Estimating myocardial attenuation from M-mode ultrasonic backscatter.

The objective of this investigation was to determine whether measurements of myocardial attenuation can be obtained from analyses of M-mode images. We exploited the inherent anisotropy of myocardial properties as a means of systematically varying the attenuation to evaluate this M-mode image-based method for myocardial tissue characterization. A commercially available ultrasonic imaging system was used to acquire M-mode images of 24 excised cylindrical specimens from six formalin-fixed sheep hearts that were analyzed using video signal analysis. Data were compensated for the presence of bright intramural myocardial echoes, a potentially significant contributor to uncertainty in measurements of attenuation from backscattered ultrasound. The estimated attenuation coefficient in dB/cm at an effective center frequency of 2.75 MHz as a function of angle of insonification for measurements obtained from analyses of M-mode images is presented. Given a linear frequency-dependence of attenuation in myocardial tissue over frequencies ranging from 1.5 MHz to 8 MHz, as has been previously reported, M-mode image-based analyses were used to estimate the slope of attenuation. Results showed slopes of attenuation (over a -10 dB transmit bandwidth of 1.875 MHz to 3.75 MHz) ranging from 1.00 +/- 0.07 to 1.81 +/- 0.08 dB/(cm.MHz) for perpendicular and parallel insonification, respectively. These values were in good agreement with contemporaneously measured values (0.99 +/- 0.02 to 1.77 +/- 0.04 dB/(cm.MHz)) obtained over a frequency bandwidth of 4 MHz to 7 MHz using a through-transmission radio-frequency-based approach. These data suggest that robust measurements of myocardial attenuation can be obtained from analyses of M-mode images and that this method may be diagnostically feasible in the clinical setting.

New sesquiterpenes from Erigeron annus.

Four new and five known eudesmane-type sesquiterpenes were isolated from the aerial parts of Erigeron annus. The structures of these new compounds were elucidated by spectral means, including 2D-NMR analysis, to be 1 beta,4 beta-dihydroxyeudesman-11-ene; 1 beta,7 alpha-dihydroxyeudesman-4(15)-ene; (1 R,5 S,6 S,7 S,10 R)-1 beta, 6 alpha-dihydroxyeudesman-4-one and 1 beta- D-glucopyranosyloxy-6 alpha-hydroxyeudesman-4(15)-ene. In addition, a chemical transformation of a known compound 1 beta,6 alpha-dihydroxy-eudesman-4 (15)-ene was carried out and the derivative 1beta,6alpha-dihydroxy-4beta(15)-epoxyeudesmane was obtained. Some of the compounds were evaluated for their in vitro antitumor activities.

Removal of earthy-musty odorants in drinking water by powdered activated carbon.

The removal of four earthy-musty compounds, 2-methylisoborneol (MIB), 2,4,6-trichloroanisole (TCA), 2-isopropyl-3-methoxy-pyrazine (IPMP), and 2-isobutyl-3-methoxy-pyrazine (IBMP), by powdered activated carbon (PAC) were investigated under the simulative condition of Xicun water plant, Guangzhou. The adsorption kinetics of odor compounds by two PACs show that main removal of odor compounds occurs within contact time of 1 h. Compared with the wood-based PAC, the coat-based PAC evidently improves the removal efficiency of poorly adsorbed compounds like MIB. The effects on removal efficiency such as optimum PAC dosage, initial concentration of the organics, chlorine residual, background organics, and changes of water quality were investigated. The percent removal of trace odorants at any given time for a particular carbon dosage is irrelative to the initial concentration of the odor compounds. Adsorptive capacity of PAC for target compounds is reduced by chlorine residual and background organics. Characteristics of raw water have vast influence on the removal of target compounds by PAC.

Aprt/Opn double knockout mice: osteopontin is a modifier of kidney stone disease severity.

