Suitability of the YNZ22 (D17S5) VNTR polymorphism for legal medicine investigations in the population of Catalonia (Spain).

Allele and phenotype frequencies for the YNZ22 locus were determined in a population sample from Catalonia (Spain) using the polymerase chain reaction (PCR). In 311 unrelated individuals, 14 alleles and 56 phenotypes were observed. No deviation from Hardy-Weinberg equilibrium was found. The observed heterozygosity was 81.35%. The YNZ22 polymorphism is useful for paternity testing with a CE value of 70% and an Essen-Möller value of 9.35 (log.).

[Left hemicolectomy and intraoperative antegrade lavage in emergency surgery of the left colon].

We report our results with a left colonic resection and intraoperative antegrade colonic irrigation technique with primary anastomosis. Thirty five consecutive patients operated on in the Emergency Surgical Ward are presented. Twenty five with large bowel occlusion and 10 with sigma perforation. Anastomotic leakage (2 patients, 5.7%) and postoperative hospital stay (mean 15 days) were similar to cases of elective surgery. The intraoperative antegrade colonic irrigation technique has become the first choice in our Department to treat any patient with left colonic occlusion or perforation. Only patients with faecal peritonitis or ischemic colon were excluded.

Multiple use of blotted polyvinylidene difluoride membranes immunostained with nitro blue tetrazolium.

A simple procedure for reusing polyvinylidene difluoride membranes with bound proteins reacted with antibodies and developed by the nitro blue tetrazolium method (M. S. Blake, K. H. Johnston, G. J. Russell-Jones, and E. C. Gotschlich (1984) Anal. Biochem. 136, 175-179) is described. The blue diformazan precipitate produced by the reduction of the nitro blue tetrazolium salt was dissolved by soaking the membrane in dichloro- or trichloromethane for 10-60 min at room temperature. After equilibration in methanol and then in distilled water the first and second antibodies were removed by incubating the membrane in 0.2 M glycine-HCl buffer, pH 2.2, for 90 min at room temperature. These cleaned membranes could be successively reused with different monoclonal antibodies or preimmune serum up to 21 times. The intensity of the spots or electroblotted bands and their specific reaction pattern with different monoclonal antibodies remained unchanged, and no background was observed when the membranes were reacted with preimmune serum. This procedure makes it possible to reuse membranes freshly developed or stored for long periods without loss of resolution.

Effect of age on serum concentrations of the third component of complement in dogs.

Single radial immunodiffusion was used to determine the concentration of the third component of complement (C3) in canine sera from 48 normal dogs in the following age groups: < 12 months, 12 to 24 months, and > 24 months. Mean serum C3 concentrations, expressed as the percentage of pooled reference sera from 20 dogs, showed a significant (P < 0.017) correlation with age in dogs less than 12 months old, while the character of this correlation was negative for dogs more than 24 months old (P < 0.056). Two-way analysis of variance, for age and sex, revealed significant differences between C3 values from this two groups that were not present when compared with dogs aged between 12 and 24 months old. Sex had no effect on serum C3 concentrations.

Distribution of transferrin subtypes in Tarragona (east Spain).

The distribution of transferrin (TF) subtypes was determined by isoelectric focusing of sera from 284 unrelated individuals from Tarragona (south of Catalonia). The allele frequencies observed, TF*C1 = 0.805, TF*C2 = 0.162, TF*C3 = 0.026 and TF*B = 0.007 were similar to those reported for other Spanish populations.

Brief communication: immunoglobulin (Gm and Km) allotypes in two populations of Catalonia (Spain).

Serum samples from two populations of Catalonia, Spain, 208 from Olot (Gerona) and 209 from Tortosa (Tarragona), were typed for G1m (1, 2, 3, 17), G3m (5, 10, 11, 13, 14, 15, 16, 26), and Km (1). The Gm patterns of the Catalonian populations are characterized by the presence of four haplotypes, Gm 1,17;21,26 Gm 1,2,17;21,26 Gm 1,3;5,10,11,13,14,26 and Gm 3;5,10,11,13,14,26. The homogeneity for haplotype Gm 1,17;21,26 among our data and other European populations suggests the existence of an isofrequency line which starts from the Mediterranean zone of Iberian Peninsula and continues through the northwestern part of Europe. From this line a decreasing cline towards the south can be observed. For the haplotype Gm 1,2;17,21,26, affinities are observed between Catalonian populations and other populations from central Europe. This confirms the existence of a gradient towards low values from NW to SE. The presence of the typical Mongoloid haplotype Gm 1,3;5,10,11,13,14,26 is discussed in this paper. No significant differences in the frequencies of the Km1 allele were observed among the European populations.

Detection of double-stranded RNA by ELISA and dot immunobinding assay using an antiserum to synthetic polynucleotides.

An antiserum against polyinosinic-polycytidylic acid (In-Cn) was used to detect double-stranded RNA (dsRNA) by several serological techniques. DsRNA was readily detected by indirect ELISA (ELISA-I) and dot immunobinding assay (DIA). Addition of the antigen to poly-L-lysine-precoated plates and blocking with uncreamed milk powder allowed detection levels of 100 pg.ml-1 In-Cn by ELISA-I. Concentrations as low as 1 ng.ml-1 were detected by DIA using polyvinyliden difluoride (PVDF) membranes. Detection capacity with nitrocellulose membranes was 1000 times lower than with PVDF. ELISA-I and DIA enabled detection of dsRNA in enriched fractions from cucumber mosaic virus (CMV)- and citrus tristeza virus (CTV)-infected plants and from virus-infected Penicillium chrysogenum mycelium. These techniques showed similar or higher sensitivity for detection of dsRNA than separation by polyacrylamide gel electrophoresis and silver staining.

[Keratitis caused by Pseudomonas aeruginosa and secondary endophthalmitis].

We report three patients with Pseudomonas aeruginosa keratitis. Two patients in the Intense Care Unit had exogenous bacterial endophthalmitis assessed by histopathology; the infection had probably a nosocomial origin and the outcome was fatal. The third case was associated with the use of soft contact lenses and even though clinically endophthalmitis was suspected there was a favourable response to treatment. We review diagnostic aspects and discuss the management of bacterial keratitis and endophthalmitis.

Frequency of the ESD*5 allele in a Spanish population.

The red blood cell esterase D (ESD) polymorphism was studied by means of IEF in a North-East Spanish population (Barcelona). Gene frequencies in 430 unrelated individuals were ESD*1: 0.888, ESD*2: 0.091 and ESD*5: 0.021. Our data confirm previous results showing that ESD*5 occurs in polymorphic frequency and has a Caucasian origin.

Glyoxalase I polymorphism in Catalonia (Spain).

Blood samples from two Catalonian populations (North-East of Spain) were analysed for glyoxalase (GLO) I polymorphism. Gene frequencies of GLO1 (0.46 and 0.41) are comparable to those reported for Central-European Caucasoid populations.
