An ultrastructural study on the ear cartilage of rabbits after the administration of papain. Appearance of cross-striated collagen segments of an atypical FLS-type.

Crude papain was administered intravenously to young rabbits and the cartilage of the collapsed ear was examined electron-microscopically. Degeneration and recovery of chondrocytes, and decrease in and recovery of the electron-density of elastic fibers, were observed during the collapse and restoration of the ear. Some samples were stained with ruthenium red. In the collapsed ear, with a marked decrease of proteoglycan in the cartilage, loss of ruthenium red-positive granules was observed in the extracellular matrix. Collagen fibrils in the cartilage appeared to be somewhat increased in number, some of their diameters became slightly greater, and a part were assembled into bundles, occasionally accompanied by periodic cross-striation. Decrease of proteoglycan in the cartilage matrix probably brought about the unmasking and the assembly of collagen fibrils. In one of the experimental animals, collagen fibrous segments of an atypical fibrous long spacing (FLS-)type with symmetrical cross-striation were found around the chondrocytes in the ear cartilage, during the period of recovery. Some kind of the endogenous sulfated carbohydrate may have acted to affect the arrangement of type II collagen or procollagen molecules newly produced by the recovering chondrocytes.

Expression of antigenic determinants of the hemagglutinin gene of a human influenza virus in Escherichia coli.

Antigenic determinants of influenza virus hemagglutinin were expressed in Escherichia coli. DNA coding for presequences of hemagglutinin were removed and an ATG codon was placed before DNA coding for mature hemagglutinin. A number of expression plasmids were constructed in which various segments of this reconstructed hemagglutinin DNA were fused to DNA coding for bacterial beta-galactosidase. The fusion proteins exhibited specific binding to antiviral antibodies. This binding could be competitively inhibited by excess viral hemagglutinin, demonstrating that these fusion proteins contained antigenic determinants of hemagglutinin.

A new device for free skin grafting in rats.

We have employed this device in more than 200 grafting operations with complete success. Its advantage is to avoid the variation of fixing pressure on the grafts that greatly interferes with the experimental results. In addition, this method allows us to inspect the graft during the first week without having to remove the protector each time, which helped us greatly. We have presented a new device for skin grafting in rats that allows us to obtain temporary stabilization and protection of a skin graft. In addition, it allows us to inspect the graft without having to remove the protector.

Oscillation of intracellular Cl- activity and membrane potential in cultured fibroblasts (L cells).

The intracellular Cl-activity (aCli) and the membrane potential (Vm) were simultaneously measured in cultured L-strain fibroblasts, using a Cl- selective liquid ion-exchanger microelectrode and a standard microelectrode. The oscillations of aCli and Vm were found to occur concomitantly. A decrease in the aCli value was associated with the spontaneous hyperpolarization. The depolarization produced by current injection resulted in an increase in aCli. These data suggest that the membrane permeability to Cl- is significantly high during the Vm oscillation in the L cell.

[Hemodynamic effects of some beta-adrenergic blocking agents in conscious spontaneously hypertensive rats (author's transl)].

The hemodynamic effects of propranolol, pindolol and practolol were investigated i conscious spontaneously hypertensive rats (SHR). Mean blood pressure (MBP), heart rate (HR) ard cardiac output (CO) were measured and total peripheral resistance (TPR) was calculated. CO was determined by the modified dye dilution method using an arterio-venous shunt between the left carotid artery and the right jugular vein. Intraperitoneal administration of propranolol (5 mg/kg), pindolol (0.1 mg/kg) or practolol (50 mg/kg) caused a sustained antihypertensive effect of a similar degree. The antihypertensive effect of propranolol (20-30 mmHg, 0.5-8 hr) was attributed to an early decrease of CO (0.5-4 hr) followed by a reduction of TPR (6-8 hr), while that of pindolol or practolol (30-50 mmHg, 0.5-8 hr, 30 mmHg, 2-8 hr) was apparently related to a reduction of TPR with a CO increment. HR decreased with propranolol but increased with pindolol, while only slight changes were observed with practolol. Antihypertensive effects of propranolol, pindolol or practolol were associated with characteristic hemodynamic changes.

[Effect of calcium and magnesium ions on the interaction of corticosterone with the cytosol receptor(s) in the rat brain].

The effects of calcium and magnesium ions on the corticosterone binding to rat brain cytosol receptor protein(s) were investigated. The increasing amounts of CaCl2 or MgCl2 up to 5.0 mM were added, the specific [3H] corticosterone binding increased 1.3-fold and 1.5 respectively. The addition of MnCl2 and KCl did not affect this binding. The binding of corticosterone with rat brain cytosol receptor(s) were decreased by increasing amounts of EDTA and complete inhibition was observed at concentration equal to and greater than 2.5 mM. Inhibition of this binding by EDTA was less than by EGTA. Either theophylline or dibutyryl cyclic AMP had no effect on this binding.
