A new familial variant of antithrombin III: 'antithrombin III Paris'.

A 59-year-old woman presented a recurrent history of thromboembolism. A qualitative defect of antithrombin III (AT III) was suggested by the discrepancy between a normal amount of AT III antigen and a decreased heparin cofactor activity. Six members of the same family showed a similar defect although clinically asymptomatic. The qualitative abnormality of AT III was confirmed by two-dimensional immunoelectrophoresis. In the absence of heparin, a single peak was obtained with both control and patients' plasmas. In the presence of heparin, two peaks of AT III were observed in the patients' plasmas: the mobility of one peak was similar to that of the control, whereas the other showed a decreased mobility, suggesting a lack of binding to heparin. The two populations of AT III were separated by affinity chromatography on heparin-agarose. 50% of the patients' AT III bound to the agarose beads. The remainder, recovered in the supernatant, migrated in two-dimensional immunoelectrophoresis as a single peak with the same mobility in the presence or absence of heparin, and was devoid of heparin cofactor activity. This familial AT III variant characterized a reduced affinity for heparin is tentatively named 'Antithrombin III Paris'.

[Properties of small nebulizers and their suitability for aerosol therapy].

The current methods of measurement for particle size spectra do not seem to be applicable for small Hand-Held Nebulizers without impairing the relations of the amounts of fine and coarse particles. Therefore a simple method was developed that before-hand allowed a rough determination of these amounts. Measurements on four Hand-Held Nebulizers, three of them of the rubber ball nozzle-type and one ultrasonic, revealed that 90-95% of the medicament was unable to pass a 15 cm long rippled aerosol hose of 25 mm diameter. Therefore most of the drug is transformed into coarse particles and the application of Hand-Held Nebulizers is justified only for the therapy of the upper airways and for diseases where a resorption of the medicament in the main bronchi will suffice.

[Dynamic action of nebulizers and the influence of breathing technic on the deposition of aerosolized drugs].

Ultrasonic Nebulizers show a variation of aerosol density during the breathing cycle that affects deposition. First there is a relatively dense fraction of aerosol that penetrates deeply into the lung, while the subsequently delivered aerosol reaches only the upper parts of the airways. Calculations show that the effect has a pronounced influence on deposition. The percentage of medicament deposited in the lower airways is significantly increased, although the total amount of medicament is lower than with an aerosol of uniform density.

Kinetics and bioavailability of mexiletine in healthy subjects.

Single-dose kinetics of mexiletine (MEX) was studied in six healthy subjects after three different formulations. The respective doses were 200 mg (intravenous infusion), 400 mg (two conventional capsules), and 432 mg (sustained-release dosage forms). By a three-compartment open model with lag time the kinetic parameters of the drug were calculated from the experimental plasma level data. The mathematical analysis of the processes of distribution and elimination was restricted to the intravenous data only, and the resulting transfer constants were introduced into the evaluations of the oral experiments. With this procedure one common value for the plasma t 1/2 of elimination was obtained (t 1/2 gamma = 6.34 +/- 1.5 hr). Mean values for the total volume of distribution (Vdtot) and the total body clearance (Cltot) were 5.5 l/kg and 10.3 ml/min/kg. After capsules, peak plasma concentrations (Cmax = 0.77 microgram/ml) were reached after 2.2 hr. and the sustained-release form built up a flat maximum of concentration (Cmax = 0.34 microgram/ml) after 9.2 hr. Mexiletine is highly bioavailable, almost identical for the two oral formulations: 87.3% (capsule) and 78.7% (slow release). Under physiologic urinary pH1 7.5% to 9.2% of the dose was excreted unchanged by the kidneys.

Influence of growth temperature on structure, thermostability and kinetic properties of the ATPase coupling factor AF1 of a thermophilic blue-green alga (cyanobacterium).

