Histochemical AMD electron microscopic studies of intercellular matrices of papilloma induced by painting DMBA on the skin of mice.

The localization and distribution of glycosaminoglycans (GAG) in papilloma tissue which was introduced by painting DMBA on the back skin of ICR mice were observed by light and electron microscopy. GAG consisting mainly of hyaluronic acid could be detected histochemically on the cell surface and extracellular matrices of the basal layer of the papilloma, but it could not be observed in the non-neoplastic epidermis. After incubation of tissue segments in a medium containing 35SO4, an autoradiograph was made, and 35S-radioactivity was observed in the interface between the papilloma epidermis and dermis. Ultrastructurally, the cell surface of keratinocytes in the basal layer of the papilloma was stained intensely with ruthenium red. A cell line was obtained from the papilloma, and GAG could be demonstrated on the cell surface and in the intercellular matrix of the cultivated keratinocytes by treatment with hyaluronidase. The significance of GAG in the proliferating squamous cells was discussed.

Androgenic granulosa cell tumors of the ovary. A clinicopathologic analysis of 17 cases and review of the literature.

We reviewed the clinicopathologic features of 17 androgenic granulosa cell tumors. The patients, two of whom were pregnant, ranged from 2 to 58 years of age (average age, 22.5 years). Fourteen of the patients were masculinized as evidenced by various combinations of facial acne, temporal recession of hair, clitorimegaly, deepening of the voice, and a male escutcheon. Five of these patients had amenorrhea; three of the patients demonstrated hirsutism only. The plasma testosterone level is known to have been elevated preoperatively in nine patients, including the three patients with hirsutism. Four prepubertal patients also demonstrated hyperestrinism as manifested by sexual pseudoprecocity. At laparotomy, all of the tumors were stage lai. Five of the tumors formed unilocular and two of the tumors formed multilocular thin-walled cysts that ranged from 23 to 36 cm in diameter. Nine of the remaining tumors were solid, and one tumor was solid and cystic; these tumors averaged 7 cm in diameter. Eleven tumors were granulosa cell tumors of the adult type, and six tumors were juvenile granulosa cell tumors. The only clinically malignant tumor in the series was a unilocular, cystic, adult granulosa cell tumor that recurred 18 months postoperatively and was fatal within four years.

Picosecond laser fluorometry of FAD of D-amino acid oxidase-benzoate complex.

Formation of a complex of D-amino acid oxidase (D-amino acid:O2 oxidoreductase (deaminating), EC 1.4.3.3) and benzoate, an enzyme-substrate complex model, was studied by measuring the fluorescence life-time of the coenzyme FAD of the complex by using a mode-locked Nd:YAG laser and a streak camera. The value of lifetime was 60 +/- 10 ps in the monomer of the complex and it was extremely short (much less than 5 ps) in the dimer of the complex. Since the values of fluorescence lifetime of the coenzyme are 130 ps in the monomeric form of free enzyme and 40 ps in the dimeric form of free enzyme, the decrease in the lifetime upon complex formation with benzoate is slight in the monomer (reduced to one-half) whereas marked in the dimer (reduced to less than 1/10). By analyzing the fluorescence decay curve, a dissociation constant of the monomer-dimer equilibrium of the complex was evaluated to be 0.4 +/- 0.3 microM, which is much smaller than that in free enzyme. Fluorescence analysis under steady state excitation revealed that the apparent dissociation constant (K) of FAD from the enzyme was decreased by 1:1000 upon the complex formation. Relative quantum yield of the fluorescence of FAD in the complex to that of free FAD exhibited appreciable dependence on the complex concentration: greater in the monomer and less in the dimer. These results suggest that a molecular interaction between FAD and amino acid residue(s) is strengthened by the complex formation, which contributes to a remarkable conformational change in the protein moiety of the complex.

Choroid plexus papilloma. Light and electron microscopic study.

Choroid plexus papilloma (CPP) was observed by light and electron microscopy, using surgically excised tissues in 7 cases and cultivated cells. CPP cells had numerous microvillous processes showing balloon-like features, lysosomes containing haemosiderin, and highly electron-dense irregular granules about 150-300 nm in diameter. Interstitial cells with highly electron-dense cytoplasm inserted their long and thin processes into the invagination of basal plasmalemma of CPP cells, occasionally breaking down the basal lamina. Many of them were located in the intercellular space among CPP cells, sometimes adhering to the ventricular surface of CPP cells. Ruthenium red stain was positive on the surface of CPP cells and was especially intense on the surface of microvilli and cilia. In culture, CPP cells and interstitial cells migrating from the CPP cell mass showed a phagocytic activity after treatment with Latex.

