Detection of human papillomavirus types 6, 11, 16 and 18 in mucosal and cutaneous lesions by the multiplex polymerase chain reaction.

A multiplex polymerase chain reaction (PCR) based on the simultaneous amplification of human papillomavirus (HPV) types 6/11, 16 and 18 in a single-step procedure was developed, using primers chosen in the E6-E7 region. The specificity and sensitivity of this technique have been proved by amplifying mixtures or various amounts of plasmid-containing HPV DNA; it allowed the detection of as few as 5-25 HPV DNA copies. Application of the multiplex PCR to 71 clinical samples showed that HPV DNA was detected in 80% (45/57 cases) of mucosal biopsies and 35% (5/14 cases) of cutaneous specimens. HPV 16 was predominant in high-grade CIN whereas HPV 6 and 11 were detected more frequently in genital condylomas and laryngeal papillomas. In cutaneous Bowen's disease HPV 16, 18 or 6/11 + 16 were detected and in squamous cell carcinomas HPV 6/11 or 16 were found. After sequence amplification with primers of one HPV type, the clinical samples displayed the same HPV types but the frequency of positive and coinfected lesions increased. Thus, multiplex PCR is a valuable technique for typing HPV DNA but coinfections may be underestimated.

Preparation of conjugates between oligonucleotides and N-vinylpyrrolidone/N-acryloxysuccinimide copolymers and applications in nucleic acid assays to improve sensitivity.

A simple approach has been developed to prepare conjugates between oligodeoxyribonucleotides (ODNs) and linear copolymers of N-vinylpyrrolidone (NVP)/N-acryloxysuccinimide (NAS). These conjugates were of higher than expected molecular weight, as determined by light scattering. Side reactions between amino groups on heterocyclic bases, especially of cytosine, are proposed to explain the formation of aggregates. ODN-NVP/NAS conjugates used in both capture and detection steps in sandwich hybridization increased assay sensitivity. Sensitivity to 10(7) DNA molecules/mL was reached for a 2.3 kb PCR fragment of hepatitis B virus (HBV) genomic DNA.

[Importance of silicone bases in complete dentures].

The use of a silicone base under a total prosthesis makes possible the solution of certain problems of tolerance or maintenance of such denture. I.--Full denture in patients who have received radiotherapy. II.--Full denture with intolerant mucosa. III.--Following failure of classical prosthesis. IV--Prosthesis fitted following pre-prosthetic surgery. The technical problems associated with the fabrication of such prosthesis and possible adjustements are discussed.

[Experimental study on the role of the mylohyoid muscle in the instability of dental prostheses].

Using cinematographic and radiocinematographic recording techniques, the authors studied the movements of inferior prosthesis unstable in the lingual region. Pronunciation of the diphthong La... La... La... causes upward and forward projection of the prosthesis. Electromyography records contraction of the mylo-hyoid during pronunciation of these diphthongs. In the totally edentulous patient, the crest of the mylo-hoid is very close to the alveolar crest. From a clinical standpoint, functional exploration of the mylohyoid is easy, the index finger being placed along the mylo-hyoid crest and the patient asked to say La... La... La... If the index finger is pushed forwards and up, surgical intervention to lower the mylo-hyoid (Trauner's operation) is indicated, in association or not with other procedures designed to treat the other causes of instability or the prosthesis.
