New biotinylating reagent utilizing carbodiimide function.

A biotin derivative having a carbodiimide function (fig.1 Carbo-biotin) was synthesized and reacted with DNAs. This compound was used to label biotin to DNAs, by reaction of carbodiimide group and nucleotides as they form stable adducts. Our experiments showed that it is able to bind to both single and double stranded DNAs. The DNA oligomer labeled with this reagent was shown to hybridize to complementary DNA chain. From our results, this compound is expected to be a useful biotinylating reagent for non-radioactive detection.

Distal arterial reconstruction using Esmarch's bandage technique to salvage upper extremity function in thoracic outlet syndrome caused by cervical ribs: a report of two cases.

We present herein the cases of two patients with thoracic outlet syndrome (TOS) who required arterial reconstruction due to gangrene of the fingers and/or hand. In both patients, the cervical ribs had produced intimal injury of the subclavian arteries, and the successive distal arterial embolism brought about severe ischemia of the affected upper extremity. To treat the TOS, the cervical ribs were resected through a supraclavicular incision. In the first patient, arterial reconstruction was performed from the subclavian artery to the radial collateral artery, a branch of the deep brachial artery, which resulted in minimizing amputation of the gangrenous hand. In the second patient, resection and direct anastomosis of the injured subclavian artery were performed, and bypass surgery from a brachial artery to an interosseous artery was carried out, preserving finger function. Reversed saphenous vein grafts were utilized and Esmarch's bandage technique was applied as a substitute for a vascular clamp in both patients. Following these case reports, we discuss the technique of performing distal bypass in the upper extremities and comment on the usefulness of Esmarch's bandage technique for preserving upper extremity function.

Hereditary nephrotic syndrome with progression to renal failure in a mouse model (ICGN strain): clinical study.

The clinical course of murine hereditary nephrotic syndrome (ICGN strain) was determined by examining 201 animals under different conditions. In the early stage, significant hypoproteinemia and hypoalbuminemia developed (p < 0.001) in parallel with a progressive rise in urinary protein concentration (p < 0.001). In the middle stage, the concentrations of total cholesterol, triglyceride, and beta-lipoprotein markedly increased (p < 0.01, p < 0.001, and p < 0.001, respectively), suggesting that type IIb hyperlipoproteinemia developed as in human nephrotic patients. Systemic edema appeared in 8 of 24 animals. In the terminal stage, both BUN and creatinine values greatly increased (p < 0.001), indicating rapid deterioration of renal function. The present study suggests that ICGN mice could be a useful model to study the pathophysiology of human nephrotic syndrome and its progression to renal failure.

[Atheromatous embolization as a cause of postoperative renal dysfunction in infrarenal aortic reconstructive surgery].

Influences of atheromatous changes in the aortic wall at the site of proximal clamping were studied retrospectively on postoperative renal function in an infrarenal aortic reconstruction. Atheromatous lesion was evaluated on CT scan as low density layer in the aortic wall observed in cases with the thickened wall exceeding 3mm, and they were classified into atheromatous group (group II) consisted of 21 patients. In these patients, the postoperative serum creatinine showed significant elevation from preoperative value of 0.89 +/- 0.32 mg/dl to 1.67 +/- 0.90 mg/dl on 1st postoperative day (p < 0.01), 1.25 +/- 0.99 mg/dl on 7th day (p < 0.05) and 1.24 +/- 0.89 mg/dl on discharge (p < 0.05). Correlation between the severity of the postoperative renal dysfunction and the degree of the atheromatous changes evaluated by preoperative CT scan was also defined and 2 patients with severe changes died of acute renal failure. On the contrary, 39 patients without thickened wall (group I) showed no significant elevation of serum creatinine after operation. These results strongly suggest that atheromatous embolization to the kidney plays an important role to cause postoperative renal impairment following infrarenal aortic reconstructive surgery for the patients with atheromatous changes in the aorta near the site of proximal clamping.

Failure to verify high levels of pyrroloquinoline quinone in eggs and skim milk.

The levels of pyrroloquinoline quinone (PQQ) in eggs, skim milk and milk was re-examined by the use of gas chromatography/mass spectrometry. PQQ was extracted from the samples, after addition of [U-13C]PQQ as internal standard, with n-butanol and Sep-Pak C18 cartridges. After derivatization of PQQ with phenyltrimethylammonium hydroxide, molecular peaks at m/z 448 and 462 were used for detection of PQQ and [U-13C]PQQ, respectively, by selected ion monitoring. PQQ could be detected in eggs, skim milk and milk, but their levels were very low (e.g., egg yolk: 7.0 ng/ml; skim milk: 2.5 ng/g dry weight). Our data for eggs and skim milk are far below those measured by the redox cycling method of Gallop's group.

Ascorbate radical levels in human sera and rat plasma intoxicated with paraquat and diquat.