BACKGROUND: Osteopontin (OPN) is reported to have two distinct functions in kidney disease: Promotion of inflammation at sites of tissue injury, and inhibition of calcium oxalate monohydrate stone formation. However, many of the studies supporting these functions were carried out in animal models of acute renal injury or in cultured cells; thus, the role of OPN in chronic renal disease is not well defined. We examined the role of OPN in adenine phosphoribosyltransferase (Aprt) knockout mice, in which inflammation and formation of 2,8-dihydroxyadenine (DHA) kidney stones are prominent features, by generating Aprt/Opn double knockout mice. METHODS: We characterized the phenotypes of six- and 12-week-old Aprt-/- Opn-/-, Aprt-/- Opn+/+, Aprt+/+ Opn-/-, and Aprt+/+ Opn+/+ male and female mice using biochemical, histologic, immunohistochemical, and in situ hybridization techniques. RESULTS: At 6 weeks of age, there was no difference in phenotype between double knockout and Aprt knockout mice. At 12 weeks, there was increased adenine and DHA excretion, renal crystal deposition, and inflammation in double knockout versus Aprt knockout male mice. Double knockout and Aprt knockout female mice at 12 weeks had less pathology than their male counterparts, but kidneys from double knockout females showed more inflammation compared with Aprt knockout females; both genotypes had similar levels of DHA crystal deposition. CONCLUSION: We conclude that (1) OPN is a major inhibitor of DHA crystal deposition and inflammation in male mice; and (2) OPN is a major modifier of the inflammatory response but not of crystal deposition in female mice. Thus, separate mechanisms appear responsible for the tissue changes seen in DKO males versus females.

Adeno-associated virus-mediated bone morphogenetic protein-7 gene transfer induces C2C12 cell differentiation into osteoblast lineage cells.

AIM: To investigate the effects of bone morphogenetic protein-7 (BMP7)-expressing recombinant adeno-associated virus (AAV) vector on the differentiation of C2C12 cells. METHODS: AAV-BMP7 was packaged by infecting the stable cell clone BHK-21 (integrated with recombinant AAV vector plasmid pSNAV-BMP7) with recombinant herpes simplex virus type 1, which expresses AAV-2 Rep and Cap and possesses AAV packaging functions. Following infection with AAV-BMP7 at multiplicities of infection of 1 x 10(5) vector genomes per cell and subsequent culture, C2C12 cells were assessed qualitatively for BMP7 production, alkaline phosphatase activity, osteocalcin production and Cbfal and MyoD expression. RESULTS: C2C12 cells transduced with AAV-BMP7 could produce BMP7 protein until d 28. Alkaline phosphatase in the cultured C2C12 cell lysate was elevated. Secreted osteocalcin in the culture medium was detectable at d 12 and Cbfal mRNA expression level was upregulated, coinciding with downregulation of MyoD in a temporal manner. CONCLUSION: The present in vitro study demonstrated that AAV-BMP7 could infect and efficiently convert C2C12 cells from myoblasts into osteoblast lineage cells.

Measurements of the anisotropy of ultrasonic velocity in freshly excised and formalin-fixed myocardial tissue.

The objective of this study was to quantify the anisotropy of ultrasonic velocity in freshly excised myocardial tissue and to examine the effects of formalin-fixation. Through-transmission radio-frequency-based measurements were performed on ovine and bovine myocardial specimens from 24 different hearts. A total of 81 specimens were obtained from specific locations within each heart to investigate the possibility of regional differences in anisotropy of velocity in the left ventricular wall and septum. No regional differences were observed for either lamb or cow myocardial specimens. In addition, no specific species-dependent differences were observed between ovine and bovine myocardium. Average values of velocity at room temperature for perpendicular and parallel insonification were 1556.9 +/- 0.6 and 1565.2 +/- 0.7 m/s (mean +/- standard error), respectively, for bovine myocardium (N=45) and 1556.3 +/- 0.6 and 1564.7 +/- 0.7 m/s for ovine myocardium (N=36). Immediately after measurements of freshly excised myocardium, ovine specimens were fixed in formalin for at least one month and then measurements were repeated. Formalin-fixation appears to increase the overall velocity at all angles of insonification and to increase the magnitude of anisotropy of velocity.

A brief conceptual tutorial of multilevel analysis in social epidemiology: linking the statistical concept of clustering to the idea of contextual phenomenon.

STUDY OBJECTIVE: This didactical essay is directed to readers disposed to approach multilevel regression analysis (MLRA) in a more conceptual than mathematical way. However, it specifically develops an epidemiological vision on multilevel analysis with particular emphasis on measures of health variation (for example, intraclass correlation). Such measures have been underused in the literature as compared with more traditional measures of association (for example, regression coefficients) in the investigation of contextual determinants of health. A link is provided, which will be comprehensible to epidemiologists, between MLRA and social epidemiological concepts, particularly between the statistical idea of clustering and the concept of contextual phenomenon. DESIGN AND PARTICIPANTS: The study uses an example based on hypothetical data on systolic blood pressure (SBP) from 25,000 people living in 39 neighbourhoods. As the focus is on the empty MLRA model, the study does not use any independent variable but focuses mainly on SBP variance between people and between neighbourhoods. RESULTS: The intraclass correlation (ICC = 0.08) informed of an appreciable clustering of individual SBP within the neighbourhoods, showing that 8% of the total individual differences in SBP occurred at the neighbourhood level and might be attributable to contextual neighbourhood factors or to the different composition of neighbourhoods. CONCLUSIONS: The statistical idea of clustering emerges as appropriate for quantifying "contextual phenomena" that is of central relevance in social epidemiology. Both concepts convey that people from the same neighbourhood are more similar to each other than to people from different neighbourhoods with respect to the health outcome variable.

