Production and characterization of the extracytoplasmic portion of the human interleukin-2 receptor.

We made a mutant cDNA clone of the human interleukin-2 (IL-2) receptor by introducing a termination codon at the eighth amino acid residue upstream from the putative transmembrane region. Ltk- cells were transfected by this mutant cDNA and cell-lines secreting the extracytoplasmic portion of the IL-2 receptor were established. The artificial secretory molecule of the IL-2 receptor named "bottom-less" receptor was able to react with anti-Tac antibody and to bind IL-2. We purified the bottom-less receptor to homogeneity by affinity chromatography using an IL-2-Sepharose column. The affinity (Kd value) of the 125I-labeled bottom-less receptor to IL-2 coupled to Sepharose beads was estimated to be 4.5 microM. All these results confirm the putative membrane topology of the IL-2 receptor based on the primary structure, and suggest that the bottom-less receptor may maintain the basic structure and function of the extracytoplasmic portion of the IL-2 receptor.

46XX male; report of case.

Chromosomal studies on a prepubertal 7-year-old boy with a normal male phenotype except for undescended testes showed a 46XX normal female karyotype. An H-Y antigen assay done at the same time on peripheral blood was positive in a titer similar to that of normal male subjects. Although the microscopic findings for both testes were normal, hypergonadotropic hypogonadism was suggested from both LH-RH and hCG stimulation tests.

[A case of complete remission of distant metastases of a renal pelvic tumor by combination chemotherapy with mitomycin, cyclophosphamide and 5-fluorouracil].

A 48-year-old woman was admitted to our hospital with the complaint of gross hematuria in March, 1982. The diagnosis of left renal pelvic tumor and bilateral neck lymph-node metastasis was made. Left nephroureterestomy and partial cystectomy were carried out in April, 1982. After the operation, combination chemotherapy with mitomycin, cyclophosphamide and 5-fluorouracil was carried out. The neck lymph-node disappeared completely in May, 1982. No recurrence or metastasis has been detected up to April 1985.

[Diagnosis of renal trauma].

We experienced 25 cases of renal trauma in our hospitals during the past 5 years, 20 cases were male and 5 cases were female, ranging in age 10 to 51 years old with average 26 years old, and the chief complaints were gross-hematuria and pain of renal region. We studied the IVP and CT patterns of these cases. IVP revealed 13 cases of renal contusion, 11 cases of renal lacerated wound and 1 case of renal fragmentation. The CT scan of 14 cases showed that it was useful for the diagnosis of the degree of renal trauma, especially, for hematoma surroundings the kidneys. The hematoma surrounding the kidneys was clearly noted in 7 cases by CT scan, but the unclearness of renal contours and iliopsoas muscle was considered to suggest the existence of hematoma surrounding the kidneys by IVP. All of the hematoma surrounding the kidneys was absorbed by conservative therapy, but 5 of the 7 cases were treated within 3 months.

Tissue reaction to implanted biomaterials.

Tissue reactions to the single-crystal sapphire, titanium, Co-Cr-Mo alloy, and PMMA were histologically evaluated after implantation into the subcutaneous tissue and tibia of rats. In subcutaneous tissue the reaction from early necrotic change and acute inflammation to final encapsulation by fibrous connective tissue was least pronounced around single-crystal sapphire implants. Tissue reactions of a slightly greater degree were found around titanium and Co-Cr-Mo alloy implants. The most prominent tissue reaction was observed around PMMA implants. In the tibia, single-crystal sapphire, titanium, and Co-Cr-Mo alloy implants were encapsulated by newly formed compact bone, but bone was not so evident around PMMA implants. These findings suggest that single-crystal sapphire implants have more favorable tissue compatibility than those of titanium, Co-Cr-Mo alloy, and PMMA.

Impaired natural killer cell activity in Bloom's syndrome could be restored by human recombinant IL-2 in vitro.

