Separation and characterization of needle and xylem maritime pine proteins.

Two-dimensional gel electrophoresis (2-DE) and image analysis are currently used for proteome analysis in maritime pine (Pinus pinaster Ait.). This study presents a database of expressed proteins extracted from needles and xylem, two important tissues for growth and wood formation. Electrophoresis was carried out by isoelectric focusing (IEF) in the first dimension and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) in the second. Silver staining made it possible to detect an average of 900 and 600 spots on 2-DE gels from needles and xylem, respectively. A total of 28 xylem and 35 needle proteins were characterized by internal peptide microsequencing. Out of these 63 proteins, 57 (90%) could be identified based on amino acid similarity with known proteins, of which 24 (42%) have already been described in conifers. Overall comparison of both tissues indicated that 29% and 36% of the spots were specific to xylem and needles, respectively, while the other spots were of identical molecular weight and isoelectric point. The homology of spot location in 2-DE patterns was further validated by sequence analysis of proteins present in both tissues. A proteomic database of maritime pine is accessible on the internet (http://www.pierroton.inra.fr/genetics/2D/).

Expression of cholinergic muscarinic receptor subtypes mRNA in rat blood mononuclear cells.

The cholinergic system plays a role in the neuroimmune network. White blood cells have been found to express muscarinic and nicotinic cholinergic receptors as well as acetylcholinesterase on their surface, and cholinergic agents have been shown to modulate immune functions. Although the presence of muscarinic receptors on white blood cells has been well documented by binding and functional studies, the subtype(s) of muscarinic receptor present on these cells is still unknown. We have previously shown the absence of m2 muscarinic receptor subtype mRNA in rat mononuclear cells (Costa et al., 1994). The expression of m1, m3, m4 and m5 mAChR mRNA was analyzed in this study in rat peripheral blood mononuclear cells by Northern blotting hybridization experiments and reverse transcription-polymerase chain reaction. Only mRNA of the m3 subtype was detected in rat mononuclear cells, at levels about 100 times lower than in brain. Traces of m4 subtype mRNA were found in lymphocytes, at levels about 10,000 times lower than the cerebral cortex, while m5 mRNA was undetectable.

Activation of neutrophil oxidative metabolism during cardiopulmonary bypass with membrane oxygenators.

Neutrophil oxidative metabolism was prospectively assessed in 9 consecutive patients undergoing cardiopulmonary bypass (CPB) with polypropylene hollow fiber oxygenators for open cardiac operations. Blood samples were collected at 10, 20, and 30 min of CPB. The generation of oxygen-free radicals by neutrophils was measured as luminol-enhanced chemiluminescence after stimulation with opsonized Zymosan and Phorbol Myristate Acetate. A significant increase in light emission was detected by using both the chemiluminescence stimulators. These data suggest that the contact of patient blood with foreign surfaces during cardiopulmonary bypass results in activation of phagocyte cells.

Antibiotic and antithromboembolic prophylaxis in prosthetic surgery of the knee.

The prevention of deep sepsis and venous thromboembolism in knee replacement surgery is of great importance for the final success of treatment. General rules of prevention must be followed when using antimicrobial drugs (environment, surgical method, patient selection). Although it does have some negative aspects (overuse, high costs, development of resistant bacteria) the administration of antibiotics is recommended, but only when it is timely, correct, and carried out for a suitable amount of time. The prevention of thromboembolism is based first on the selection of patients who risk the occurrence of angiology-related diseases, on continuous passive postoperative movement, and, finally, on intentional normovolemic hemodilution, for which the procedures used and the advantages obtained are described.

The locked intramedullary Grosse Kempf nail in the treatment of proximal diaphyseal fractures of the femur.

The authors studied the effectiveness of locked intramedullary nailing (Grosse-Kempf) in the treatment of proximal shaft femoral fractures. They reviewed 24 cases with an average two-year follow-up and which had obtained positive clinical radiographic results in 80% of cases as compared to 88% in a previous revision of femoral shaft fractures. The difference is due to the greater difficulty encountered in intraoperative reduction of fractures from which defects in axial (varus) and rotatory (extra) alignment arise. They discuss indications as regards locking type, weight-bearing, as well as surgical techniques, which may prevent some of the most frequent problems observed.

Immunologic recurrent abortion: comparison between immunotherapies.

The Authors record the results in a group of patients with immunologic recurrent abortion (IRA) treated with two different immunoprophylaxis regimen. The first one is an active prophylaxis therapy with preparation and administration from donor mononucleates. According to their previous original experience, the Authors started giving high doses of IV-Ig (HD IV-Ig) in a second group of pregnant patients with the same diagnosis of immunologic recurrent abortion (IRA). The results of this not randomized study show better reproductive outcome in the group treated with use of HD IV-Ig.

Adult polycystic kidney disease: prenatal diagnosis with DNA polymorphic markers.

A prenatal diagnosis of adult polycystic kidney disease (ADPKD) by DNA testing is reported. Evidence showing a linkage between the disease and the DNA markers on chromosome 16 was obtained in the family by linkage analysis and homogeneity testing with Italian families of the linked type. Prenatal diagnosis was performed either by polymerase chain reaction (PCR) of GGG1 fragment either by Southern blotting analysis of the others chromosome 16 markers. Diagnostic results were available by PCR analysis in a few hours and then were confirmed by Southern blotting of the others probes. The foetus was monitored by ultrasounds. At 26th week the foetal kidney were enlarged with small cysts and, at birth, the newborn had bilateral renal cysts, confirming the foetal genotype prediction based on flanking markers.

[Female urinary stress incontinence and insufficient closing pressures. Results of perineal sphincter electromyography].

Prior to a clinical evaluation of the efficacy of sphincter and perineal rehabilitation in female urinary stress incontinence due to striated sphincter incompetence, and in order to define the cause, 32 patients with stress incontinence with very low urethral closing pressure on urodynamic studies, underwent a perineal electromyographic investigation Three types of sphincteric lesion were detected: an isolated lesion of the striated muscle fibres, an isolated neurogenic lesion of the internal pudendal nerve and a neurogenic lesion of the internal pudendal nerve in a context of sensorimotor polyneuropathy.