A brief conceptual tutorial on multilevel analysis in social epidemiology: investigating contextual phenomena in different groups of people.

STUDY OBJECTIVE: (1) To provide a didactic and conceptual (rather than mathematical) link between multilevel regression analysis (MLRA) and social epidemiological concepts. (2) To develop an epidemiological vision of MLRA focused on measures of health variation and clustering of individual health status within areas, which is useful to operationalise the notion of "contextual phenomenon". The paper shows how to investigate (1) whether there is clustering within neighbourhoods, (2) to which extent neighbourhood level differences are explained by the individual composition of the neighbourhoods, (3) whether the contextual phenomenon differs in magnitude for different groups of people, and whether neighbourhood context modifies individual level associations, and (4) whether variations in health status are dependent on individual level characteristics. DESIGN AND PARTICIPANTS: Simulated data are used on systolic blood pressure (SBP), age, body mass index (BMI), and antihypertensive medication (AHM) ascribed to 25 000 subjects in 39 neighbourhoods of an imaginary city. Rather than assessing neighbourhood variables, the paper concentrated on SBP variance between individuals and neighbourhoods as a function of individual BMI. RESULTS: The variance partition coefficient (VPC) showed that clustering of SBP within neighbourhoods was greater for people with a higher BMI. The composition of the neighbourhoods with respect to age, AHM use, and BMI explained about one fourth of the neighbourhood differences in SBP. Neighbourhood context modified the individual level association between BMI and SBP. Individual level differences in SBP within neighbourhoods were larger for people with a higher BMI. CONCLUSIONS: Statistical measures of multilevel variations can effectively quantify contextual effects in different groups of people, which is a relevant issue for understanding health inequalities.

A brief conceptual tutorial on multilevel analysis in social epidemiology: interpreting neighbourhood differences and the effect of neighbourhood characteristics on individual health.

STUDY OBJECTIVE: Using a conceptual rather than a mathematical approach, this article proposed a link between multilevel regression analysis (MLRA) and social epidemiological concepts. It has been previously explained that the concept of clustering of individual health status within neighbourhoods is useful for operationalising contextual phenomena in social epidemiology. It has been shown that MLRA permits investigating neighbourhood disparities in health without considering any particular neighbourhood characteristic but only information on the neighbourhood to which each person belongs. This article illustrates how to analyse cross level (neighbourhood-individual) interactions, how to investigate associations between neighbourhood characteristics and individual health, and how to use the concept of clustering when interpreting those associations and geographical differences in health. DESIGN AND PARTICIPANTS: A MLRA was performed using hypothetical data pertaining to systolic blood pressure (SBP) from 25 000 subjects living in the 39 neighbourhoods of an imaginary city. Associations between individual characteristics (age, body mass index (BMI), use of antihypertensive drug, income) or neighbourhood characteristic (neighbourhood income) and SBP were analysed. RESULTS: About 8% of the individual differences in SBP were located at the neighbourhood level. SBP disparities and clustering of individual SBP within neighbourhoods increased along individual BMI. Neighbourhood low income was associated with increased SBP over and above the effect of individual characteristics, and explained 22% of the neighbourhood differences in SBP among people of normal BMI. This neighbourhood income effect was more intense in overweight people. CONCLUSIONS: Measures of variance are relevant to understanding geographical and individual disparities in health, and complement the information conveyed by measures of association between neighbourhood characteristics and health.

Adrenomedullin impairs the profibrotic effects of transforming growth factor-beta1 through recruiting Smad6 protein in human renal tubular cells.

Adrenomedullin (AM) was originally identified as a vasodilator peptide, and has recently been shown to be an antiproliferative factor in renal mesangial cells, suggesting that adrenomedullin may impair the progression of glomerulosclerosis. This study was to investigate the effect of adrenomedullin on transforming growth factor-beta1 (TGF-beta1)-stimulated cell growth, synthesis of extracellular matrix (ECM) components and the related molecular mechanism in a human tubular epithelial cell line HK-2. TGF-beta1 inhibited cell proliferation induced by fetal bovine serum, but neither AM itself affectted cell proliferation, nor did AM influence TGF-beta1-caused cell growth arrest. However, AM beginning at 10(-8) M alleviated the action of TGF-beta1-stimulated cellular collagen synthesis and secretion of fibronectin into cell culture supernatant. Activation of Smad proteins is known to be the key signaling pathway of the profibrotic effect of TGF-beta1, AM at 10(-8) M exerted no effect on TGF-beta1-induced Smad2 phosphorylation, but prevented the suppression of the inhibitory Smad6 protein by TGF-beta1 and restored Smad2-Samd6 complex formation. Our results suggest that AM can attenuate TGF-beta1-mediated renal tubulointerstitial ECM turnover via an antagonistic mechanism of inhibitory Smad in TGF-beta1-elicited signaling.