To clarify the toxicological mechanism of action of paraquat and diquat, the level of ascorbate radical, an oxidized product of ascorbic acid, was examined using the electron spin resonance (ESR) method. The ascorbate radical level increased to twice the normal level in sera from humans acutely intoxicated with a 1:1 mixture of paraquat and diquat, and to more than 1.5 times the normal level in rat plasma containing moderate levels of paraquat. The dosage level for rats was 0.025% paraquat dichloride in the diet. The ascorbate radical in both intoxicated human sera and rat plasma decayed much faster than that in normal samples.

A case of appendiceal mucocele showing massive mucous production with concomitant colonic cancer.

A 58-year-old male patient presented with abnormal discharge of 200 ml transparent fecal mucus. Irregular protuberance of the ascending colon into the adjacent ileocecal region was observed by colonofiberscopy, and a cystic lesion in the ileocecal region was suggested by computerized axial tomography. Pale yellowish semitransparent jellied substances were observed exudating from the site of ileocecal resection. The diagnosis established was moderately differentiated colonic adenocarcinoma and a mucocele of the appendix. The mucus discharge disappeared after resection. Biochemical analysis of the mucus suggested the mucocele as a source of the discharged mucus. We report an extremely rare case of mucocele of the appendix that may be related to the cause of mucous stool.

Effects of pyrroloquinoline quinone (PQQ) and PQQ-oxazole on DNA synthesis of cultured human fibroblasts.

The effects of pyrroloquinoline quinone (PQQ) and PQQ-oxazole (PQQ-glycine adduct) on DNA synthesis were examined using cultured human fibroblasts. Confluent fibroblasts were cultured in serum-free Dulbecco's modified Eagle's media, and various concentrations of PQQ and PQQ-oxazole were added to the media. After incubation for 24 h, [3H]thymidine was added to the media as an indicator for DNA synthesis of the cells. The thymidine incorporation into the cells was significantly enhanced even in the presence of very low concentrations of PQQ (0.003-0.03 microM); it remained significantly enhanced, up to 30 microM PQQ. However, the incorporation remarkably decreased at 750-1500 microM of PQQ. In contrast to the results for PQQ, DNA synthesis was not stimulated by addition of 0.003-3 microM PQQ-oxazole, but it was slightly enhanced at concentrations 15-750 microM. In morphological examination of the cultured human fibroblasts, cell density was increased by addition of 0.003-30 microM PQQ when compared with that of the control, supporting the above biochemical data. However, there were no distinct differences in morphological effect between PQQ and PQQ-oxazole.

Positive- and negative-ion mass spectrometry of diphenylmethane antihistaminics and their analogues and rapid clean-up of them from biological samples.

Positive-ion electron impact (PIEI), positive-ion chemical ionization (PICI) and negative-ion chemical ionization (NICI) mass spectra are presented for 15 compounds of diphenylmethane antihistaminics and their analogues, and each fragmentation pathway was analyzed. In the PIEI mode, molecular peaks were very small or missing for most compounds. Peaks at m/z 58, due to a dimethylaminomethyl group liberated, constituted base peaks in five compounds. Peaks at m/z 165 and/or 167, due to diaromatic rings plus a methyl group, appeared in most compounds. In the PICI mode, peaks due to M+H and M+C2H5 appeared in all compounds. Peaks due to diaromatic rings plus a methyl or ethyl group constituted base peaks in five compounds, which had an ether bond in their structures. In the NICI mode, anions at m/z M-H appeared in most compounds. Peaks at m/z 35 were observed for compounds having a chlorine group in their structures. Detection limits for total ion monitoring of these compounds were 20-50 ng on column in the PIEI mode, 100-200 ng in the PICI mode and 500-1000 ng in the NICI mode. A rapid and simple clean-up procedure of these drugs with use of Sep-Pak C18 cartridges is also presented. The drugs could be detected by gas chromatography with DB-1 and DB-17 capillary columns with satisfactory separation from impurities in their underivatized forms. The recovery of the drugs, which had been added to whole blood and urine, was more than 60% except for meclizine.

Maclura pomifera agglutinin-binding glycoconjugates on converted apatite from synthetic octacalcium phosphate implanted into subperiosteal region of mouse calvaria.

We have previously shown that the mineral in granules of synthetic octacalcium phosphate (OCP) implanted subperiosteally in mouse calvariae was converted to apatitic crystals and that the OCP implantation stimulated bone formation. The matrix components accumulated on the converted apatite were very similar to those of bone nodules (starting locus of calcification) in intramembranous osteogenesis. In the present study, the nature of the matrices accumulated on OCP implants in calvariae was compared with that of the matrices accumulated in abdominal subcutaneous implants. The comparison was facilitated by the use of Maclura pomifera agglutinin (MPA) lectin which is known to have a high affinity for the primary intramembranous bone matrix. Micro-beam x-ray diffraction indicated conversion of the implanted OCP to apatitic crystals in situ, both in subperiosteal and subcutaneous sites, after 10 days. Additional bone formation was detected on the converted apatite after 13 days in subperiosteal implantation, whereas bone was not formed in the subcutaneous implantation. MPA reaction was strongly manifested after 10 days in matrices accumulated on the converted apatite in both subperiosteal and subcutaneous implantations. Biochemical data showed that intensely and weakly MPA-blotted molecules (53.0 and 152.6 kDa, respectively) were in all the mouse sera, in the guanidine HCl-EDTA extracts of mouse calvarial bone and in the extracts of the implanted OCP in both subperiosteal and subcutaneous sites. These findings indicated that the glycoconjugates accumulated on the converted apatite from OCP were similar to the glycoconjugates in the serum in terms of reactivity with MPA and molecular weights. Furthermore, the results suggest that MPA-binding glycoconjugates which had accumulated on the converted apatite may be a requisite for the differentiation of mesenchymal cells into osteoblasts in periosteum but not in subcutaneous sites.

