Autoradiographic demonstration of deoxyglucose uptake in guinea pig cochlea.

The 2-deoxyglucose autoradiographic method was applied to whole-body cryosectioning to include the cochlea. The highest levels of 2-deoxyglucose uptake were observed in the vascular stria, spiral ligament and spiral prominence. The cochlear nerve showed the next highest level of uptake, while the organ of Corti and the spiral ganglion showed low levels. The functional significance of the results was briefly discussed.

Localization of lysozyme in the frozen section of nasal mucosa by protein A-gold technique.

The location of lysozyme was studied in frozen sections of the nasal gland by protein A-gold technique, modified for the frozen section. Serous glands were enriched with lysozyme. Within the glands, the apical portion of the serous cells was most heavily stained. The epithelial layer was virtually unstained for lysozyme. The result was essentially the same as that obtained by the peroxidase-antiperoxidase method. Electronmicroscopic examination of the stained sections supports the possible application of this method for pre-embedding immunostaining, although further improvement of the technique is necessary.

Experimental immune complex otitis media: localization of IgG by protein A-gold technique.

Immune complex (IC) was made of bovine serum albumin (BSA) and anti-BSA guinea pig serum, and instilled into the tubotympanic cavity of untreated guinea pigs. The location of IgG was determined by using the protein A-gold technique to trace IC in the tubotympanic mucosa of the otitis media with effusion (OME) caused by the instillation. IgG was found on the effusion, degenerated ciliated cells and on granules of goblet cells. There was no evidence of intense accumulation of IgG on the basement membrane of the mucosal epithelium or the capillaries. It is likely that IC activates complements in the tubotympanic cavity to cause OME.

Duane's syndrome associated with crocodile tear and ear malformation. A case of stapes anomaly and its reconstruction.

Duane's syndrome is a congenital eye movement disorder characterized by marked limitation or absence of abduction, variable limitation of adduction, and narrowing of the palpebral fissure with retraction of the globe on attempted adduction. We treated a patient with bilateral Duane's syndrome associated with crocodile tear and external, middle, and inner ear anomalies. Exploratory tympanotomy showed an absence of the oval window and a rudimentary superior structure of the stapes that was connected to the abbreviated incudal lenticular process with connective tissue. A small hole was drilled into the vestibulum and the ossicular chain was reconstructed by inserting a Teflon wire piston to minimize the air-bone gap. Pertinent literature is reviewed, and the pathogenesis of Duane's syndrome and the aspect of concomitant anomalies are discussed.

The localization of motoneurons innervating the canine pharyngeal constrictor muscles in the posterior larynx by the fluorescent double-labeling technique.

The localization of motoneurons innervating the canine pharyngeal constrictor muscles in the posterior larynx, i.e., the hyopharyngeal muscle (HP), the thyropharyngeal muscle (TP) and the cricopharyngeal muscle (CP), was investigated by the fluorescent labeling technique. Labeled cells were found in the rostral portion of the ipsilateral nucleus ambiguus. The rostral ends of these cell columns were located at nearly the same level. The caudal ends of the HP cell column and the TP cell column were also located at nearly the same level. However, the CP cell column was elongated a little more caudally. The HP cells and the TP cells were intermingled in the dorsal part of the nucleus. The CP cells were mainly located in the ventral part of the nucleus. No double-labeled cells were detected.

Vocal rehabilitation by tracheogastric shunt method after pharyngolaryngoesophagectomy for malignancy.

After pharyngolaryngoesophagectomy had been performed to remove a malignant tumor, a shunt was created between the elevated gastric tube and the trachea for vocal rehabilitation in two patients. By covering the tracheostoma, the patients were able to produce a substituted voice without any reflux. However, strange vibrations of the gastric tube slightly disturbed the intelligibility of the shunt voice.

Localization of triphosphoinositide in the cochlea. An electronmicroscopic immunocytochemical study.

Triphosphoinositide (TPI) has been demonstrated to be a receptor for aminoglycosides in the cochlea and may regulate ionic permeability by its binding with Ca++. This phospholipid was localized by a protein A-gold technique in the cochlea at the electronmicroscopic level. TPI was prepared by a neomycin column and antibodies to it were raised in rabbits. The antibody used in this study reacted virtually only to TPI among the tested lipids. TPI was localized mainly at stereocilia, cuticular plates, head plates of Deiters' cells, plasma membrane, and mitochondria of various cells in the organ of Corti. In the vascular stria, TPI was found mainly at the plasma membrane of basal infoldings of the marginal cells. Possible physiological and pathophysiological roles of TPI in the cochlea are briefly discussed.

Freeze-replica observations of guinea pig middle ear muscles.