Malignant papillary cystadenoma lymphomatosum. Light and electron microscopic study.

A case of malignant papillary cystadenoma lymphomatosum was investigated by light and electron microscopy. Histologically, the tumor consisted mainly of a characteristic papillary cystadenoma lymphomatosum, but the feature of epithelial components varied greatly in separate portions of the tissue; papillary adenoma-, tubular adenocarcinoma-, acinic cell tumor- or oncocytoma-like features were observed, the tumor cells showing "atypia" in major components. Numerous intracytoplasmic inclusions were found in many oncocytic cells arranged in acinar structure, and stained with PAS, AZAN or Alcian blue. Electron microscopically, large numbers of very large, swollen and closely packed mitochondria were observed in almost all the tumor cells. The intraplasmic inclusions were composed mainly of a long dense body enclosed by a single unit membrane, and the contents constituted bundles of fine filament running parallel to the longer axis. The inclusions were often observed to unite with lysosomes and to co-exist with secretory granules in the cytoplasm.

Case report of neuroendocrine carcinoma of the skin, histochemical and electron microscopic study.

A rare case of neuroendocrine carcinoma of the skin in a 83-year-old male Japanese was reported. Histological, electron microscopical and cytological studies were performed on the surgically removed tumor tissue and the cultured tissue. The tumor occurred at the junction of the dermis and subcutaneous tissue of the right elbow, and spread to the right brachial region and axilla. Histologically, the tumor consisted mostly of small anaplastic cells, closely resembling lymphocytes. They showed a characteristic uniformity without rosette or trabecular formation in the usual histological sections, but an epithelial-like arrangement of tumor cells was readily observable both in the tissue culture and imprint preparations. Grimelius' stain was weakly positive in the periphery of the cytoplasm. Neuron Specific Enolase (NSE) activity was high both in the serum and in the tumor mass. No gastro-entero-pancreatic hormones were detected. Electron microscopically, membrane-bound granules of neurosecretory type, 90 to 170 nm in diameter were observed in the cytoplasm. These granules were characteristically distributed along the periphery of cytoplasm beneath the plasma membrane and in clusters in the cell processes. The tumor cells had sparse rudimentary desmosome-like junctions and a few cytoplasmic finger-like projections. Either a neurogenic or APUD cell origin of the tumor was suspected. The serum NSE value, suggesting to be a neurogenic origin before the histological examination in the present case, is considered to be a valuable marker substance for screening and therapeutic monitoring of neurogenic tumors.

Fourteen cases of left atrial myxoma with emphasis on related histology.

With the availability of diagnostic ultrasonic equipment, there is an increase in the number of diagnosed cases of left atrial myxoma. Fourteen such patients underwent surgical treatment. One major concern was the possible recurrence of the tumor. Histologic examination revealed that the tumor cells did not invade the muscular layer beyond the layer of elastic fiber adjacent to the endocardium, in all cases. We remove the stalk with the Fossa Ovale when the tumor grows on the Fossa Ovale. When the tumor is attached to the muscular septum or free wall, we remove the stalk with the endocardium and a part of the adjacent myocardial layer. The total resection of the muscular septum may not be necessary as far as the myxoma is concerned.

Influence of fixed fibroblasts on glycosaminoglycan synthesis of human gastric carcinoma cells in vitro.

The influence of fixed fibroblasts on the glycosaminoglycan (GAG) synthesis of gastric carcinoma cells was examined by incubation along with [3H]glucosamine. In well-differentiated adenocarcinoma cells, the amount of 3H-GAG in the interface material between the carcinoma cells and the fixed fibroblasts was much larger (about twenty times) than in the interface between the carcinoma cells and the bare culture plates, and 3H-GAG consisted mainly of heparan sulfate, with a small amount of dermatan sulfate and chondroitin sulfate. On the other hand, in poorly differentiated carcinoma cells, the amount of 3H-GAG in the interface material produced by the carcinoma cells on the fibroblast was almost the same as on the bare culture dish. In a conventional monolayer culture, well-differentiated adenocarcinoma cells produced a much greater amount of GAG, consisting mainly of dermatan sulfate, chondroitin sulfate and heparan sulfate, than poorly differentiated carcinoma cells. Almost the same amount of hyaluronic acid was secreted into the medium by both types of carcinoma cells.

