[The enzymes of the enterocyte. Prospects for a functional exploration].

The authors recall the enzymatic activity and transport activities which were localised in the glycocalyx, the microvillous membrane, the intracellular compartment and the basolateral membrane of the enterocyte. The relationships between the processes of digestion and absorption, on the one hand, active transport, energy metabolism and sodium and potassium dependent ATPase, on the other hand are considered. The application of these data to functional investigation of the enterocyte is discussed, emphasizing the complementary character of the biological tests used currently and the importance of sodium and potassium dependent ATPase in the phenomena of active transport.

On the inhibition of elastase by serum. Some distinguishing properties of alpha1-antitrypsin and alpha2-macroglobulin.

1. The influence of serum on the elastolytic and esterolytic activity of elastase has been studied. With both substrates the inhibition curves are linear. 1 ml of normal human serum inhibits the activity of 0.77 mg of pure porcine elastase. 2. Elastase binds faster with alpha2-macroglobulin (k = 3.4-10(6) M-1 S-1) than it does with alpha1-antitrypsin (k = 5-10(5) M-1 S-1). 3. The dissociation constant of the alpha-antitrypsin -elastase complex is much lower than that of the alpha2-macroglobulin-elastase complex but both complexes are very stable (Ki less than 10(-10) M). 4. Protein pi (inter-alpha-inhibitor) does not inhibit elastase.

Influence of a long or medium chain triglyceride diet on intermediary hepatic metabolism of the rat.

Two groups of rats were given, ad libitum, diets containing 20% fat for 4-5 weeks. The fat consisted of either long chain (LCT) or medium chain triglycerides (MCT). A third group of rats was given a low fat control diet. No difference in weight increase could be found between LCT and MCT rats. Total lipids and cholesterol concentrations in the plasma were the same whatever the diet. The high fat diets caused an increase in hepatic acetyl-CoA, citrate, malate and ketone body concentrations. Concentration of ketone bodies in the blood, however, decreased. No ketonuria occurred in any case. Hepatic levels of adenosine 5'-triphosphate, adenosine 5'-monophosphate and inoganic phosphate seemed higher in MCT rats than in LCT rats. The levels of these substrates were increased in both groups compared with the control group. LCT rats had hepatic (NS) and blood (HS) lactate and pyruvate concentrations below the corresponding figures for control rats. Hepatic and blood lactate concentrations in MCT rats were at least equal to or slightly higher (NS) than those of control rats. Hepatic glucose and glycogen and blood glucose concentrations were higher in rats given the high fat diet.

Studies on intestinal adenosine triphosphatases. I. Application of a semiautomated method to the rat intestinal brush borders.

Elaboration of a semiautomated kinetic test on LKB 8600 apparatus for ATPase is described, using the PK-LDH system. As optimal ionic conditions 3 mmol-1 - minus 1 potassium chloride and 100 mmol-1 - minus 1 sodium chloride are proposed for measurement of (Na+-K+)-ATPase activities of rat intestinal brush borders. NH+4 can substitute for K+. The coefficients of variation of the method are 2.4% for Mg2+-ATPase and 4.9% for (Na+-K+)-ATPase determinations.
