[Systemic artery-pulmonary vein fistula. Apropos of a case].

Fistulas between a systemic artery arising from the aorta and the pulmonary vein with absence of a branch of the pulmonary artery and without abnormality of the bronchial tree are very seldom. We report the case of a child which had a continuous vascular murmur with heart failure secondary to a left-left shunt. A left inferior lobectomy was performed. The inferior lobe was the site of a huge arterio-venous fistula between an artery arising, from the aorta and the pulmonary veins.

The pharmacological effects of acute and chronic clenbuterol treatments after lesions of central noradrenergic nerve terminals.

Acute administration of clenbuterol, a lipophilic beta-adrenergic agonist, decreases motor activity and antagonizes the reserpine-induced hypothermia in mice. After chronic administration of clenbuterol, the acute effect on motor activity disappears (tachyphylaxis) and the acute effect on reserpine hypothermia is potentiated (facilitation). These effects of clenbuterol (either acute or chronic + acute treatments) are not abolished after specific lesions of the noradrenergic system by the neurotoxin DSP-4 which reduces the cerebral levels of norepinephrine to 30% of controls. Although it cannot be excluded that a 70% lesion may be insufficient, another explanation is that beta-adrenergic receptors involved in hypomotility and in reserpine-induced hypothermia may not be located on noradrenergic neurons or may be different from the post-synaptic beta-adrenergic receptors which become hypersensitive after DSP-4 denervation.

Dose related sequence of effects induced by the DA agonist 2-(N,N-dipropyl)-amino-5,6-dihydroxytetralin.

2-N,N-dipropylamino 5,6-dihydroxytetralin (nPr2 ADTN), a potent dopaminergic agonist, induces the same polyphasic effects observed with apomorphine by Protais et al., (1983). The dose related sequence of behaviours in mice can be summed up as followed: to 0.9 to 3.9 micrograms.kg-1, a decrease in motor activity without any other effects; to 7.8 to 15.6 micrograms.kg-1, a return to control level or an increase in motility alone according to experimental conditions; to 31 to 62.5 micrograms.kg-1, a second phase of hypomotility when hypothermia develops; above 125 micrograms.kg-1, a second phase of hypermotility when both stereotypies and climbing behaviour appear. Hypothermia moreover, disappears. The effects of the nPr2 ADTN were compared with those of apomorphine obtained in others studies (Puech et al., 1974; Protais et al., 1983). The tetralin derivative differs from apomorphine in that the first phase of hypermotility it induces is greater, and that hypothermia disappears at high doses.

Adaptation of the tail suspension test to the rat.

In the tail suspension test (TST), the rat is suspended by the tail for 6 min during which the animal shows periods of agitation and immobility. The duration of the immobility is measured. Desipramine decreased the duration of immobility. The main advantages of this procedure are: the use of a simple, objective test situation; the concordance of the results (for desipramine) with the "behavioral despair" test described by Porsolt; the avoidance of the hypothermia induced by immersion in the Porsolt test.

[Interactions between hypnotics and psychotropics. Various aspects of experimental pharmacology].

Fourteen french pharmacologists working for the pharmaceutical industry were asked about the tests they used for the primary screening and the study of hypnotic drugs. They were also asked about the tests using interactions between hypnotics and the studied drugs, either psychotropic or not. The results of survey are presented. A systematic study was performed concerning the interaction of psychotropic drugs with either barbital or pentobarbital in mice. The utilization of these two tests is useful for the research of possible inductory or inhibitory effect on hepatic enzymatic systems. For a complete study of drugs with a profile of psychostimulant or antidepressant.