Development of preimplantation guinea-pig embryos in serum-free media.

This study was undertaken to determine how far preimplantation guinea-pig embryos can develop normally in vitro with serum-free media generally used for mouse (Whitten's medium, WM) and rabbit (Kane's medium, KM) embryos. Preimplantation guinea-pig embryos were recovered from naturally mated females at each day of pregnancy between Day 1 and Day 6, and were cultured in vitro with either WM or KM at 37 degrees C in 5% CO2 in air. One-cell (Day-1) and 2-cell (early Day 2) embryos cultured in WM developed to the 4-cell and morulae stages respectively, whereas the corresponding embryos cultured in KM arrested development at the 2-cell and 8-cell stages respectively. In contrast, 4-cell (late Day 2 and Day 3) embryos proceeded to the blastocyst stage in KM, but not in WM. Eight-cell embryos (Day 4) and morulae (Day 5) developed to blastocysts in either medium. However, hatching of the blastocysts and trophoblast outgrowth on the culture dish were supported only by KM even when collected at the blastocyst stage. The present study suggests that, in guinea-pig embryos, a distinct change in nutritional requirements occurs around the time of the second cell division and that, unlike in the mouse, trophoblast outgrowth in the guinea-pig requires no supplementation of serum or extracellular matrices.

Are diabetic hearts more resistant to ischemia/reperfusion injury?

The present study was designed to investigate the cardiac impairment during ischemia/reperfusion injury in rats with streptozotocin-induced diabetes vs controls. Male rats were divided into three groups: controls, one-week diabetic rats (1wDM) and four-week diabetic rats (4wDM). In the isolated working heart, left ventricular developed pressure (LVDP), left ventricular end-diastolic pressure (LVEDP), and maximum positive and negative dP/dt were measured before ischemia and after 20 min of global ischemia followed by 30 min reperfusion. In the pre-ischemic state, hearts of 4wDM showed a significant (p < 0.05) depression of the maximum positive and negative dP/dt compared with those of controls and 1wDM. There were no significant differences in LVDP and LVEDP among the three groups. The incidence of reperfusion-induced ventricular fibrillation (VF) was 75% for controls, 15% for 1wDM rats, and 27% for 4wDM rats. In hearts without reperfusion-induced VF, there were no differences in the three groups, between the pre- and post-ischemic values in LVDP, LVEDP, and maximum positive and negative dP/dt. These findings suggest that diabetic hearts exhibit no susceptibility to ischemia/reperfusion injury.

Effects of diabetes on lipid peroxidation and scavenger activity in ischemia/reperfusion injury.

This study examined the effects of ischemia/reperfusion on cardiac function and diabetes-associated lipid peroxidation and scavenger induction in isolated working hearts. Cardiac function in diabetic rats was depressed in proportion to the duration of diabetes. Plasma levels of malondialdehyde (MDA) were increased significantly after diabetes of 1-week duration (1wD). MDA levels in the myocardium were increased significantly after 4-weeks (4wD) and tissue superoxide dismutase (SOD) activity was also elevated significantly in 8-week diabetic rats (8wD) as compared to control values. In both controls and diabetics, tissue MDA concentrations increased significantly during the first 3 min of reperfusion following 20 min of ischemia, compared with preischemia levels. At the end of reperfusion, tissue MDA levels decreased in controls and 1wD hearts, but remained elevated in the 4wD and 8wD cases. The SOD activity in 8wD hearts decreased significantly after 20 min of global ischemia followed by 30 min reperfusion, compared with preischemia levels. By contrast, there were no significant differences between preischemic and after-reperfusion data for parameters of cardiac function in 8wD rats. Thus, lipid peroxidation appears to have little influence on cardiac function during ischemia/reperfusion in rats with chronic diabetes.

Hereditary hydronephrosis in C57L/MsNrs mice.

Inbred C57L/MsNrs mice (66 males and 72 females) maintained at the National Institute of Health, Japan, were examined grossly for hydronephrosis. Unlike the hydronephrotic strains of mice reported so far, C57L mice showed no sex-related difference in the incidence (62% in males and 75% in females; P > 0.05) and severity of hydronephrosis. The right kidney was more severely affected than the left kidney in both sexes. Age appeared to have no influence on the incidence.