Natural killer (NK) activity against K562 tumor cells was evaluated in peripheral blood nonadherent mononuclear cells (PBMNC) obtained from four patients with Bloom's syndrome. NK activity of PBMNC from all patients was found to be depressed to less than half of age-matched control values. However, all patients showed the normal percentages of large granular lymphocytes and of cells expressing the NK-cell-associated determinants (OKM1, Leu-7, and Leu-11). Although recombinant interferon-beta or -gamma augmented NK activity of patients' PBMNC in a manner similar to their effects on NK activity of normal individuals, impaired NK activity of PBMNC from Bloom's patients could be restored to normal ranges only by recombinant interleukin 2 (IL-2). These results suggested that IL-2 and interferons might participate in boosting NK activity of Bloom's syndrome in different ways.

Light-Induced Alkalinization of the Suspending Medium of Guard Cell Protoplasts from Vicia faba L.

The light-dependent pH changes in the suspending medium of guard cell protoplasts (GCP) from Vicia faba were studied. Upon illumination, the medium was initially slightly alkalinized and then acidified. The extent of alkalinization was lower in CO(2)-free air than in normal air. This initial alkalinization was inhibited by DCMU. Acidification in CO(2)-free air became observable in shorter duration of light exposure than that in normal air. The rate of acidification was higher in CO(2)-free air than in normal air. The CO(2) level of the medium decreased in the light, and increased in the dark. (14)CO(2) uptake was enhanced 2- to 3-fold by light, but not in the presence of DCMU. These results indicate that photosynthetic CO(2) fixation does take place in GCP and that the initial alkalinization is due to this photosynthetic CO(2) uptake. Diethylstilbestrol, a nonmitochondrial membrane-bound ATPase inhibitor, inhibited the acidification, suggesting that the acidification resulted from H(+) extrusion by GCP. The acidification in light was also prevented by KCN, and partly by DCMU. Possible mechanisms of alkalinization and acidification are discussed in relation to guard cell metabolism.

Light Activation of NADP-Malate Dehydrogenase in Guard Cell Protoplasts from Vicia faba L.

Light-induced swelling of guard cell protoplasts (GCP) from Vicia faba was accompanied by increases in content of K(+) and malate. DCMU inhibited the increase of K(+) and malate, and consequently swelling.Effect of light on the activity of selected enzymes that take part in malate formation was studied. When isolated GCP were illuminated, NADP-malate dehydrogenase (NADP-MDH) was activated, and the activity reached a maximum within 5 minutes. The enzyme activity underwent 5- to 6-fold increase in the light. Upon turning off the light, the enzyme was inactivated in 5 minutes NAD-MDH and phosphoenolpyruvate carboxylase (PEPC) were not influenced by light. The rapid light activation of NADP-MDH was inhibited by DCMU, suggesting that the enzyme was activated by reductants from the linear electron transport in chloroplasts. An enzyme localization study by differential centrifugation indicates that NADP-MDH is located in the chloroplasts, NAD-MDH in the cytosol and mitochondria, and PEPC in the cytosol. After light activation, the activity of NADP-MDH in guard cells was 10 times that in mesophyll cells on a chlorophyll basis. The physiological significance of light-dependent activation of NADP-MDH in guard cells is discussed in relation to stomatal movement.

Effects of dimethylpropranolol (UM-272) on the electrophysiological properties of guinea-pig ventricular muscles.