The histological characteristics of the middle ear muscles, Musculus tensor tympani and M. Stapedius, of the guinea pig were studied using freeze-fracturing technique. No gap junction was observed but the tight junction was found in the tensor tympani muscle, possibly between muscle cells. No distinct difference was found between the size distributions of the apertures and caveolae on the fracture face of the sarcolemma of the two kinds of muscles.

Studies of the substructures of the lysozyme-rich secretory granule of the serous cell in the human nasal gland.

Using a protein A-gold immunohistochemical technique, the lysozyme rich core and the lysozyme free peripheral rim were differentiated in the secretory granule of the serous cell in the human nasal mucosa, under the electronmicroscope. The shperical lysozyme rich core, which had been excreted into the gland lumen, was also wrapped with the peripheral rim. This finding suggests that the peripheral rim defends the integrity of the core and inhibits the release of lysozyme from the core.

The effect of tonsillar T cell subpopulations in antigen-specific lymphocyte proliferative response. Evidence of T cell suppression.

The presence of specific T cell subsets in tonsils, during in vitro generation of peripheral blood lymphocyte proliferative response was investigated. Tonsil T cells were selected for their activity with the monoclonal antibodies OKT4 and OKT8. The suppressor cell assay revealed the effects of tonsillar T cell subpopulations identified as OKT4- and OKT8-. The OKT8- had no inhibitory effects on proliferation of peripheral lymphocytes by Dermatophagoides farinae antigen in vitro.

A multi-centre, double-blind study of serrapeptase versus placebo in post-antrotomy buccal swelling.

A multi-centre, double-blind, placebo-controlled trial was carried out to investigate the clinical efficacy of the anti-inflammatory enzyme serrapeptase in a total of 174 patients who underwent Caldwell-Luc antrotomy for chronic empyema. Eighty-eight patients received 10 mg serrapeptase 3 times on the day before operation, once on the night of the operation and 3 times daily for 5 days after operation; the other 86 received placebo. Changes in buccal swelling after operation were observed as a parameter of the response to treatment. The degree of swelling in the serrapeptase-treated patients was significantly less than that in the placebo-treated patients at every point of observation after operation up to the 5th day (p less than 0.01 to p less than 0.05). Maximal swelling throughout all the post-operative points of observation was also significantly smaller in size in the serrapeptase-treated group than in the placebo-treated group. No side-effects were reported.

Effect of anti-interferon serum of influenza virus infection in mice.

Mice were infected by an aerosol of influenza virus Type A (0.5 LD50) and subsequently treated with 4 intranasal instillations of anti-interferon antiserum over a period of 72 h. All the mice treated with antiserum died within 7 days post-infection, whilst the mice in the control groups survived. In mice that did not receive the antibody, virus titers in the lung peaked on day 3 and then decreased again. Also, interferon was detectable both in lung homogenates and serum. In mice treated with antiserum, no interferon was detectable and the virus concentrations in the lung increased until death. These results suggest that interferon produced in the respiratory tract plays an important role in the early stages of influenza virus infection.

[Clinical studies of cefotiam in otorhinolaryngologic infections].

Cefotiam (CTM) was intravenously given to 12 patients with infectious diseases in the field of otorhinolaryngology including 10 cases with chronic suppurative otitis media. Daily doses of CTM were 1 to 4 g. Clinical responses were excellent in 3 patients, good in 5, fair in 1 and poor in 3. The rate of overall clinical effectiveness was 67%. Staphylococcus species were most frequently isolated from the patients and all of them were eradicated by the CTM treatment. No side effects and no abnormal laboratory findings relating to the drug administration were observed.

[Clinical study of cefsulodin on infectious diseases in otorhinolaryngological field].

The clinical effect of cefsulodin (CFS) on Pseudomonas species infections in otorhinolaryngologic field was studied in 10 patients. The overall clinical effectiveness was 80% (excellent 4, good 4, poor 2). The bacteriological response of CFS against Pseudomonas species was evaluated from 11 strains. Eradication rate of Pseudomonas species was 82%. In 1 case, body temperature was rised after injection. No abnormal laboratory findings, relating to the drug, were observed after treatment.

Occurrence of non-ototoxic antibiotic (cephazolin) in the inner ear.

Cephazolin, a non-ototoxic antibiotic, and kanamycin, an ototoxic antibiotic, were administered to the guinea pig systemically and their tissue concentration was measured. Not only kanamycin but also cephazolin did occur in the inner ear as well as in the kidney and other tissues. Therefore, it is most likely that the accumulation of an antibiotic inside the inner ear forms only a sine qua non for its ototoxicity. The mechanism of the ototoxicity should exist in the antibiotic-vulnerable cells (hair cells) themselves.
