Attempts were made to prepare antisera monospecific for Lp(a) lipoprotein and to investigate the distribution of subjects according to plasma levels of Lp(a) in Japanese controls and patients with coronary heart disease or cerebral infarction. Positive plasma reactions to the double diffusion test for Lp(a) (Ouchterlony) were observed in 32.3% of the healthy Japanese subjects, which is similar to results previously reported in western countries. The plasma threshold level of 17 mg/dl was considered an appropriate point for dividing subjects into positive and negative groups depending on reactions to the double diffusion test. When subjects were divided into two groups at 17 mg/dl, a significant association was found between a high plasma level of Lp(a) and either coronary heart disease or cerebral infarction in the distribution of the cortical artery. These results suggest that Lp(a) may play an important part as a risk factor for coronary heart disease and cerebral infarction.
Two cases of thalamic tumour, spreading through the midbrain into the pons ventrolaterally, showed suppression of cerebellar blood flow on the opposite side to the tumour, and also showed reduction of blood flow in the pons on the side of the tumour in positron emission tomographic studies. The pathophysiological mechanism of crossed cerebellar diaschisis may be due to functional suppression of the pontine nuclei by destruction of descending fibres from the cerebral cortex.
The role of gastric mucous glycoprotein (GP) in cytoprotection by prostaglandin E2 (PGE2) and an anti-ulcer drug, sofalcone, which increases the endogenous prostaglandins (PGs), was investigated in rats with ethanol-induced gastric lesions. Oral administration of absolute ethanol in rats caused a marked increase in low-molecular weight luminal GP and a marked decrease in macromolecular weight GP in the gastric tissue. These changes were significantly prevented by pretreatment with PGE2 or sofalcone. The adhesive intensity of mucous GP to the surface epithelium was estimated by comparing the amount of GP released into the medium in an in vitro incubation system. Our observations indicate that the adhesion of mucous GP is markedly decreased by oral administration of absolute ethanol, and this decrease is prevented by pretreatment with PGE2 or sofalcone. The adhesion of the secreted mucous GP to the gastric epithelium is probably one of important factors in prostaglandin cytoprotection.

The tumor-specific transplantation antigens (TSTAs) of BALB/c mouse colon tumor C-C36 were characterized by gel filtration (TKS-gel FPLC), ion exchange chromatography (Mono Q FPLC) and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis. The cell surface antigens were solubilized noncytolytically with 2.5% n-butanol. It was demonstrated that the C-C36 antigens responsible for tumor rejection activity against C-C36 cells had a molecular weight of less than 200,000, and were eluted at 0.4-0.5M NaCl. On the other hand, the cross TSTAs expressed on C-C26 cells were eluted 0.2M NaCl. SDS-PAGE analysis of the partially purified antigens showed that the C-C36 TSTAs had four bands corresponding to molecular weights of 75,000, 68,000, 60,000 and 40,000. In contrast, the cross TSTAs expressed on C-C26 cells gave several major bands corresponding to 110,000, 94,000, 60,000 and 38,000, in addition to faint bands of 75,000 and 68,000. These results indicate that, although the partially purified C-C36 and C-C26 colon tumor antigens inducing rejection of C-C36 tumor challenge in syngeneic hosts showed differences in the ionic strength at which they were eluted, there are three common molecules with molecular weights of 75,000, 68,000 and 60,000, suggesting that these common molecules might be the tumor antigens responsible for the C-C36 colon tumor rejection.
The tumorigenicity of 9-3',4',5',6'-tetrachloro-o-carboxy phenyl-6-hydroxy-2,4,5,7-tetraiodo-3-isoxanthone sodium (CAS: 632-68-8) [also called Food Red 105 (FR 105) or Rose Bengal B], which is widely used for food or cosmetic coloring in Japan, was examined in (C57BL/6N X C3H/N)F1 mice. Animals were divided into 6 groups with 50 mice in each group, and they were given solutions of 0, 0.125, and 0.5% FR 105 in the drinking water starting at 6 weeks of age and ending at 101 weeks. The mean relative thyroid weights in both sexes of mice given 0.125 and 0.5% FR 105 were significantly increased compared to those of controls. Enlarged thyroid glands were composed exclusively of colloid goiters characterized by distended follicles lined with flattened follicular cells. The male mice given 0.5% FR 105 had follicular adenomas in the thyroid gland at an incidence of 22.9%, which was significantly higher than the incidence in controls (P less than .005). Radioactive 131I uptake in colloid goiters was markedly decreased compared to that in intact thyroid glands. The results indicate that FR 105 can induce colloid goiters and thyroid adenomas in a dose-dependent manner.