[Terramycin wastewater treatment with combination hydrolysis-denitrification-nitrification].

Successive hydrolysis-denitrification-nitrification process is adopted to treat terramycin wastewater in lab scale for 70 days. Two sludge bed reactors are used respectively for hydrolysis and denitrification, and two biofilm reactors are used for nitrification. When the COD and NH4+ -N concentrations in influent were 2 200 - 3 000mg/L and 400 - 460mg/L, more than 80% COD and TN removals were achieved under a total HRT of 56h. The COD of the effluent was reduced to 293mg/L through coagulation under a polyferric sulfate dose of 48mg/L as Fe.

Sesquiterpenes and other constituents from Achillea wilsoniana.

From the methanol extract of the whole plant of Achillea wilsoniana, 23 compounds were isolated. Their structures were elucidated by spectroscopic methods and chemical transformations. Three of them are new: 4E, 10E-9beta-hydroxy-3-(2-methylbutyroyloxy)-germacra-4,10(1)-diene-12,6alpha-olide (1), 4E,10E-3-(2-methylbutyroyloxy)-germacra-4,10(1)-diene-12,6alpha-olide (2) and 1beta,6a-dihydroxy-10beta-methyl-5alphaH,7alphaH-eudesm-4-one (3). In addition, 1beta-hydroxy-alpha-xyperone (5) and 9beta-acetoxy-3-(2methylbutyroyloxy)-germacra-4,10(1)-diene-12,6alpha-olide (1a) exhibited effective antibacterial activity against Staphylococcus aureus.

Efficient and quick inactivation of SARS coronavirus and other microbes exposed to the surfaces of some metal catalysts.

OBJECTIVE: To study the two metal catalysts Ag/Al2O3 and Cu/Al2O3 that interdict the transmission pathway for SARS and other respiratory infectious diseases. METHODS: Two metal catalysts Ag/Al2O3 and Cu/Al2O3 were pressed into wafers. One hundred microL 10(6) TCID50/mL SARS-CoV, 100 microL 10(6) PFU/mL recombinant baculovirus expressing hamster's prion protein (haPrP) protein and roughly 10(6) E. coli were slowly dropped onto the surfaces of the catalyst wafers and exposed for 5 and 20 min, respectively. After eluted from the surfaces of wafers, the infectivity of viruses and propagation of bacteria were measured. The expression of PrP protein was determined by Western blot. The morphological changes of bacteria were observed by electronic microscopy. RESULTS: After exposure to the catalysts surfaces for 5 and 20 min, the infectivity of SARS-CoV in Vero cells and baculovirus in Sf9 cells dropped down to a very low and undetectable level, and no colony was detected using bacteria culture method. The expression of haPrP protein reduced to 21.8% in the preparation of Sf9 cells infected with recombinant baculovirus exposed for 5 min and was undetectable exposed for 20 min. Bacterial membranes seemed to be cracked and the cytoplasm seemed to be effluent from cell bodies. CONCLUSION: Exposures to the surfaces of Ag/Al2O3 and Cu/Al2O3 destroy the replication and propagation abilities of SARS-CoV, baculovirus and E. coli. Inactivation ability of metal catalysts needs to interact with air, utilizing oxygen molecules in air. Efficiently killing viruses and bacteria on the surfaces of the two metal catalysts has a promising potential for air-disinfection in hospitals, communities, and households.

[Expression of midkine fusion protein and preparation and application of its monoclonal antibodies].

AIM: To express midkine (MK) protein, prepare its monoclonal antibody (mAb), and detect its expression in tumor cells. METHODS: The human MK gene fragment was cloned from kidney cancer tissues by RT-PCR and inserted into the prokaryotic expression vector pMS-31b. The MS2-MK fusion protein was expressed in E.coli after temperature induction. The purified fusion protein was used to immunize BALB/c mice to prepare mAbs against MK by routine hybridoma technique. The Ig subclasses of the mAbs were identified by ELISA. The expression of MK in tumor cells was detected with the mAbs. RESULTS: MK gene was cloned and MS2-MK fusion protein was expressed successfully. Two hybridoma cell lines secreting anti-MK mAbs were obtained. The Ig subclass of the 2 mAbs was IgG1 and IgG2a, respectively. MK was expressed in gastric carcinoma cell line MGC803 and gastric cancer tissue. CONCLUSION: The MS2-MK fusion protein was expressed in E.coli and the mAbs against MK were pepared, which allow futher research on MK function.

