[Serum apolipoproteins A-1 and B in young males with a history of myocardial infarction].

The content of the protein-lipid components of lipoproteins of individual classes was studied in 87 coronary heart disease patients surviving myocardial infarction under 40 years of age. All the patients lived in Sverdlovsk. The control group was made up of 49 age-matched males without any symptoms of CHD. CHD patients aged 30-39 years showed a number of differences as compared to healthy men; these included elevated plasma levels of cholesterol, triglycerides and low-density lipoprotein cholesterol as well as an increase in the apoprotein B levels and the apo-B/apo-A1 ratio. In a group of CHD patients aged 20-29 years enhanced plasma concentrations of apo-B and an increase in the apo-B/apo-A1 ratio, the latter approaching 1 were the only change in the lipoprotein system as against control.

[Disruption of the cholesterol-transport function of lipoproteins in the blood plasma of patients with ischemic heart disease].

A ratio cholesterol esters/apo B in very low and low density lipoproteins (VLDL and LDL), characterizing their cholesterol-transport function, as well as a ratio cholesterol esters/apo A-I in high density lipoproteins (HDL) HDL2 and HDL3, demonstrating the loading of cholesterol per unit of their main protein, were studied in 16 patients with ischemic heart disease accompanied by coronary atherosclerosis documented by angiography and in 36 healthy men. In patients with coronary atherosclerosis and low level of HDL cholesterol, particles of atherogenic lipoproteins were overloaded with cholesterol esters, while the ratio cholesterol esters/apo A-I was decreased in HDL2 and HDL3 as compared with controls. These alterations might be related to an increase in efficiency of cholesterol esters transport from HDL into VLDL and LDL as a result of dissimilar transformations in chemical and structural characteristics of surface monolayer of VLDL and HDL particles studied earlier.

[Apolipoproteins A-1 and B of the blood plasma and apolipoproteins A-1 of 2 high-density lipoprotein subclasses in ischemic heart disease patients with various lipoprotein spectra].

Apolipoproteins (apo A-1 and apo B) were measured by rocket immunoelectrophoresis in 53 patients 40-59 years of age with coronary heart disease. Patients with hyperlipoproteinemia and low HDL cholesterol level as well as those with normal lipid levels were compared with age-matched healthy controls. Apo A-1 blood plasma level reduction was found in the former group. The reduction is shown to be due to the decrease of apo A-1 composing HDL3 in type IIa and IIb hyperlipoproteinemia and in normal lipoprotein level. HDL2 apo A-1 reduction is also observed in type IV hyperlipoproteinemia. The rise of apo B level was noted in the blood plasma of patients with coronary heart disease, the changes being mainly connected with the increase in the number of apo B-containing particles. It is shown, that patients with coronary heart disease have an elevated apo B/apo A-1 ratio as compared to the control group, with its value exceeding 1 whatever the lipoprotein level. Changes in the quantitative composition of plasma apolipoproteins are considered to be a factor, ensuring prevalence of cholesterol flow to peripheral tissues over its acceptance and escape.

[Changes in blood plasma lipid and apolipoprotein content in the course of acute myocardial infarction].

Patterns of changes in blood plasma lipids and apolipoproteins were examined in 16 patients with acute large-focal myocardial infarction. Six of those were investigated during unstable angina pectoris. Lipoproteins responsible for cholesterol transport to tissues (apo-beta-containing lipoproteins) and its outflow (apo-A-I-containing lipoproteins) were shown to undergo a variety of metabolic changes in the course of myocardial infarction. Possible prognostic implications of patterns of post-infarction changes in lipoprotein and apolipoprotein spectrum are discussed with respect to predicting further development of atherosclerosis.

Flow cytofluorometric study of lipoprotein interactions with cultured endothelial cells.

Accumulation of rhodamine B isothyocyanate-conjugated low density lipoproteins (R-LDL) in cultured endothelial cells from human umbilical cord was studied with a fluorescence activated cell sorter. R-LDL uptake was blocked at 0 degrees C, inhibited by addition of excess of nonlabeled low density lipoprotein, high density lipoprotein-2 or high density lipoprotein-3. High density lipoprotein-2 was about twice as effective in inhibition of R-LDL uptake as high density lipoprotein-3. Endothelial cells that formed a contact-inhibited monolayer lost the ability to incorporate R-LDL via a receptor-mediated pathway. Using R-LDL, it was possible to distinguish cells with different levels of R-LDL incorporation.

The peculiarities of lipid and protein components of the high- and low-density lipoproteins in patients suffering from coronary atherosclerosis with different number of affected coronary arteries.

In 81 patients with coronary atherosclerosis, with different number of affected coronary arteries determined by selective coronarography, the blood plasma levels of low-density (LDLP) and high-density (HDLP) lipoproteins were determined, and their lipid and protein components were analyzed. In patients with two and three arteries affected, there were found -- in comparison with the control group (patients with neurocirculatory asthenia) and patients with one coronary artery affected: higher total cholesterol, triglycerides, cholesterol of very-low-density lipoproteins (VLDLP), LDLP cholesterol, and moderate value of the VLDLP+LDLP cholesterol/HDLP cholesterol ratio(s.c. atherogenicity coefficient). The HDLP cholesterol level was decreased in all groups of patients with coronary atherosclerosis. Along with the increasing number of arteries affected the percentage of esterified cholesterol in the HDLP decreased, free cholesterol in the HDLP and esterified cholesterol in the LDLP rose.

