Comparison of immunofluorescence and hemagglutination inhibition tests and enzyme-linked immunosorbent assay for detection of serum antibody in rats infected with hemorrhagic fever with renal syndrome virus.

Serum antibodies against the B-1 strain of hemorrhagic fever with renal syndrome (HFRS) virus in wild and experimental rats were investigated by the indirect immunofluorescent antibody (IF) test, the hemagglutination inhibition (HI) test, and an enzyme-linked immunosorbent assay (ELISA). In the newly developed ELISA, monoclonal antibodies to the B-1 strain were applied as coating antibody. In sera from wild rats, the results obtained by the ELISA agreed quite well with those obtained by the HI test, but some serum samples that gave a negative reaction in the HI test gave a positive reaction in the IF test, although their IF titers were very low. In serum samples from experimental rats that had been kept in an animal house infected with HFRS virus, a group with high titers and a group with low or negative titers were clearly differentiated by all the tests.

[Toxic effects of cisplatin in the treatment of malignant bone and soft tissue tumors].

A total of 31 patients with malignant bone and soft tissue tumors were evaluated for toxic effects after receiving cisplatin (104 courses) at doses of 2.0-3.0 mg/kg with aggressive hydration. Nausea was common and in 86% of cisplatin courses severe vomiting was observed. Nephrotoxicity was observed in less than 10% of all courses but ototoxicity, especially tinnitis, developed in 5 of 6 patients whose total doses of cisplatin were over 500mg, and one of them showed hearing disorder in the upper frequency above 5,000 Hz. There were no other toxic effects such as severe myelosuppression, allergic reaction or peripheral neuropathy.

[Surgical treatment of metastatic hepatic cancers].

Main primary sites of hepatic metastatic cancers were the stomach, colon, pancreas and esophagus. Those were found in 16.1% of 335 stomach cancer, 11.9% of 270 colon cancer, 27.1% of 70 pancreas cancer and 3.8% of 131 esophageal cancer at the time of operation. However, the metastatic lesions were resectable in 13%, 43%, 0% and 20%, respectively. Prognosis after hepatectomy was favorable only in colon cancer. 7 of 13 patients who underwent resection of the colon cancer and hepatic metastases are still surviving. The others all died. In inoperable cases, chemotherapy, especially intrahepatic arterial infusion, prolonged the survival time for months.

[UFT, UFTM, and UFTM-Dpc treatment of gastric cancer].

It became increasingly evident that UFTM chemotherapy, a combination of UFT and mitomycin C is one of the most recommendable therapy for advanced gastric cancer, especially for the treatment of Borrmann type 4 gastric cancer. Objective responses were achieved in 18 out of 26 (69.2%) patients with Borrmann type 4 gastric cancer. Moreover, UFTM-Dpc treatment, a combination therapy of UFTM plus D-penicillamine (suppressor of collagen synthesis), is proposed for the treatment of Borrmann type 4 gastric cancer from the point of view of collagen synthesis inhibition. It is likely that D-penicillamine might inhibit the peritoneal metastatic involvements, which is lethal to the patients finally.

Introns and their flanking sequences of Bombyx mori rDNA.

We obtained two different clones (16 kb and 13 kb) of B. mori rDNA with intron sequence within the 28S-rRNA coding region. The sequence surrounding the intron was found to be highly conserved as indicated in several eukaryotes (Tetrahymena, Drosophila and Xenopus). The 28S rRNA-coding sequence of 16 kb and 13 kb clone was interrupted at precisely the same sites as those where the D. melanogaster rDNA interrupted by the type I and type II intron, respectively. The intron sequences of B. mori were different from those of D. melanogaster. In 16 kb clone, the intron was flanked by 14 bp duplication of the junction sequence, which was also present once within the 28S rRNA-coding region of rDNA without intron. This 14 bp sequence was identical with those surrounding the introns of Dipteran rDNAs.

The role of alpha and beta adrenergic receptors in constriction and dilation of the systemic capacitance vessels: a study with measurements of the mean circulatory pressure in dogs.