[A study of cerebral evoked potentials response to esophageal distention in non-erosive gastroesophageal reflux disease].

OBJECTIVE: To study the mechanism of visceral hypersensitivity in patients with non-erosive gastroesophageal reflux disease (NERD) and to get further objective evidence in the abnormal alteration of the afferents involved in mediating esophageal sensation by cerebral evoked potentials (CEP). METHODS: We recruited 21 NERD patients and 10 normal healthy volunteers for the study. Mechanical distention stimulation was performed using a balloon-affixed polyvinyl multilumen catheter. First, maximally tolerated pain threshold of all subjects were recorded, then esophageal mechanical stimulation at an intensity of 75% maximum tolerated intensity and a frequency of 0.2 Hz was inflated in a total of 64 times by means of a computer-controlled barostat. The alternation of esophageal CEP was recorded before and after acid perfusion with a multi-channel international 10-20 system of electroencephalograph. RESULTS: Esophageal mucosal distention may evoke recognizable and reproducible multi-peak CEP. CEP morphology of the NERD patients was characterized by randomly distributed patterns and the peak latencies for N1, P1, and N2 were significantly shorter for mechanical stimulation as compared with the control group (respectively, P = 0.016, 0.003, 0.031), and the amplitude of the P1-N2 components was significantly increased in NERD patients (P = 0.03). CONCLUSIONS: Characterization and alternation of CEP morphology and peak latencies and P1-N2 amplitudes elicited by esophageal distention in NERD patients provides evidence for defective hypersensitivity of afferent neural pathways and cortical processing.

[Study on the combination of preozonation and post-ozonation-BAC process for drinking water treatment].

This study was conducted to illustrate the effectiveness of preozonation or O3-BAC or combination of these two process in controlling DBPs for treating a dam source water in South China through SBR and continuous flow (200 L/h) experiments and the variables in the treatment train included the point of preozonation with respect to coagulation; the point of ozonation with respect to BAC, the ozone dosage required for preozonation and the Br- content of raw water. Results indicated the reduction in DBP formation potential closely paralleled the reduction in UV absorbance, and trihalomethane and haloacetic acid formation potential can be removed under a dose of preozone at 0.5 - 1.0 mg/L, at the same time, ozone by-products (bromate and formaldehyde) can be controlled in low level. The continuous flow experiment results show that combination of preozonation and O3-BAC process has obviously positive effects on the removals of particles with a size above 2 microm, COD(Mn), and TOC. However, DBPs formation can be controlled by the removal of organic matters.

Silicon device scaling to the sub-10-nm regime.

In the next decade, advances in complementary metal-oxide semiconductor fabrication will lead to devices with gate lengths (the region in the device that switches the current flow on and off) below 10 nanometers (nm), as compared with current gate lengths in chips that are now about 50 nm. However, conventional scaling will no longer be sufficient to continue device performance by creating smaller transistors. Alternatives that are being pursued include new device geometries such as ultrathin channel structures to control capacitive losses and multiple gates to better control leakage pathways. Improvement in device speed by enhancing the mobility of charge carriers may be obtained with strain engineering and the use of different crystal orientations. Here, we discuss challenges and possible solutions for continued silicon device performance trends down to the sub-10-nm gate regimes.

Biomass production of yeast isolate from salad oil manufacturing wastewater.

Conversion of oil-rich salad oil manufacturing wastewater (SOMW) into protein source for animal feed through biomass production of yeast isolate was investigated in this study. Five species of yeasts, including Rhodotorula rubra, Candida tropicalis, C. utilis, C. boidinii, Trichosporon cutaneum, were isolated from SOMW following enrichment culture. Of them, C. utilis was chosen as the sole biomass producer in the study due to its greatest oil uptake rate, 0.96 kg oil kg(-1) biomass d(-1), and highest specific growth rate, 0.25 h(-1). The cells of C. utilis contained 26% protein, 9% crude lipid, 55% carbohydrate and balanced amino acid compositions. The initial N:C ratio in SOMW drastically influenced oil reduction efficiency, biomass production and protein content of C. utilis, and therefore a range between 1:6 and 1:8 was recommended in consideration of these three factors simultaneously.