[Change in the properties of model membranes interacting with atherogenic and "antiatherogenic" classes of lipoproteins with varying physicochemical characteristics].

The work was undertaken for the purpose of studying the interaction of human blood plasma lipoproteins (LP) with bi-layer lipid membranes (BLM), which is regarded to be a model of nonreceptive interaction of LP with the cell membranes. The surface LP of very low density (LPVLD) layer is much "denser" in hyperalpha-lipoproteinemia than in hypoalpha-lipoproteinemia. It was found that atherogenic LP classes induce an increase in BLM conductivity in much lesser concentration than the subclasses of antiatherogenic LP of high density. LPVLD of individuals with their increased content in plasma increase the electrical capacity of BLM to a lesser extent than LPVLD of healthy individuals. In most experiments with LPVLD of individuals with their high level. BLM resilience diminished, whereas in experiments with LPVLD of persons with a normal LP spectrum, BLM resistance was found to be increased. It is thus shown that despite the absence of protein receptors in the BLM some of their physico-chemical characteristics change on interaction with blood plasma LP.

[High-density lipoprotein subfractions and sex hormones in the blood plasma of men 40 to 59].

Examination of 50 males aged from 40 to 59 years showed that an increase or decrease in the level of cholesterol of high-density lipoproteins (HDLP) occurs mostly at the expense of cholesterol of the HDLP2 subfraction. Changes in the concentration of sex hormones in the blood plasma are attended by a change in the number of HDLP2 particles, evidence of which was a change in the content of HDLP2 cholesterol and phospholipids with the level of HDLP3 cholesterol and phospholipids remaining unaltered. A higher content is attended by a rise in the level of HDLP2 cholesterol and HDLP2 phospholipids. A negative correlation between the level of estradiol and that of triglycerides in blood plasma (r=-0.61; p less than 0.01) and a positive correlation between the content of estradiol and the level of HDLP2 cholesterol and phospholipids (r=+0.34; p less than 0.05 and r=+0.37; p less than 0.05, respectively were revealed in males with estradiol concentration of 1--8 ng/dl. There was no correlation between the above-mentioned values when the concentration of estradiol in blood plasma was higher (8--16 ng/dl). In hypoalphalipoproteinemia and hypertriglyceridemia, the relation in very low density lipoproteins of apoprotein SH, which inhibits lipoprotein lipase, and apoprotein SP, which activates this enzyme, was higher than normal while the relation of these apoproteins in HDLP was lower.

[Characteristics of cholesterol transport by various classes of lipoproteins and their apolipoproteins in hypo- and hyperalphalipoproteinemia].

The content in the blood plasma of the main apolipoproteins of lipoproteins of very low and low density apo-B and tetramethylurea-soluble apo-LP as well as lipoproteins of high density apo-A and the relation to apo-LP cholesterol of these classes of lipoproteins were studied in 34 males with different levels of alpha-LP cholesterol. It is shown that in hypoalphacholesterolemia the plasma apo-A content is reduced with the cholesterol/apo-A ratio being less than in normoalphacholesterolemia; at the same time, the content in the plasma of total triglycerides, cholesterol, and protein of very low density lipoproteins is increased and cholesterol saturation calculated for protein of very low and low density lipoproteins does not change. In hyperalphacholesterolemia the plasma apo-A content is increased both in the absolute amount and in relation to apo-B and TMU-soluble proteins; the content of cholesterol on apo-A in high density lipoproteins is the same as under normal conditions and higher than in hypoalphacholesterolemia. The results obtained are discussed in association with the data in the literature concerning the functions of lipoproteins and apo-LP in relation to the transport of cholesterol into the vascular wall and its drainage.

[Content of testosterone and of different classes of lipoproteins in the blood plasma of men 40-59 years old].

The blood plasma of males aged from 40 to 59 (227 practically healthy individual and 48 persons suffering from ischemic heart disease) was tested for the content of testosterone, cholesterol, triglycerides, cholesterol of various classes of lipoproteins and the lipoprotein spectrum by electrophoresis in polyacrylamide gel. It was found that a change in the testosterone level in the blood plasma, even within the limits of physiological values, may affect the plasma lipoprotein spectrum: a decrease in the testosterone concentration is attended with an increase in the content of atherogenic (beta- and pre-beta-) pipoproteins and a decrease in the content of alpha-lipoproteins.

[Study of low density lipoproteins in hyperlipoproteinemia by the method of double immunodiffusion].

By way of gel double immunodiffusion a certain heterogeneity of low density lipoproteins was observed to manifest itself in an additional band of immunoprecipitation. The incidence of this additional band does not exceed 1/3 of the cases of low density lipoproteins studies in normal individuals, while in those with ischaemic heart disease the additional band is found twice-thrice as often, the highest incidence being noted in patients with ischaemic heart disease and Type IIb hyperlipoproteinemia (93% of cases). The revealed immunochemical heterogeneity of low density lipoproteins was shown to be not connected with the appearance of any new, additional antigen in their structure. Most probably it is attributable to the presence of lipoprotein particles with a quantitatively different protein and lipid composition, probably of intermediate lipoprotein metabolites in blood plasma, or conformation changes in the structure of low density lipoproteins.
