Mutagenesis by neocarzinostatin in Escherichia coli and Salmonella typhimurium: requirement for umuC+ or plasmid pKM101.

Neocarzinostatin, a protein with antibiotic activity, is a bacterial mutagen. We have investigated the mutagenicity of neocarzinostatin towards Salmonella typhimurium and discovered that, unlike the situation in Escherichia coli, neocarzinostatin will revert base pair substitution mutations (missense or nonsense). However, when the R46 factor derivative, plasmid pKM101, was introduced, the mutagenicity of neocarzinostatin towards base pair substitution-carrying mutants of S. typhimurium was readily detected. Neocarzinostatin had only modest activity in reverting a frameshift mutation in S. typhimurium, but that activity, too, required the presence of pKM101. Mutant pKM101 plasmids which no longer enhanced mutagenesis also lost their ability to promote neocarzinostatin-induced mutations. Finally, the umuC36 mutation, which renders E. coli nonmutable by ultraviolet light, also rendered the bacteria nonmutable by neocarzinostatin. The effect of the umuC36 mutation was suppressed by plasmid pKM101.

Palliative orbital decompression for metastatic melanoma to the orbit.

Although relatively rare, metastatic carcinomas to the orbit are the second most common orbital neoplasm in the adult. There are only ten cases reported in the literature of cutaneous melanoma metastatic to the orbit and, of these, only two had disease involving both orbits. This paper presents the third case of bilateral orbital melanoma; however, in this case, the primary site was the uveal tract in the first eye, with contralateral involvement being a metastasis of the orbital primary. The patient developed severe unilateral proptosis in his only seeing eye with progressive loss of vision. A palliative orbital decompression was performed which successfully restored visual acuity for an extended period of time. A brief discussion of the rationale for this procedure in this type of case is presented.

Ellobius lutescens Th. (Rhodentia, Microtinae): Q-, R-, and replication banding patterns. Chromosome 1 polymorphism in the male and presumptive heterogamety in the female.

Five specimens of Ellobius lutescens, four males and one female, were studied by several banding methods. Structural polymorphism occurred in the longest chromosome pair of the karyotype of two males: a pericentric inversion in one animal and a paracentric inversion in the other. In the female studied, one chromosome 1 differs from those of the males by a marked elongation of a proximal euchromatic band in the long arm. Thus, the female seems to represent the heterogametic sex. Chromosome 9 showed the same banding pattern in both sexes. The possible meaning of this chromosome constitution for the sex-determining mechanism of E. lutescens is discussed.

Cowden's disease: a cutaneous marker of breast cancer.

Cowden's diseases features facial trichilemmomas (a benign tumor of follicular epithelium), acral keratoses on the limbs, and oral mucosal papillomas and fibromas; it may also involve thyroid, gastrointestinal tract, ovaries, uterus, and breasts. Among 32 known cases of Cowden's disease, 21 are women, in 10 of whom breast cancer has already developed (bilateral in 4). The 11 women in whom breast cancer has not yet developed have fibroadenomas, fibrocystic disease, virginal hypertrophy of the breast, and malformations of nipples and areolae. Their median age is only 36 years. Two have mothers with breast cancer and in one both mother and maternal grandmother had breast cancer. Dermatologic lesions, including pathognomonic multiple facial trichilemmomas, precede the development of malignancy and can identify women with ahigh risk of developing breast cancer.

[Comparative effects of different inhibitors of phenylalanine hydroxylase and dihydropteridine reductase. In vivo and in vitro study in rats].

The effects of inhibitors of phenylalanine hydroxylase and dihydropteridine reductase have been tested in vivo and in vitro to induce classical phenylketonuria and the variant due to dihydropteridine reductase deficiency. In addition two inhibitors of intestinal absorption of phenulalanine were investigated. Blood, urine, and hepatic parameters have been measured and are discussed.

[On experiences with the new ultrasonic inhalation generator TuR USI 50 (author's transl)].

The paper deals with our experiences with the new TuR ultrasonic nebulizer USI 50, whose nebulizer chamber consists of glass and stainless steel. It works with an electronic feed system and has an electronically controlled heater for the production of thermoaerosols. In an alternating series of nearly 1,000 inhalations with normal and thermoaerosol, about 3/4 of the patients preferred thermoaerosol, about 20% preferred normal aerosol while 10% did not find any difference. According to our experiences the TuR USI 50 can be recommended for medical application.

[The measurement of aerosol density of ultrasonic inhalation generators (author's transl)].

Measurements of the behaviour of ultrasonic USI-Nebulizers during operation were carried out using a simple photometric arrangement. It shows, that the electrically controlled feed system of USI 3 and USI 50 is superior to the far simpler chicken feed system of USI 3 and USI 50 is superior to the far simpler chicken feed system. For the inhalation of small amounts of medicament not using the feed system the measuring procedure for the remaining amount of medicament should be reconsidered.

[Vocational rehabilitation after lung resection due to bronchial carcinoma (author's transl)].

Problems of vocational rehabilitation after radical operation of 307 patients with bronchial carcinoma, who are still living 3 years after the operation are analysed. Preoperative relation of professional groups: Employees 46%, industrial workers 28%, independent professions 15%, agricultural professions 11%. The success of rehabilitation is dependent on profession extent of resection, and other concomitant diseases.
