Enhanced tumor metastases in rats following cryosurgery of primary tumor.

The incidence of tumor metastases was studied in WKA/Hok rats after cryosurgery and surgical excision of primary tumors. When rats with syngeneic fibrosarcoma, KMT-17, were treated by cryosurgery 5 days after transplantation of the tumor, 15 out of 31 rats (48.4%) died with tumor metastases. In contrast only 4 out of 34 (11.8%) died with tumor metastasis after surgical excision. Because tumor cells either in the regional lymph nodes or in the peripheral blood were already detectable in about half of the rats treated by either cryosurgery or surgical excision, mechanical and physical control of enhanced metastasis by cryosurgery is ruled out. The development of anti-tumor immunity was delayed in the rats treated by cryosurgery as compared with that in rats treated by surgical excision. This was investigated by means of Winn's assay. A marked reduction of tumor metastasis was seen after the surgical removal of cryonecrotized tumor tissue, while implantations of cryonecrotized tumor tissue into the rats treated by surgical excision resulted in an increment of metastasis. The mechanism of the enhanced metastasis observed in this experimental investigation is discussed from an immunological point of view.

The significance of measurements of lipid peroxide and a newly-discovered amidase in inflammation.

Inflammation is a biological defence mechanism against noxious stimuli. Over-strong and oversuppressed inflammatory responses both lead to tissue damage and delay of tissue repair. Determination of inflammatory responses on the basis of chemical mediators, however, only informs about the degree of these responses themselves. It cannot show whether these responses are over-strong or oversuppressed. We have developed a prototype standard for safe judgement of the appropriateness of inflammatory responses: measurements of lipid peroxide (a product of tissue damage) and a newly-discovered amidase (related to wound healing).

Estimation of kinetic parameters, amount of endogenous substrate and contaminating enzyme activity in a target enzyme reaction.

New analytical methods were devised to estimate the Michaelis constant (Km), the maximum velocity (V), the concentration of endogenous substrate (x) and the activity of contaminating enzyme (u) in an impure enzyme reaction with single substrate. In the non-radiometric assay, the linear plot was developed on the basis of Eisenthal-Cornish-Bowden plot [1] by transforming the equation for reaction rate (v) consisting of Km, V, x, u and the concentration of substrate (S'). To confirm the accuracy of the linear plot, the non-linear fitting method was simultaneously devised in terms of a modification of the method of Cleland [2]. In the radiometric assay, the linear and non-linear kinetic analyses were applied to the equation for the radiometric rate (v*), expressed by Km, V, x and S* (concentration of radioactively labelled substrate) as in the non-radiometric assay. In both assays, the values of Km, V and x (with u value in the non-radiometric assay) were obtainable at x1 and x2 with a known ratio of x2/x1. The validity of the above methods was proved by the model experiments with purified enzymes; and the radiometric model experiment offered a good example for a new enzymatic assay method of many substrates. These methods were successfully applied to the practical experiments.

The importance of local hemostatic procedures in dental extractions and oral mucosal bleeding of hemophiliac patients.

Deficient coagulation factor replacement therapy is often indispensable for dental extractions and oral mucosal bleeding in hemophiliac patients. High concentrates are often infused in large amounts of volume, until complete hemostasis is achieved. Considering the various side effects of replacement therapy the volume infused should be reduced to a minimum. Several aspects associated with replacement therapy for the control of oral bleeding are studied. Our conclusion is that local hemostatic procedures are even more important than replacement therapy. The use of surgical packs and band retainers proved to be of benefit in this study.
