Failure of histiocytosis X cells to express i blood group antigen.

Expression of HLADR, I, i blood group antigen and T6 antigen were studied in Histiocytosis X cells and pulmonary alveolar macrophages using double labelling immunofluorescence technique or immuno-peroxidase procedure. Alveolar macrophages express simultaneously HLADR and i blood group antigen. Histiocytosis X cells, characterized by HLADR and T6 antigens, and by their ultra-structural marker do not express i antigen. These results confirm the hypothesis that histiocytosis X cells constitute a specialized sub-population of the mononuclear phagocyte system.

Blood group antigen expression in nephroblastoma.

The histological distribution of A, B, H, Lewis, Pr, i and I blood group antigens (BGA) was studied in 19 nephroblastomas and one mesoblastic nephroma by means of the indirect immunofluorescence technique. In addition to general conclusions about the relatively limited BGA expression in the tumour cells, their histological distribution was found to be related to the differentiation level of these renal embryonic tumours. By comparison with normal embryonic, fetal and adult kidney it was shown that the nephroblastoma differentiation level is not higher than that of a 3-month fetal kidney. Furthermore, i antigen appears to be a non-specific marker of the histiocytic macrophage cells of the nephroblastoma stroma.

Distribution of blood group antigens in adult pancreas.

Numerous blood group antigens are present in different human tissues where they appear as immunological markers of certain structures. The Pr antigen is the only antigen present in the islet of Langerhans. The A, B, H and Lewis antigens are present in the centro acinar cells and the Pr and Lewis antigens are found on the membrane of the pancreatic ducts. A, B, H and Pk antigens are expressed in the capillaries. These kinds of studies could be important for the transplantation of the endocrine pancreas.

Haemagglutination kinetics using a continuous-flow system.

An automated technique for measuring haemagglutination kinetics is described. Equipment used in this test is very simple and already present in most blood transfusion centres. This method may be used with advantage to differentiate homozygous and heterozygous subjects in Rhesus, MNSs, and Kell systems, and to study antigen variations, especially in genetic investigations.

ABO-blood-group-related idiotypic network: mimicry of oligosaccharide epitope by rabbit antiidiotypic antibodies to murine monoclonal anti-A antibody.

The idiotypy of antibodies (Ab) specific for oligosaccharide determinants of blood groups of the human ABO system was studied through a cascade. Xenogenic antiidiotypic Ab (Ab2) raised in rabbits to the murine monoclonal anti-A61 (Ab1) were screened for reactivity with various anti-ABH Ab. Three anti-A and three anti-A,B monoclonal antibodies (mAb) which were developed in the same mouse strain as that producing Ab1, as well as a human polyclonal anti-A, were found to share cross-reactive idiotopes (CRI) with Ab1. CRI on murine mAb could be due to a Biozzi recurrent Id on anti-A Ab reacting with anti-Id "à la Oudin", while CRI on human anti-A Ab suggested the presence of paratope-induced anti-Id. Inhibition by Ab2 of haemagglutination of A, B or O human red blood cells by many murine anti-ABH mAb, and by polyclonal or monoclonal human anti-A, strongly supported the occurrence of anti-Id mimicking ABH epitopes belonging to type 2 determinants carried by human erythrocytes. Furthermore, a rabbit immunized with Ab2 produced a potent Ab3 response characterized by anti-H-type-2 specificity. Altogether, these results are consistent with the first successful production of anti-Id Ab that mimics the tridimensional shape of a well defined and strictly carbohydrate epitope, eliciting a haemagglutinating Ab3.
