An autopsy case of adult T cell leukemia associated with hypercalcemia and hyperplastic parathyroid glands is reported. A 51-year-old man complained of systemic lymphadenopathy and hepato-splenomegaly; a diagnosis of adult T cell leukemia was made based on clinico-pathological findings. The pathogenesis of hypercalcemia in adult T cell leukemia is discussed. However, cases associated with hyperplasia of the parathyroid gland are very rare. We emphasize the role of the parathyroid gland in the pathogenesis of hypercalcemia in adult T cell leukemia.
Local cerebral blood flow (LCBF), local cerebral glucose utilization (LCGU) and capillary permeability were studied in rats with tumor-induced brain edema. Moreover, effects of methylprednisolone on these physiological parameters were studied to analyse a possible role of steroid on treatment of peritumoral brain edema. A cubic millimeter pellet of Walker 256 tumor was transplanted to the left sensorimotor cortex of the rat brain. Animals were randomly divided into two groups. One group was treated with methylprednisolone (15 mg/kg/day) for 5 days starting at 5 days after tumor inoculation, and the other group received no treatment. These rats were used for autoradiographic study at 10 days after tumor inoculation. Local CBF, LCGU and capillary permeability were measured with 14C-iodoantipyrine, 14C-deoxyglucose and 14C-alphaaminoisobutyric acid, respectively. In the untreated group, LCBF and LCGU were widely depressed in the cortex and deep structures of the hemisphere ipsilateral to the tumor. Of the methylprednisolone treated animals LCBF and LCGU were significantly better than that of untreated animals. Capillary permeability of the untreated animals were highly increased in the viable part of the tumor. Some increase was also noted in the peripheral edge of the tumor and adjacent brain. In the methylprednisolone treated groups, capillary permeability was significantly lower than that in untreated group. The data suggest that methylprednisolone decreases capillary permeability in the viable part of the tumor.(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of a new azole antimycotic, 1-[(4-biphenylyl)-phenylmethyl]-1H-imidazole (bifonazole, Bay h 4502, Mycospor) on the morphology of hyphae of Trichophyton mentagrophytes grown in a liquid medium was studied by high-resolution scanning electron microscopy. With bifonazole at concentrations ranging from 1 to 10 ng/ml that were much lower than the MIC value of the drug for this test strain (630 ng/ml), the hyphal growth was significantly inhibited and several morphological changes of the hyphae were produced. They were characterized by: (1) development of wavy or curled hyphae, (2) occasional formation of swollen cells often arranging in chain; (3) partial exfoliation of hyphal walls; and (4) excretion of fibrillar materials. These morphological changes of the hyphae became more prominent with increasing drug concentrations up to 200 ng/ml, and many hyphae were collapsed and distorted. These results strongly suggest that subinhibitory concentrations of bifonazole profoundly affected the normal growth and induced degenerative changes of the hyphae of T. mentagrophytes probably by damaging the cell membrane.
A simple high performance liquid chromatographic (HPLC) method has been developed for determining malondialdehyde (MDA) in vegetable oils. MDA was reacted with dansyl hydrazine in an acidic medium, and the product, 1-dansyl-pyrazole, was determined by HPLC, using a Zorbax sil column with mixed mobile phase of n-hexane-methylene chloride. MDA can be determined as 1-dansyl-pyrazole by fluorometric detection at a level of 0.01 ppm in vegetable oils.

Uterine estrogen receptors (ER) of the rat during pregnancy and postpartal lactation were assayed. Both the cytoplasmic and nuclear ER were determined by the controlled pore glass (CPG) method as previously described in detail. The cytoplasmic ER level slightly decreased as pregnancy advanced and then increased remarkably during postpartal lactation, with a tendency to decrease at 5 weeks of lactation when the estrous cycle could have recurred despite the nursing behavior of the rats. On the other hand, no remarkable difference in nuclear ER was noted during pregnancy and lactation. A high concentration of plasma progesterone could have reduced the translocation of the cytoplasmic ER to the nucleus and the replenishment of the cytoplasmic ER, despite a high concentration of plasma estrogen during pregnancy. In order to keep an environment of a high plasma concentration of estrogen similar to that of pregnancy, a long-acting estrogen (estradiol dipropionate) was injected in to the proestrous rats. The cytoplasmic ER rapidly translocated to the nucleus, followed by a prolonged maintenance of a low cytoplasmic and a high nuclear ER level until 7 days, with a gradual replenishment of the cytoplasmic Er and a gradual decline of the nuclear ER thereafter. A translocation of the cytoplasmic Er to the nucleus, similarly seen in the hysterectonized-adult rats treated with estradiol, was noted in the rats injected with estradiol at 3 weeks of lactation. In general, low estrogen and high prolactin may be held in plasma during lactation. These results indicate that the fluctuation of uterine ER simply depends on the plasma level of estrogen during pregnancy and postpartal lactation in rats.
The activities and kinetic properties of argininosuccinate synthetase in cultured skin fibroblasts of three citrullinemic patients with qualitative or quantitative abnormalities of hepatic argininosuccinate synthetase were examined. The cultured skin fibroblasts of a citrullinemic patient with a low hepatic argininosuccinate synthetase activity showed low activity and abnormal kinetics similar to those of the hepatic enzyme. This suggests a similar genetic origin of the fibroblast argininosuccinate synthetase as of the hepatic enzyme. However, we found normal argininosuccinate synthetase activity in cultured fibroblasts of two citrullinemic patients who had a low hepatic enzyme activity resulting from decrease in the amount of enzyme protein but with normal kinetic properties. This suggests that part of the quantitative abnormality of hepatic argininosuccinate synthetase of citrullinemic patients may be due to an abnormal organ-specific gene expression in the liver.
A specific, precise and sensitive double-antibody radioimmunoassay for the measurement of human apolipoprotein E (apo E) was developed. Antibody specific to apo E was raised in a rabbit. This antibody did not crossreact with plasma low density lipoproteins (d = 1.030-1.063) and apo-proteins such as pao CII, CIII, AI and AII. Mean plasma level with SD of apo E in 29 normalipidemic subjects was 9.5 +/- 1.8 mg/100 ml. Plasma apo E concentrations in 25 hypertriglyceridemic patients with plasma triglyceride concentrations ranging from 169 to 1190/mg/100 ml correlated positively with plasma triglyceride concentrations (r = 0.84, p less than 0.001). Among subjects with hypertriglyceridemia, there was a negative correlation between the apo E per mg triglyceride ratio and plasma triglyceride concentrations (r = 0.49, p less than 0.02). Plasma apo E levels did not show any correlation with plasma cholesterol concentrations. These results indicate that plasma apo E is closely associated with plasma triglyceride levels, which suggests a possible connection of this apoprotein with the metabolism of plasma triglyceride.
Triacylglycerol lipase activities in adrenals and testes of rats were measured and the properties of the enzymes were characterized. The enzymes were extracted from acetone-ether dried powder of the tissues and the crude extracts were applied to a heparin-Sepharose affinity column. The enzymes bound to the column were then eluted at different ionic strengths. Two lipases could be detected in rat adrenals: one was inactivated by the antiserum raised against hepatic triacylglycerol lipase and the other had the properties of lipoprotein lipase. In testes, only lipoprotein lipase could be detected. The results are discussed in connection with the possible functions of these two lipases in organs utilizing lipoprotein-cholesterol for steroid hormone synthesis.