The coupling factor AF1 isolated from cells of the thermophilic blue-green alga Mastigocladus laminosus grown at 40 degrees C, 50 degrees C and 60 degrees C is investigated and compared with the chloroplast coupling factor CF1. It is demonstrated that the structure of AF1 is affected by the growth temperature. The AF1 from 60 degrees C shows a split alpha-band in dodecylsulfate/polyacrylamide gel electrophoresis which is not observed in AF1 from 40 degrees C or from 50 degrees C. Only the AF1 from 60 degrees C aggregates with allophycocyanin. The AF1 from 60 degrees C is stable up to 85 degrees C for 60 min whereas the AF1 from 40 degrees C and 50 degrees C denature between 65 degrees C and 70 degrees C. CF1 is much less stable. In contrast to the structure, the kinetic properties of the coupling factor are not influenced by the growth temperature. Furthermore, the reaction rates of all three AF1 preparations have the same temperature optimum which jis also identical to that of CF1. The kinetic properties of the AF1 from 50 degrees C were determined in more detail. In the absence of ADP the saturation curve for ATP shows Michaelis-Menten kinetics with Km = 480 micromol and V = 30 micromol ATP hydrolyzed X mg protein-1 X min-1. With the addition of ADP this curve becomes sigmoidal and V decreases. This behaviour and the Hill values suggest an allosteric enzyme with two active sites acting cooperatively. ADP as a product of the ATPase reaction inhibits the enzyme. The inhibition is not simply competitive, it also influences the cooperativity of the active sites.

Induction of sister chromatid exchanges by hydroxylamine, hydrazine and isoniazid and their inhibition by cysteine.

Experiments were performed in order to gain information about the primary process leading to the production of sister chromatid exchanges (SCEs). Radical-forming substances (hydroxylamine, hydrazine and the antituberculous drug isoniazid) were examined for their effectiveness in inducing SCEs. All three substances proved successful in the induction of SCEs in the V-79 cell line of the Chinese hamster. By simultaneous application of a sulfhydryl compound (cysteine), a reduction of the hydrazine- and isoniazid-induced SCEs was achieved. Isoniazid was additionally examined in the in vivo SCE-test. At concentrations of 2-100 mg/kg body weight, it does not increase the rate of SCEs in the bone marrow of the Chinese hamster.

Synergistic action of cysteamine and BrdU-substituted DNA in the induction of sister chromatid exchanges.

The effect of different BrdU-concentrations on the cysteamine-induced SCE-rate was investigated in V-79 Chinese hamster cell monolayer cultures. Both cysteamine and its auto-oxidation product cystamine act synergistically with BrdU in the induction of SCEs. A given concentration of these substances produces a low SCE-frequency at low BrdU-concentrations -- but the incidence of SCEs is significantly increased at increased BrdU-concentrations. -- Using one-cycle substitution experiments and the determination of the relative level of substitution by means of 3HBrdU-incorporation, this synergism was shown to depend on the BrdU incorporated in the DNA and on the extent to which this incorporation takes place.

The effect of sulfhydryl compounds on sister-chromatid exchanges.

We examined the SH compounds glutathione (GSH), cysteine (CYS), cysteamine (MEA) and 2-mercaptoethanol (MET) with regard to their capacity for inducing SCEs. Whereas cysteamine and 2-mercaptoethanol increased the SCE frequency, this did not happen with glutathione and cysteine. By a comparative analysis of oxidized forms and structurally analogous compounds, it was confirmed that the induction of SCEs has no connection with the reducing property of the SH compounds. These findings are discussed in terms of other cytogenetic effects produced by SH compounds and the possible causes responsible for the induction of SCEs by SH compounds.

The SCE-inducing capacity of vitamin C: investigations in vitro and in vivo.

In contrast with results in vitro, vitamin C did not lead to an induction of sister-chromatid exchanges (SCEs) in the bone marrow of Chinese hamsters in the SCE test in vivo. The test dose ranged from 200 to 10 000 mg/kg body weight and was administered orally as well as by intraperitoneal injection. Sulfhydryl compounds such as cysteine and glutathione inhibited the SCE induction by vitamin C in vitro owing to their reducing capacity. These results are discussed with regard to the primary damage that may lead to an induction of SCEs by ascorbate, and the possible mutagenic danger to humans posed by vitamin C.
