Severe impairment of dietary folate utilization in the elderly.

Absorption of folates, vitamin B6, pantothenate, and riboflavin from a natural food source (yeast) and from synthetic folylmonoglutamate was studied in 24 elderly subjects (age range, 73 to 101 years) and in 12 healthy younger subjects (age range, 24 to 42 years). All subjects absorbed riboflavin, vitamin B6, and pantothenate from yeast. Ingested folypolyglutamates (the preponderant folates in yeast) proved to be a very poor source of folates for the elderly subjects, whereas synthetic folylmonoglutamate served as a good source. In the younger subjects, yeast was a significant folate source. It is suggested that the folate deficits so common in the elderly are caused by impaired ability to obtain folate from ingested foods. The resultant folate deficit may induce changes in the epithelial structure and enzyme secretion of the small bowel, which may further exacerbate folate malabsorption.

Biopterin content of human neuroblastoma cells in culture.

The biopterin (Crithidia activity) content of several cell lines of human neuroblastoma was quantified. Adrenergic cell lines contained markedly higher levels than those found in cholinergic neuroblastoma cells, fibroblast-like cells and glioblastoma cells in culture.

Vitamin levels in low-birth-weight newborn infants and their mothers.

The cord blood of 50 normal-birth-weight neonates (more than 2,500 grams) and 50 low-birth-weight neonates (less than 2,500 grams) and the respective mother's blood were analyzed for folate, vitamin B6, riboflavin, nicotinate, pantothenate, thiamin, biotin, vitamin B12, vitamin A, and beta-carotene concentrations at parturition. No mothers had received supplemental vitamin intake. Except for vitamin A and beta-carotene, maternal vitamin levels were lower than those of neonates in all instances. Vitamin levels in the blood of low-birth-weight neonates were the same of those of normal-birth-weight infants except for significantly lower folate, vitamin B12, and pantothenate levels.

The cell content and secretion of water-soluble vitamins by several freshwater algae.

Three green algae, Chlamydomonas reinhardii, Chlorella vulgaris and Scenedesmus obliquus, and one blue-green alga, Anabaena cyclindrica, were grown in chemically defined media. All the algae examined contained folates, beta-carotene and vitamins C and E; several of the B-vitamins and vitamin A were found in varying amounts in some but not in all the algae examined. All the green algae secreted significant amounts of folate and biotin and all but Scenedesmus secreted pantothenate into their growth medium; Anabaena secreted folate and pantothenate.

A double-blind comparative study of 150 mg flurbiprofen daily and 75 mg indomethacin daily in the treatment of osteoarthrosis of the hip joint.

Thirty patients with osteoarthrosis of the hip were studied in a double-blind crossover comparison of 150 mg flurbiprofen daily and 75 mg indomethacin daily, each drug being given for 2 weeks separated by a wash-out period of 1 week on placebo. Statistical analysis of the results from 26 patients with complete records showed that both drugs produced statistically significant improvements over baseline after 1 and 2 weeks in assessments of severity of pain, night pain, duration of morning stiffness, and intermalleolar straddle. No statistical differences, however, were found between the two drugs. There were no reports of side-effects with flurbiprofen, but 6 reports from 3 patients whilst receiving indomethacin.

[DNA--"rejoining" in lymphycytes of patients with a minifestation of rheumatoid arthritis in older age (author's transl)].

Relations between the time of manifestation of the disease and the course of the rheumatoid arthritis were found from the clinic frequently. Patients with later first manifestation of rheumatoid arthritis show clear differences against patients with earlier manifestation of the disease at which the late first manifestation a high activity of the process of the disease exhibit. In males first manifestation of rheumatoid arthritis in middle age of 54,5 years was observed, while in a first manifestation of rheumatoid arthritis in middle age of 47 years a normal course of the disease was found. The difference between the both courses was proved as significantly. In former investigations we could prove that in patients with rheumatoid arthritis the 3H-Thymidin-build in the course of not programmed DNA-synthesis in the DNA of lymphocytes after damage with ionizing was retarded. The rate of not programmed DNA synthesis seems to be in old persons equal. The ligase reaction is possibly diminished because the number of DNA-strand breakes with the age is strong increasing.

Potentiation of ethanol fatty liver in rats by chronic administration of nicotinic acid.