The effects of dimethylpropranolol (UM-272) on transmembrane action potentials were examined in isolated right ventricular papillary muscles of the guinea pig. UM-272 (10(-5) to 3 X 10(-4) M) caused a dose-dependent decrease in the Vmax of the action potential. At 3 X 10(-4) M, a slight decrease in the amplitude of action potential was also observed. The resting potential and the action potential duration were not affected by the drug. In the presence of UM-272, trains of stimuli at rates higher than 0.1 Hz led to an exponential decline in Vmax (onset rate, 0.13-0.28 per action potential) to a new plateau level. This use-dependent block was augmented at the higher stimulation frequency. The time constant for the recovery of Vmax from the use-dependent block (offset) was 7.1 to 7.3 sec. In depolarized papillary muscles with 8 or 10 Mm [K+]0, the inhibitory action of UM-272 on Vmax of the first action potential after a long quiescent period (tonic block) was augmented markedly, but the rates of onset and offset of the use-dependent block were similar to those in normally polarized preparations under 5 mM [K+]0. The curves relating membrane potential and Vmax in preparations stimulated infrequently were shifted by 7.2 mV with UM-272 at 10(-4) M in the direction of more negative potentials. These findings suggest that UM-272 has kinetically similar use-dependent inhibitory action of the fast sodium channels of cardiac muscles as other Class Ia antiarrhythmic drugs like quinidine or procainamide.(ABSTRACT TRUNCATED AT 250 WORDS)

[The role of lymphadenectomy in the treatment of renal cell carcinoma].

The usefulness of radical nephrectomy combined with lymphadenectomy for improving the prognosis of renal cell carcinoma was evaluated in 146 patients with this disease during the 21-year period from 1963 to 1983. Eighty-six out of the 146 underwent radical nephrectomy alone, while the remaining 60 patients received radical nephrectomy combined with lymphadenectomy. In cases of stage I and stage IV B, lymphadenectomy produced no improvement in the survival rate. On the other hand, of the 58 patients who had stage II and stage III disease, in the 23 who underwent lymphadenectomy, the survival rate improved significantly as compared to the 35 who did not.

[Fever in renal cell carcinoma].

Fever developed in 65 (37.8%) out of 172 patients with renal cell carcinoma, being the third most frequent symptom following hematuria and renal mass. Patients who developed fever were associated with symptoms such as palpable renal mass, general malaise anorexia, weight loss, acceleration in ESR, liver dysfunctions and an increase in alpha 2-globulin significantly more frequently than those who did not. Histologically also, high graded cases were more common with a significant. Moreover, the prognosis was significantly more poor.

Autoimmune disease and malignant lymphoma associated with host-versus-graft disease in mice.

BALB/c mice neonatally injected with (BALB/c X C57BL/6)F1 hybrid spleen cells develop host-versus-graft disease (HVGD) with immunopathological features characteristic of either systemic lupus erythematosus (SLE), or immunoblastic lymphadenopathy (IBL) in man. HVGD mice manifest polyclonal hypergammaglobulinemia with various autoantibodies, generalized lymphadenopathy, hepatosplenomegaly, immune complex glomerulonephritis and evolve in the end to malignant lymphoma. The necessary prerequisite for HVGD induction between donor and host can be summarized as follows: Histoincompatibilities in the H-2 region between donor and host are needed; predominant F1 donor cells needed for HVGD induction are, if not sole, steroid resistant, nylon wool nonadherent and Thy-1 positive T cells; the role of donor T cells is not only to present H-2 complex to the host, but also to interact with and proliferate in the host; strain differences are found for the susceptibility of HVGD induction in the host. It has been found that HVGD evolves to a malignant lymphoma of host T cell origin. The HVGD mouse model may, therefore, contribute to the understanding of the cell to cell interactions at work in the pathogenesis of IBL, as well as SLE, in man.

Calcium source for excitation-contraction coupling in myocardium of nonhibernating and hibernating chipmunks.

The amplitude of the early plateau phase of the action potential and the slow action potential of cardiac muscle were much lower in hibernating chipmunks than in nonhibernating chipmunks. The frequency-dependent contraction was decreased in hibernating animals but increased in nonhibernating animals. Caffeine caused a negative inotropic effect in hibernating animals but a positive inotropic effect in nonhibernating animals. Ryanodine caused greater inhibition in hibernating animals than in nonhibernating animals. These results suggest that the respective roles of the sources of calcium for cardiac excitation-contraction coupling are changed during hibernation.