The response of the mean circulatory pressure (MCP), an index of the tone of the systemic capacitance vessels, to the infusion of an alpha-adrenergic receptor stimulant (phenylephrine) and a beta-adrenergic receptor stimulant (isoproterenol) was studied in anesthetized, open-chest dogs. Provided that the blood volume (particularly, extra volume) remains constant, an increase in the MCP indicates an increase in the tone of the capacitance vessels ("venoconstriction") and a decrease in the MCP indicates a decrease in the tone of the capacitance vessels ("venodilation"). It was almost definitely concluded that the stimulation of the alpha-adrenergic receptor led to the increased tone of the systemic capacitance vessels and the stimulation of the beta-adrenergic receptor did not decrease the tone of the systemic capacitance vessels in anesthetized, open-chest dogs, but the stimulation of the beta-adrenergic receptor decreased the tone of the systemic capacitance vessels, when the tone had been previously elevated by angiotensin-II.

Scrub typhus and chiggers in northern Thailand.

It was reconfirmed that L. deliense was the most dominant chigger during the rainy season in northern Thailand. Overall prevalence of IgG antibody against R. tsutsugamushi among the rural villagers in the northern most part of Thailand was nearly 60% at the level over 1 : 10 by an indirect immunofluorescent test. The level of IgG and IgM antibody titers was considerably high as greater than or equal to 1 : 2560 in some sera, but did not markedly vary by age groups or sexes. Based on detection of IgM antibody, the probability of new or reinfections were considered.

[Evaluation of severe side effects of high-dose methotrexate in osteosarcoma].

Severe toxicity associated with high-dose methotrexate (HD-MTX) combined with citrovorum factor rescue (CR-Rescue) was evaluated in the 365 courses in 54 patients with osteosarcoma. Anaphylactoid reaction developed in 8 patients (14.8%), 8 courses (2.2%). The mean age was 14 years old. The total dose given was 85770mg in mean and 1840 mg per body weight (kg). The occurrence was proportional to the dose and the number of the course. Plasma MTX concentration remained under 10(-7) mol/l during the episode. Delayed clearance of MTX from plasma was treated with massive CF-Rescue in 10 patients (19%) (the mean age of 13.6), in 12 courses (3.3%), which was observed on an average at the 7th course when the total dose reached to 59530 mg, or 1390 mg per body weight (kg). Neurotoxicity was observed in only one patient (1.9%), one course (0.3%). Severe toxicity associated with HD-MTX therapy tended to occur according to dose escalation and the number of administration. To overcome severe toxicity careful observation of clinical symptoms and signs as well as adequate treatment of side effects without delay are of importance.

[Preservation of the affected limbs in the treatment of osteosarcoma in children].

The pre-operative radiotherapy and systemic chemotherapy is effective in reducing the size of tumor and making the tumor resectable. The results achieved with systemic chemotherapy are ultimately reflected in improved survival. Moreover, limb-saving procedure in the treatment of osteosarcoma has become possible with rationality in carefully selected patients. Sixty-seven patients with osteosarcoma within a range of 5 to 27 years of age were treated at Chiba Cancer Center Hospital in the recent 12 year-duration. Seventeen of 67 patients underwent en bloc resection for osteosarcoma of the femur, (12 patients) and the tibia (5 patients). No patients had evidence of local recurrence of the tumor and overall 15 patients are alive without distant metastasis after 9 to 120 months follow-up. Based on the effective chemotherapy schedule, the limb salvation in the treatment of osteosarcoma is most desirable if it is indicated without risk of decreased radicality of the treatment.

[Pulmonary metastases from osteosarcoma].

On the basis of prognosis in conjunction with tumor behavior, 22 patients with pulmonary metastatic osteosarcoma who had a systematic with combination of surgery and adjuvant chemotherapy were compared with thirteen patients treated randomly in the initial period. The thirteen patients had median survival of 5 months with median tumor-free interval of 6 months. Six of the twenty two patients, however, are alive and 16 had median survival of 15 months with median tumor-free interval of 14 months. In 20 of the twenty two treated with repeated or bilateral thoracotomies, there was a correlation between decrease number of metastatic lesions with number of thoracotomies and survival. Wedge excision performed repeatedly produced only minimal loss of vital capacity, compared with lobectomy. Dn bloc resection of pericardium, chest wall, and diaphragm enhanced no survival. The patients with tumor-free interval of greater than 8 months survived or succumbed to the disease and was associated with mitotic ratio of more than 10 or less. Both of chest X-ray and CT scan had a positive diagnostic rate of 41% and 67%, respectively. Currently, we conclude that modified Thomfold 's indication is applicable to the patient with pulmonary metastatic osteosarcoma with bilateral or multiple nodules and more than 8 months of tumor-free interval.