Two groups of rats were fed isocalorically on alcohol and control semi-liquid diet for 28 days; two other groups had the same diets except for supplementation with nicotinic acid at 50 mg/100 ml of diet, Blood ethanol levels were unaffected by nicotinic acid administration, even though nicotinic acid was well absorbed and stored in the liver. Lipid analyses of liver and plasma after 28 days revealed that nicotinic acid, per se, stimulated fatty infiltration of the liver and this effect was potentiated when given in conjunction with ethanol.

[Biochemistry of benign-symmetrical lipomatosis (adenolipomatosis Launois-Bensaude, Madelung's disease)].

1. Total lipids, total cholesterol, cholesterol esters, phospholipids, triglycerides and free fatty acids as well as the fatty acids profiles of the different lipid classes were determined in serum, lipomatous and normal adipose tissue. Triglycerides were elevated in patient L's serum. The distribution of serum lipoproteins in this patient's serum showed a type IV according to Fredrickson. All other lipid parameters were within the normal range. Palmitoleic acid was increased nearly in all lipid fractions of the patients' sera as well as in the lipids of lipomatous subcutaneous adipose tissue. 2. The lipomatous adipose tissues of the patients showing no histological abnormalities revealed higher levels of cyclic AMP than normal subcutaneous adipose tissue. 3. Serum uric acid was normal (patient E.), between the normal and pathological range (patient L.) and elevated (patient W.). Urinary uric acid excretion was increased in all three patients. 4. 14C-glycine was overincorporated into urinary uric acid in all three patients. 5. Adenine phosphoribosyltransferase activities in the hemolysates were within the normal range. A decrease of hypoxanthine-guanine phosphoribosyltransferase activities could be demonstrated in two patients' (e., w.) erythrocytes. Erythrocyte phosphoribosylpyrophate synthetase activity was slightly increased in patient L.'s and twice the normal value in patient W.'s erythrocytes.

Effects of a single intoxicating dose of ethanol on the vitamin profile of organelles in rat liver and brain.

The effects of a single, oral, intoxicating dose of ethanol (EtOH, 9 g/kg body weight) on the distribution of water- and fat-soluble vitamins in various subcellular fractions of eight rat livers and eight rat brains was surveyed. Results were compared to eight other rats receiving a similar dose of saline (controls). Both organs showed redistribution of vitamins among the various organelles after EtOH was given. In saline-treated animals the nucleus and cytosol of the liver contained the highest proportion of all vitamins and the microsomes and lysosomes contained the least. As compared to controls, EtHO ingestion generally caused the mitochondria and microsomes to accumulate vitamins, while vitamins in the lysosomes were decreased. A different pattern was seen in the brain after EtOH administration. Except for thiamin and niacin, the whole brain vitamins were higher than those in controls; they were primarily concentrated in the brain nucleus and mitochondria. It was concluded that changes in the organelle vitamin content of the liver were the result of EtOH injury. The simultaneous attempt to repair the injury was manifested by the by the sequestration of vitamins in the organelles of EtOH-treated animals as compared to controls.

The effect of folic acid on gingival health.

On days 0 and 30 of a double blind study, two groups of 15 subjects each were evaluated using a plaque index, a gingival index, a gingival exudate flow and fasting plasma folic acid levels. Group I received 2 mg of folic acid twice daily for 30 days while Group II received a placebo. Results of the study seem to indicate that folic acid supplemented to the diet may increase the resistance of the gingiva to local irritants and thus lead to a reduction in inflammation.

Absorption, utilization and clinical effectiveness of allithiamines compared to water-soluble thiamines.

Oral administration of lipid-soluble allithiamines [thiamine propyl disulfide (TPD) and thiamine tetrahydrofurfuryl disulfide (TTHF)] rapidly increased thiamine activity in whole blood, red blood cells, cerebrospinal fluid, and urine in normal and thiamine-deficient subjects. These thiamine congeners also restored red blood cell transketolase to normal in alcoholics with thiamine deficiency. Such repletion equaled that produced by parenteral, water-soluble thiamine hydrochloride (THCl) or thiamine pyrophosphate (TPP). Oral administration of water-soluble thiamines (THCl, TPP) neither elevated thiamine activity in biological fluids nor restored transketolase activity to normal in alcoholics with thiamine deficiency presumably due to their rate-limited intestinal transport. Oral administration of TPD eliminated lateral rectus palsy in patients with Wernicke's encephalopathy. Orally administered allithiamine vitamers are therefore recommended for prophylaxis and treatment of thiamine deficits because while having essentially the same biological properties as parenterally administered water-soluble thiamines they have not produced any untoward effects after long-term administration and are far more efficiently utilized.
