Intracellular folate distribution in cultured fibroblasts from patients with the fragile X syndrome.

Altered folate metabolism has been suggested as a possible reason for expression of the fragile X chromosome in low-folate medium. However, there were no significant differences in the total folate content or in the distribution of folate cofactors between fibroblasts from patients with the fragile X chromosome and those of controls both before and after a period of folate starvation. Fragile X and control fibroblasts lose folate at an equivalent rate. Insofar as folate content and distribution reflect a primary abnormality of folate metabolism, there appears to be no such abnormality in the fragile X syndrome.

Significance of a (4;11) translocation in acute lymphoblastic leukemia.

This report describes a case of null cell congenital acute lymphoblastic leukemia associated with a (4;11) translocation. This chromosome abnormality is associated with acute lymphoblastic leukemia and probably more specifically with congenital acute lymphoblastic leukemia. A review of the literature is presented.

Susceptibility to cleft palate and the major histocompatibility complex (H-2) in the mouse.

The congenic mouse strains B10 and B10.A differ genetically only at the H-2 locus, B10 having the H-2b and B10.A the H-2a haplotype. The two strains also differ in susceptibility to cortisone-induced cleft palate, B10 being more resistant than B10.A. Since stage of palate closure is also associated with susceptibility to cortisone-induced cleft palate it was postulated that the H-2 haplotype may affect the cortisone-induced cleft palate susceptibility by altering the stage of palate closure. The present study shows that palate closure occurs at the same stage in the two strains, so this hypothesis must be rejected.

The genetical difference between the DBA/2 and C57Bl/6 strains in cortisone-induced cleft palate.

The DBA/2 strain of mice is about five times more sensitive than the C57B1/6 strain to cortisone-induced cleft palate. There is a matroclinous reciprocal cross difference. Appropriate backcrosses indicate that the basis of this reciprocal cross difference is not cytoplasmic. A biometric model including additive and dominance components adequately explains the data. Since dominance has been demonstrated in other strains and crosses, it is suggested that a trait correlated to cortisone-induced cleft palate has been exposed to directional selection.

Induced abortion in Belgium: clinical experience and psychosocial observations.

Abortion is illegal in Belgium, despite the pressure of public opinion, the feminist movement, and the numerous motions for liberalization presented to Parliament since 1971. Nevertheless, some hospitals and an association of outpatient clinics have practiced abortions openly since 1973. This study examines the socioeconomic and medical data of some 3,800 women who asked for an abortion in 1979, covering income, level of education, profession, religion, family structures, and personal relationships of the women and their partners. The women's reasons for seeking abortion are listed and their attitudes toward abortion and contraception are examined. Information on medical practice, stage of pregnancy, parity, and attendance at a routine follow-up visit is also given. The conclusion focuses on the probable benefits for women's well-being of a liberalization of the abortion laws.

Stage of palate closure as one indication of "liability" to cleft palate.

A new inbred mouse strain, SW/Fr, developed from a random-bred SW stock has a 6% incidence of spontaneous cleft palate without cleft lip. SW/Fr mice close their palates comparatively late in development. After cortisone treatment, the mean of the distribution (mean time to reach palate stage 5) is shifted towards later gestational ages. There is no change in the variance of the distribution. These data lend further support to the hypothesis that cleft palate in mice may fit a model where a continuous distribution is separated into discontinuous parts by a developmental threshold, and that time of palate closure is an important component of liability to cleft palate.

Double-blind crossover clinical pharmacology study comparing moxestrol (R 2858) and ethinyl estradiol in postmenopausal women.

The potency of 5 migrogram moxestrol (R 2858). 11beta-methoxy-17alpha-ethinyl-estra-1,3,5-[10]-triene-3,17-diol, administered orally, was compared to that of 25 microgram ethinyl estradiol in a double-blind crossover clinical pharmacology study of six postmenopausal women. At these doses, both compounds increased the eosinophilic index and decreased serum LH and FSH levels. A marked effect on circulating prolactin was observed only during ethinyl estradiol treatment.

Serum alpha-fetoprotein in adults, in women during pregnancy, in children at birth, and during the first week of life: a sex difference.

In order to investigate whether a sex difference exists in alpha-fetoprotein serum concentration, blood samples were collected from 133 adult men, 52 adult women, 239 pregnant women at different ages of gestation, 16 girls and 18 boys at birth, and seven girls and seven boys during the first week of life. In the case of 15 boys and 10 girls, blood samples were also collected from their mothers at the time of delivery. Serum AFP was measured by a radioimmunoassay method. In all neonates the mean value was 625 times higher than in all mothers at the time of delivery and 13,000 times higher than in adults. No difference was found between mothers of girls and those of boys both during pregnancy and at the time of delivery. However, at birth, the fetal AFP serum concentration was 1.8 (p less than 0.001) times higher in boys than in girls. This sex difference was maintained during the first week of life. The half-life of serum AFP was 5 to 6 days in the neonate. The origin and the possible physiologic significance of such sex difference in AFP deserve further investigations.

Serum prolactin levels during the menstrual cycle.

Serum prolactin has been measured in blood samples collected daily during 51 menstrual cycles using an homologous human radioimmunoassay for 17 cycles and an homologous ovine radioimmunoassay for 34 cycles. There was a progressive and significant increase in serum prolactin during the late follicular phase, with a maximal value concomitant to the LH peak. Serum prolactin levels were also significantly higher during the luteal phase than during early follicular phase. In some 70% of the individual cycles, the highest serum prolactin level was found at mid-cycle. Similar patterns were obtained with both radioimmunoassays. However, when using the same laboratory serum standard, the average serum prolactin level calculated for the entire cycle was 1.6 times higher with the homologous ovine assay than with the homologous human assay. The overall pattern of serum prolactin during the menstrual cycle resembles that reported for circulating 17beta-estradiol.

Influence of low dose oestrogen on circulating prolactin. LH and FSH levels in post-menopausal women.

The effect on serum prolactin, LH and FSH levels of 25 mug ethinyloestradiol administered daily per os during 27 consecutive days was investigated in 5 post-menopausal women aged 52-78. Blood samples were collected before, during and after treatment. The hormones were assayed in serum by radioimmunological methods. Both LH and FSH decreased progressively and significantly from 120 and 115 mIU/ml before treatment to 52 and 51 mIU/ml, respectively after three weeks of oestrogen administration. Two weeks after interruption of treatment, LH (90mIU/ml) and FSH (112 mIU/ml) were significantly higher than during the last week of treatment. Mean prolactin level increased from 127 muU/ml before treatment to 237 muU/ml after 10 days of oestrogen administration (P less than 0.001). This increase was significant after 4 to 8 days and the levels remained about twice as high as the control values for the rest of the treatment period. Two weeks after interruption of treatment, serum prolactin had fallen (136 muU/ml) to the pre-treatment levels. Such results raise the question of possible effects of elevated levels of this hormone during long term oestrogen medication in post-menopausal women on the development of breast cancer.

The influence of exogenous estrogen on the circadian periodicity of circulating prolactin in women.

Ethinylestradiol (400 pg/day) does not only stimulate prolactin release in normal cycling women but also modifies the pattern of the circadian periodicity of circulating prolactin: the nocurnal rise is of reduced amplitude but covers a larger part of the nyctohemeral period as compared to the situation during a control period in the same subjects. The values start rising long before sleep, i.e., -t 2 pm, under estrogen treatment and during sleep, i.e., at 4 am, during the control period. In both cases, however, the values remain high until 8 am and are minimal at 12 am.
