Regional cerebral blood flow in schizophrenic disorders.

Regional cerebral blood flow was measured using a 133Xe inhalation technique in 16 schizophrenic patients and 20 healthy volunteers. The bilateral frontal blood flows in the patient groups were significantly lower than in the control group. In addition, the patient group having auditory hallucination showed a significantly increased blood flow predominantly in the left temporal region. On the other hand, the patient group without auditory hallucination showed a slightly increased flow in the right temporal region. These findings indicate that there are a hypofrontal activity and also a hypertemporoparietal activity in schizophrenics.

[Regional cerebral blood flow in cerebrovascular disease studied using N-isopropyl-p-(123I) iodoamphetamine].

Twenty-one patients with cerebrovascular disease (5 with hemorrhage, 10 with infarction, 2 with TIA, 4 with motor disturbance of unknown cause) were studied using N-isopropyl-p-(123I)iodoamphetamine (123I-IMP) and single photon emission CT. In 3 of 5 cases with cerebral hemorrhage, perfusion defects were shown in and around the region of hematomas, furthermore, one of the cases with internal capsular hematoma, the perfusion defect extended to the cortical area corresponding to the neurological pathway. In one case with MCA infarction, the perfusion defect was greater and clearer than the low density area on X-ray computed tomography (CT). These results revealed the 123I-IMP study provides physiological information in contrast with X-ray CT which provides anatomical information. In 3 of 9 cases with multiple small deep hemispheric infarctions on X-ray CT, the perfusion to the basal ganglia was suspected to be decreased on 123I-IMP images. However, this visual findings was not definitive and, in fact, the diseased side was not always consistent with the clinical findings. For quantitative analysis, symmetrical regions of interest (ROIs) were constructed both basal ganglia and the ratio of average counts over the ROI to those over the whole slice was calculated. In the small infarction group, the mean +/- S.D. of the values was 0.89 +/- 0.09 in right and 0.89 +/- 0.08 in left. Although the values were not significantly different from those of normal subjects (0.99 +/- 0.02 in right, 0.97 +/- 0.03 in left), they distributed in the range less than normal in 5 of 9 cases. This method was thought to be useful and practical to evaluate the cerebral blood flow in basal ganglia of patients with deep hemispheric infarction.

Establishment and characterization of the human cholangiocarcinoma cell line HChol-Y1 in a serum-free, chemically defined medium.

A human cholangiocarcinoma cell line, designated as HChol-Y1, was established in a protein-free, chemically defined medium after a very short period of primary culture in 0.1% fetal bovine serum (FBS)-containing medium. The cell line has been propagated in this medium for 2 years. The cells grew as a monolayer and the doubling time was about 52 hours. Addition of FBS did not stimulate cell growth (population-doubling time = 50 hr) or increase saturation density. The cells grown in a protein-free medium secreted small amounts of carcinoembryonic antigen (CEA) and large amounts of carbohydrate antigen (CA) 19/9 (CEA: 12.5 +/- 2.1 ng/10(6) cells/48 hr; CA 19/9: 760 +/- 52 IU/10(6) cells/48 hr); these tumor markers were immunohistochemically demonstrated in HChol-Y1 cells. Addition of FBS slightly stimulated the production of CEA and CA 19/9. The HChol-Y1 cell line was xenotransplantable in athymic nude mice and increased the serum CEA and CA 19/9 levels in the tumor-bearing nude mice. For determination as to whether a human carcinoma cell line can proliferate and secrete CEA and CA 19/9 in synthetic medium without any protein supplements, the cells were cultivated long term (2 yr) in a protein-free, chemically defined medium. When this method of cultivation is used, it is easy to purify these substances from spent medium, because contaminating antigens such as FBS or other substances usually added to cultures are absent.

Multiple primary cancers, with special reference to occupational exposure.

Multiple primary cancer (MPC) relating to occupational exposure was studied. The study consisted of a literature survey, survey of on-going research and our project on the frequency and nature of MPC in an occupational cohort of Japanese dyestuff workers followed from 1962 through 1983. From the review of the literature and on-going research there is little information on MPC and existing findings are mostly incomplete. However, a few occupational agents known to be carcinogenic to more than two organs, i.e. arsenic or radium, have been reported to cause MPC. In our follow-up of occupational bladder cancer cases, increased MPCs compared with a matched control group of nonoccupational cases were observed, but further investigation is necessary for a final conclusion. Finally, epidemiologic problems inherent to the study of MPC, including occupational studies, are discussed.

Estimation of the prevalence of occult cancer and its application to the epidemiology of multiple primary cancer.

This paper focused on methodological issues in the epidemiology of multiple primary cancer, especially on methods to evaluate the frequency of a second cancer. The incidence density is the most widely used measure in cancer epidemiology. However, the measure of incidence might be biased to some extent, because it might be affected by extraneous factors which are related to the detection of the second cancer. A better alternative is the measure of the prevalence of occult cancer. The authors have formulated the functional relationship between the prevalence of occult cancer and some variables which can be obtained from ordinary epidemiologic observations, such as the incidence density, the growth function, and the distribution of the diameters of the cancers which were clinically diagnosed. The formula was then applied to an epidemiologic study of multiple primary cancer in the digestive system. All the patients with cancer of the digestive system except the stomach treated in the National Cancer Center Hospital between 1962 and 1981 constituted the study group. Among 1,561 male and 714 female patients, gastric cancers were found as the second primary cancer in nine males and one female within 2 months after the first cancers were detected. All the first cancers in these 10 cases were esophageal cancers. The expected numbers of second gastric cancers were calculated as 4.55 in males and 1.20 in females by using the formula. The observed number exceeded significantly the expected number in males (p value = 0.042), which suggested that esophageal and gastric cancers might share some risk factor.

Characterization of gamma-GTP in a human pancreatic cancer cell line.

Gamma-glutamyl transpeptidase (gamma-GTP) was partially purified from human normal pancreas, pancreatic carcinoma and a human pancreatic cancer cell line, HPC-Y1. The characteristics of gamma-GTP from all three samples appeared to be identical. The estimated molecular weight of samples solubilized with Triton X-100 was 210K and that of bromelain-solubilized gamma-GTP was 110K. On polyacrylamide gel electrophoresis, the main band in both the Triton- and the bromelain-solubilized gamma-GTP of these samples had similar electrophoretic mobility. The percentages binding with concanavalin A were 52%-62%, while on isoelectric focusing the pI values were 3.40-3.45. It was concluded that the heterogeneity of the gamma-GTP isoenzyme could not be identified by either gel filtration or polyacrylamide gel electrophoresis, and it is necessary to investigate the modification of carbohydrate structure on tumor.
