[Effects of rhGM-CSF on Apoptosis of HL-60 Cells Induced with VP-16 Treatment]

In order to investigate the effects of rhGM-CSF on apoptosis of HL-60 cells induced with VP-16 treatment, HL-60 cells were first incubated with rhGM-CSF before they were treated with VP-16. The apoptosis processes and the changes in apoptosis related gene bcl-2 and fas expression were observed. The morphological and ultrastructural changes were observed under optics microscope and electromicroscope. DNA fragmentation were detected by agarose gel electrophoresis, the apoptotic rate, bcl-2 and fas expression with flow cytometry. Our results showed that rhGM-CSF inhibited the apoptosis of HL-60 cells induced with VP-16 treatment, which enhanced the down-regulation of bcl-2 expression, but inhibited the up-regulation of fas expression. So it suggested that rhGM-CSF can decrease the sensitivity of HL-60 cells to VP-16, probably by down-regulation of fas expression.

Synthesis of proteins from [35S]methionine by guinea pig megakaryocytes in vivo and time course of appearance of newly synthesized proteins in platelets.

The relationship of protein synthesis to megakaryocyte maturation has been studied in guinea pigs in vivo. Guinea pigs were injected with a single dose of [35S]methionine. Megakaryocytes and platelets were isolated daily for 4 days, and proteins from both cells were isolated by DEAE-Sephacel chromatography and analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and fluorography. All proteins in megakaryocytes corresponding to stained bands on the SDS-PAGE gels were radiolabeled at 3 hours after injection. The greatest loss of radioactivity from the megakaryocytes occurred between 1 and 3 days after injection. Only trace labeling of platelet proteins was seen at 3 hours, representing almost entirely three bands at molecular weights 47,000, 52,000, and 66,000. At 24 hours only about 13% of the maximal labeling was present, but not all proteins were labeled. The maximal labeling was at 3 days. The pattern of labeling of platelets at 3 days was identical to that of megakaryocytes at 3 hours. The protein pattern of nonmegakaryocytic marrow cells was different from that of the platelets and megakaryocytes. Data presented here suggest that most protein synthesis in megakaryocytes is completed at least 24 hours before release of the platelets to the circulation, and suggest some specificity in the proteins that are synthesized at the terminal stages of maturation.

Provision of pharmacy services in community health centers and migrant health centers.

OBJECTIVES: To assess the frequency with which pharmacy services are provided in community health centers (CHCs) and migrant health centers (MHCs), the importance of these services in these settings, and pharmacists' preparedness to provide these services and to determine whether pharmacist- and site-specific characteristics are related to the provision of pharmacy services. DESIGN: A self-administered mail survey was used to collect information on the provision of traditional pharmacy services and on the provision of pharmaceutical care services in four domains: collecting, organizing, and evaluating information; formulating a course of action; providing medications and counseling patients; and monitoring and managing patient outcomes. Data were also gathered on pharmacist- and site-specific characteristics. Descriptive and inferential statistics were calculated. PARTICIPANTS AND SETTING: Surveys were mailed to a sample of 1,260 contact persons (medical/executive directors and pharmacists). MAIN OUTCOME MEASURES: Pharmacist-reported frequency of the provision of pharmacy services, respondents' perceptions regarding the importance of the provision of pharmacy services, and respondents' perceived level of preparedness to provide pharmacy services. RESULTS: Pharmacists in CHCs and MHCs provided traditional services very often, felt that the services were important, and felt very prepared to provide the services. Although frequency and perceived importance and preparedness varied across and within the four pharmaceutical care domains, pharmacists most often reported collecting, organizing, and evaluating information and providing medications and counseling patients. Least often reported was formulating a course of action, followed by monitoring and managing patient outcomes. Similarly, pharmacists felt least prepared to provide these services. Pharmacists who had received additional education and training reported that they provided pharmacy services significantly more often (P < .05) and felt significantly more prepared to provide the services. CONCLUSION: Additional pharmacy-related education and training may be helpful in preparing pharmacists in CHCs and MHCs to increase their provision of pharmaceutical care services.
