatrsophalangeal joints. The amount of load in the different gait phases must be considered for the construction of a dynamic shoe for children.