[Mode of spread of intrahepatic bile duct carcinoma].

To determine the area of surgical resection and the prognosis in patients with intrahepatic bile duct carcinoma, we studied the mode of spread of this cancer, using 27 autopsy and 4 lobectomized cases. In hilar carcinoma, there was lymphatic invasion to the extrahepatic bile duct before invation to the hepatic lobules. Scirrhous cancer spread more distantly toward the peripheral areas of the liver than non-scirrhous cancer. The occurrence of extrahepatic metastasis might involve the following factors; depth of infiltration, extent of venous invasion, intrahepatic metastasis, size of the main tumor, and number of hepatic segments invaded by the cancer.

Papillary carcinoma of choroid plexus. Light and electron microscopic study.

Two cases of papillary carcinoma, one in a 23-month-old girl and the other in a 25-month-old boy who both died within a relatively short time after operation, were studied histologically and electron microscopically. Both tumors originated in the right trigone of the lateral ventricle and spread widely via the cerebrospinal fluid. Histologically, the tumors consisted mostly of a differentiated papillary architecture closely resembling choroid plexus papilloma. Some carcinoma cells, showing cellular atypism, displayed a multilayer arrangement. The amount and distribution of PAS-, Alcian blue-, or orcein-positive substances on the cell surface and/or in the interstitial elements of the carcinomas differed from that of choroid papillomas examined in our laboratory. Electron microscopically, the carcinoma cells in some areas showed a loss of apical-basal polarity, and the formation of both microvilli and desmosome-like structures was indistinct. Papillary carcinoma is reviewed on the basis of the literature from 1906 till 1980.

Progesterone-induced smooth muscle-like cells in the subperitoneal nodules produced by estrogen. Experimental approach to leiomyomatosis peritonealis disseminata.

In order to examine the pathogenesis and histogenesis of leiomyomatosis peritonealis disseminata (LPD), guinea pigs were treated with various doses of estradiol benzoate (E) (40, 80, 100, and 200 microgram/day, three times a week, by intramuscular injection). After treatment for 3 months, each dose of E produced lesions simulating the disseminated pattern of LPD. Ultrastructurally, the nodules were considered to be composed of cells resembling those of fibroblast. In guinea pigs, pretreatment with estradiol benzoate (100 microgram/day, three times a week intramuscularly for 3 months followed by combination treatment with E and progesterone (P) (0.5 and 1.0 mg/day, once a week, intramuscularly) for 1 to 3 months produced nodules similar to those with estrogen alone. Ultrastructurally, these nodules were composed of cells resembling smooth muscle and decidual cells. According to these results, we conjectured that estrogen differentiates and proliferates cells with the features of fibroblast-like cells from the subcoelomic totipotential mesenchyme, and these cells are differentiated into smooth muscle-like cells and decidua-like cells under the influence of both estrogen and progesterone, resulting in the production of multiple subperitoneal nodules.

Phase I study of ceftizoxime, a new cephalosporin. Single-dose study.

A phase I study of ceftizoxime, a new cephalosporin, was performed in 29 subjects. No abnormalities were observed in subjective symptoms, laboratory test results, or physical test results at estimated therapeutic doses of 500 mg intramuscularly, 500 and 1000 mg intravenously, and 2000 mg drip infusion. It is concluded, therefore, that the drug is safe for clinical use. The mean peak serum concentration was dose dependent. The mean serum concentrations of ceftizoxime in man at 5 minutes after 500 and 1000 mg by intravenous bolus were 58.4 and 112.8 micrograms/ml, respectively, which exceed the MIC80 against most pathogens tested. Thus, 500 or 1000 mg was estimated to be the therapeutic dose. The distribution volume of ceftizoxime was 16.0 to 18.6 liters, the total clearance was 135.6 to 154.9 ml/min, and the half-life in the beta-phase was 1.36 to 1.39 hours. Ceftizoxime is mainly excreted in the urine as unchanged drug, at an excretion rate in the 24-hour urine of approximately 80 per cent.