Recent reports have described that the accurate preoperative diagnosis of renal angiomyolipoma is possible with computerized tomography (CT). There are, however, some cases of angiomyolipoma which are indistinguishable from renal cell carcinoma on CT. Herein we report a case of renal angiomyolipoma which CT failed to reveal preoperatively. In a 71-year-old Japanese male, a left renal tumor was incidentally suspected on excretory urogram. Then CT demonstrated a homogeneous mass in the upper part of the left kidney, 6 cm in diameter, which showed a positive attenuation coefficient, slightly denser than renal parenchyma. Selective left renal arteriogram showed hypervascularity without aneurysmal formation or arteriovenous fistula. These radiologic findings strongly suggested renal cell carcinoma and radical left nephrectomy was performed. Histological examination confirmed angiomyolipoma composed mainly of smooth muscle with thick-walled vessels and immature fat cells. We reviewed the literature and discussed the computerized tomographic-pathologic correlation of angiomyolipoma.

Prostaglandin D synthetase was purified 2,600-fold from rat brain to apparent homogeneity, as judged by polyacrylamide gel electrophoresis and ultracentrifugation. The purified enzyme was a monomeric protein with a molecular weight of 27,000 +/- 1,000. The pI value, sedimentation coefficient, and partial specific volume were 4.6, 4.1 s, and 0.73 ml/g, respectively. The enzyme was stable between pH 4 and 11 at the temperature lower than 25 degrees C and resistant to a heat treatment under alkaline conditions (pH 8-11). About 50% of the activity was detected after a heat treatment at 100 degrees C for 5 min at pH 10. However, the enzyme was readily inactivated by the isomerase reaction of prostaglandin H2 to prostaglandin D2. The enzyme required sulfhydryl compounds such as dithiothreitol, glutathione, beta-mercaptoethanol, cysteine, and cysteamine for the reaction, but stoichiometric oxidation of these sulfhydryl compounds was not observed. The optimum pH, Km value for prostaglandin H2, and the turnover number were 9.5, 14 microM, and 170 min-1, respectively. The antibody was raised against the purified enzyme in a rabbit, which showed only one positive band in immunoblotting after gel electrophoresis of crude extracts of the brain at the same position as that of the purified enzyme. More than 90% of the prostaglandin D synthetase activity in the brain was absorbed by an excess amount of the antibody, indicating that our preparation is a major component of the enzyme responsible for the biosynthesis of prostaglandin D2 in the brain.
The effects of vascular damage on platelet function were investigated in rats. Common carotid artery specimens 4 mm in length were dissected from male Wistar rats and exposed to ultrasonic treatment or rubbing of the intimal surface. In some cases, carotis was dissected after ligation for 4 hr. The specimens were fixed on a stirring bar and agitated in platelet-rich plasma added with ADP. After determining platelet aggregability, the plasma was subjected to radioimmunoassay for 6-keto-prostaglandin F1 alpha (6-K-PGF1 alpha). Anti-platelet-aggregability in the arterial specimens was significantly greater in the sonicated, rubbed or ligated specimens than in the intact group, probably due to the effect of prostacyclin (PGI2). The measurement of 6-K-PGF1 alpha levels yielded similar results. Endogenous arachidonic acid in the vessel wall seems to be utilized in the production of PGI2, which probably acts only on local platelets. The findings indicate the presence of a mechanism that inhibits the formation of platelet thrombi by the accelerated production of PGI2 under conditions favorable for the generation of thrombosis.
Bilateral persistent primitive hypoglossal arteries associated with multiple aneurysms in a 59-year-old man are reported. Angiography revealed bilateral persistent primitive hypoglossal arteries with a large ruptured intracranial aneurysm on the left side. There was one additional small aneurysm at the trifurcation of the right middle cerebral artery. This seemed to be the first case of bilateral persistent primitive hypoglossal arteries associated with multiple aneurysms.
